Effect of topically applied demecarium bromide and echothiophate iodide on intraocular pressure and pupil size in Beagles with normotensive eyes and Beagles with inherited glaucoma

Topically applied demecarium bromide (0.125 and 0.25%) and echothiophate iodide (0.125 and 0.25%) solutions were evaluated in Beagles with normotensive eyes and Beagles with inherited glaucoma. In single-dose studies, the effects of intraocular pressure (IOP) and pupil size (PS) were measured in eyes before drug treatment and in drug- and nondrug-treated eyes. Both concentrations of the 2 drugs induced long-term miosis and decrease in IOP in normotensive eyes of Beagles and of eyes of Beagles with inherited glaucoma. Demecarium bromide (0.125 and 0.5%) decreased IOP for 49 and 55 hours, respectively. Echothiophate iodide (0.125 and 0.5%) reduced IOP for 25 and 53 hours, respectively. The miosis associated with both concentrations of the 2 drugs generally paralleled the decreases in IOP.

Respiratory tract infections are prevalent in foals, yet the frequency with which the distal airways are affected in chