Molecular detection of Ehrlichia canis in Rhipicephalus sanguineus (s.l.) ticks in dogs and their domestic environment in Cuiaba, MT, Brazil

The central region of Brazil is known to be an endemic area for canine ehrlichiosis. Therefore, this study aims to determine the prevalence rates of E. canis infection in dogs and in Rhipicephalus sanguineus ticks collected from the dogs and their home environments. Serum samples and genomic DNA from the blood of 20 dogs and 299 ticks were analyzed by IFA and PCR assays in order to detect Ehrlichia canis antibodies and DNA. Nine (45%) of the 20 dogs were seropositive for E. canis, with titers ranging from 80 to 10240, and 6 dogs (30%) were positive for Ehrlichia spp. by PCR. Five free-living ticks were positive (2.89%, 95% confidence interval: 0.94-6.62%), as were six ticks attached to dogs (4.76%; 95% CI: 1.77-10.0%). The two groups showed a similar infection rate (P=0.395). Partial dsb DNA sequences of two samples from ticks were identical to each other and 100% (350/350 nucleotides) were identical to E. canis. Despite the high serological and molecular rates of canine ehrlichiosis in Cuiabá, the prevalence among infected ticks was lower than that found among dogs. However, adult ticks may remain infective much longer to ensure their infestation and infection of susceptible dogs.

Despite its important role as vector for African horse sickness virus (AHSV), very little information is available on the dissemination of this virus in Culicoides (Avaritia) imicola Kieffer (Diptera: Ceratopogonidae). This study reports on the applicability of a real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR) to detect AHSV in dissected midges. A total of 96 midges were fed on AHSV-infected blood, after which one test group was dissected into head/thorax and abdomen segments immediately after feeding and the other only after 10 days of incubation. The majority of the midges (96%) ingested the virus successfully and there was no significant difference between the virus concentration in the heads/thoraxes and the abdomens immediately after feeding. After incubation, virus was detected in 51% of the midges and it was confined to the abdomen in the majority of these. The fact that virus was detected only in the heads/thoraxes of four Culicoides midges after incubation suggests the presence of a mesenteronal escape barrier. Replication in the salivary glands was not shown. An increase of the mean virus concentration in the abdomen after incubation indicates localised viral replication. The real-time RT-qPCR is recommended for further studies investigating the replication and dissemination of AHSV in Culicoides midges.

Infectious diseases of swine, particularly zoonoses, have had a significant influence on nutritional safety and availability of pig meat as high-energy protein product since the time that pigs were domesticated back in the 7ᵗʰ century BC. The main sources of swine infectious diseases include the so-called primary sources (direct infection, i.e. through contact with infected and sick animals) and secondary sources (contaminated meat products, slaughter products, and vectors, including ticks). At present, the most serious epidemiological and economic threat to swine breeding in Europe is African swine fever (ASF). This disease, originally coming from Africa, is incurable and causes death of infected pigs and wild boars during 7−10 days after infection. Among the various factors that influence the spread of ASF, important role is played by ticks from the genus Ornithodoros, mainly from the species Ornithodoros moubata. Research on the ASF indicates that other species of ticks can also transmit the virus to healthy pigs in laboratory conditions. Sylvatic and domestic cycles of ASF virus transmission, which have been described so far, require further studies and updating in order to point the potential new vectors in the Caucasus and Eastern Europe affected by the ASF. Effective methods of control and biosecurity may significantly slow down the spread of ASF, which undoubtedly is a major threat to world pig production and international swine trade.

Some sandflies are vectors for leishmaniasis, therefore knowing the fauna of the region is important in combating the disease. Sandflies were captured in Concórdia, SC, Brazil, and Leishmania spp. in captured females. Thirty-eight specimens of six different species were collected, one of which (Brumptomyia carvalheiroi) is reported for the first time in the south of the country. However, there were no samples of sandflies positive for Leishmania spp. Despite reports of allochthonous cases in the region studied, the agent is not yet circulating in vectors, but these represent a potential risk when new cases occur.
