Donkeys, mules, and asses are domestic animals of worldwide distribution, found in all regions of Brazil. Although more health data is needed from these species, some sectors of today’s society are exploring alternatives to use these animals for milk production and the use of donkeys in social events and onotherapy. Donkeys (Equus asinus) are used in many everyday activities, but the animals need better welfare conditions. Although it is well known that endoparasite infections (i.e. cyathostomins, Strongylus spp., Parascaris sp.) can harm the clinical condition of the animals, there are limited clinical and laboratory supporting data, including registered therapeutic drugs for their control. The objective of the present work was to review the existing literature about parasite infections of donkeys, including their clinical implications, and the drug efficacy test to help practitioners improve their health management programs. To assist with that, we have developed a questionnaire to identify the risk factors of donkey farming, which is presented here in English and in Portuguese. Lastly, the present article also includes an original simulation using the susceptible-infected-recovered (SIR) model with low, moderate, and severe scenarios to help visualize the risk of parasite infections. The SIR model was based on the disease condition of donkeys in Brazil, using transient interaction levels from 0.0015 to 0.0025, infectivity levels from 0.10 to 0.40, and distinct recovery rates (R) from 0.01 to 0.08. As milk production is an expanding new activity worldwide, we want to emphasize that the first comprehensive studies must take into account differences in animal age, breed, and body condition score, local climate, pasture quality, day of sampling, and the period of lactation within the lactation curve to measure the health condition of donkeys. These data are considered important and will then be correlated to the parasite egg/oocyst (EPG or OoPG) counts in the feces using coproparasitological protocols (McMaster and Mini-FLOTAC), and even DNA-based techniques, to establish a novel data bank for donkeys. Target selective treatment has been proposed to be used as a routine in horses and will also be implemented in donkeys. This technique aims to reduce the number of anthelmintic treatments and, consequently, lessen parasite selection for resistance. The dynamic variations found by the SIR model are discussed in the text and should be taken into account when deciding the best health management implementation and parasite control programs. We think that more studies are needed to evaluate the basic physiological performance of donkeys in tropical areas to better understand their thermal adaptation, assuming that the animals are kept under acceptable clinical, nutritional, and housing conditions to express their resilient behavior. Cases of neglect and ill-treatment of animals must also be studied in infected conditions as well as their negative impacts. These strategies reinforce the concept of holistic farming with the possibility to improve specific informations, thereby improving the welfare of the donkeys.

| Developing effective cooled semen protocols is essential to increase pregnancy rates and reproductive efficiency in donkeys. This study aimed to evaluate the effect on sperm kinetic parameters and membrane integrity in cooled donkey semen diluted with defined milk proteins extender with 1% or 2% of egg yolk and the removal of seminal plasma. Twenty-four ejaculates from six jackasses were collected. Each ejaculate was divided into four aliquots that were diluted in extender with 1% (EY1) or 2% (EY2) egg yolk. One sample from each group was centrifuged, seminal plasma was removed (CEY1, CEY2 groups, respectively), and the samples were then refrigerated at 5 °C for 24 h. Fresh and cooled semen samples were assessed for sperm motility, morphology, and plasma membrane integrity. Total motility, progressive motility, sperm kinetic parameters, or live sperm cells were not statistically different when semen was cooled with an extender supplemented with 1% or 2% of egg yolk. Seminal plasma removal does not affect total motility or sperm kinetic parameters. However, progressive motility decreased (P<0.05) when semen was extended with 2% of egg yolk and seminal plasma was removed. Membrane integrity was affected (P<0.05) in centrifuged samples. In conclusion, the obtained results suggest that there is no difference in sperm kinetics and membrane integrity when 1% or 2% of egg yolk was added to the Equiplus(R) extender. Also, the removal of seminal plasma by centrifugation did not have any beneficial effect on cooled donkey semen. Further studies are needed to relate these results with in vivo fertility tests with cooled donkey semen.

| Donkeys have a long history in the development of human societies. Typically referred to as a beast of burden, traditional uses for donkeys have included the transportation of goods and people, use in agricultural and forestry activities, to access water, and provide citizens in low- and middle-income countries a means of making an income for communities. However, the rise of mechanization, the development of modern farming techniques, and the increasing availability of motorized vehicles have led to donkeys and mules becoming redundant from traditional roles in many parts of the world. We provide examples of where donkeys have successfully transitioned from traditional roles to new, non-traditional roles in Europe and North America, and demonstrate that, although the roles and use of donkeys and mules are changing in a rapidly developing world, we can learn lessons from the past and apply them to current challenges. As the need for working equids declines in transport and agriculture, they still hold great value for recreational, therapeutic, and environmentally friendly methods of animal traction.

| To better understand and manage the population of donkeys and mules, it is essential to know information about its size. In developing countries, these animals are often used as working animals, mainly by poor populations in agricultural and livestock labor, but their use is decreasing in Brazil. Brazilian official data regarding donkeys and mules started to be measured at the agricultural census of 1960 and, from that time, eight agricultural censuses were published. But until the writing of this paper, no scientific paper was dedicated to the analysis of the results of the official agricultural censuses on the population of donkeys and mules in Brazil. Thus, we aim to compile and analyze the official Brazilian data from 1960 until 2017 to enhance the actions of stakeholders, researchers, and decision-makers concerned with Brazilian donkeys and mules.

| Vehicle-animal collisions represent a serious problem in roadway infrastructure. | Growing demand for ejiao – gelatin produced from donkey skin and used in Traditional Chinese Medicine – is putting global donkey populations at risk and threatening the livelihoods of millions of people that depend on them in lower and middle-income countries. Hundreds of thousands of donkeys are slaughtered for their skins and exported annually, mostly from Africa to China. However, the trade is spreading across the globe, including Brazil. This article highlights the appalling welfare conditions for donkeys caught up in both the legal and illegal trade, as well as the effects on vulnerable people and the potential for disease spread and hazards to human health.

Objective: Humpback (hpbk) mice harbor a pathogenic mutation in the Notch3 gene and can serve as a beneficial animal model for investigating human myopathy, kyphosis, and developmental disorders, including lateral meningocele syndrome. Detection of the point mutation in hpbk mice is important for maintaining strains and scrutinizing genetic rescues, especially considering that homozygous mice are infertile and indistinguishable from their littermates at a young age. This study aimed for the development of a novel, precise, and time-saving genotyping method to identify the mutation in hpbk mice. Materials and Methods: In order to study the hpbk mouse line, we describe how we applied several tools, including quantitative polymerase chain reaction (qPCR), multiplex tetra-primer amplification-refractory mutation system (ARMS-PCR) and Sanger sequencing, toward the recognition of heterozygous and homozygous mice. Results: The Notch3 mutation was clearly identified using qPCR and ARMS assays, but the latter was a more precise and cost-effective approach. The lengths of the ARMS-PCR amplicons are 210 bp and 164 bp for the wild-type and hpbk alleles, respectively. Moreover, the genotyping results for each mouse were corroborated by Sanger DNA sequencing. Conclusion: Our newly developed PCR-based ARMS system affords a swift and precise way to genotype the hpbk mice. ARMS-PCR does not rely on any advanced equipment and is useful as a genotyping method for other model organisms that harbor a pathogenic variant. J. Adv. Vet. Anim. Objective: Present research aims to isolate, identify, and determine the virulence of the Streptococcus agalactiae (group B Streptococcus; GBS), isolated from popped eye disease affected Tilapia and Vietnamese Koi (V. Koi) fishes. Materials and Methods: A total of 330 fish samples were collected, of which Tilapia (n = 180) and V. Koi (n = 150), were collected from 35 affected ponds of four selected districts of Bangladesh. Isolation of the bacterium was done using different culture media (Nutrient broth, Plate count agar, Tryptic Soy Agar, and Blood agar), and identification by using various biochemical tests (conventional and using API 20 Strep kit) and polymerase chain reaction (PCR) using primers against 16S rRNA gene of S. agalactiae. Antibiotic susceptibility of the bacteria was performed using seven different antibiotics disc (Tetracycline, Oxytetracycline, Chlortetracycline, Streptomycin, Ciprofloxacin, Gentamicin, and Neomycin). Virulence of the isolated S. agalactiae was determined by infecting healthy Tilapia and V. Koi fishes through experimental infection. Results: Isolated bacteria were found Gram-positive paired and chained cocci, β-hemolytic and non-motile. Findings of biochemical and serological tests indicate that the isolated bacterium belongs to Group B Streptococcus of Lancefield classification. PCR result also confirmed that the bacteria were S. agalactiae. The bacterial isolates possessed resistance property against all the seven antibiotics used in this study. The isolated GBS was found highly virulent and showed 80%–90% mortality for Tilapia and V. Koi fishes in experimental infection within 1–6 days of post-infection. Conclusion: From the findings of this study, it may be concluded that isolated GBS from the Tilapia and V. Koi fishes were highly virulent and possessed multidrug-resistance properties. J. Adv. Vet. Objective: The study was carried out for molecular characterization and antibiotic resistance analysis of Escherichia coli isolated from different turkey farms in Dinajpur, Bangladesh. Materials and methods: A total of 45 samples comprising feces (n = 23) and cloacal swabs (n = 22) were collected randomly from turkeys. The samples were subjected to isolation and identification of E. coli by cultural and biochemical characteristics, followed by polymerase chain reaction and sequencing. An antibiogram of the isolated E. coli isolates was carried out by following the Kirby– Bauer disk diffusion method. Results: Out of the 45 samples, 28 (62.21%) were positive for E. coli, of which 16 (35.55%) fecal samples were positive and 12 (26.66%) cloacal swabs were positive . The antibiotic sensitivity analysis revealed that all the E. coli isolates were 100% sensitive to levofloxacin, norfloxacin, neomycin, gentamicin, and nitrofurantoin. On the other hand, all the isolates were 100% resistant to amoxicillin, azithromycin, erythromycin, tetracycline, bacitracin, cephalexin, nalidixic acid, vancomycin, methicilin, piperacillin, pefloxacin, novobiocin, cefepime, trimethoprim, netilmicin, and aztreonam. Conclusion: This study’s results uncover the occurrence and antibiotic resistance pattern of E. coli in the study area’s turkeys. J. Adv. Vet. Anim.
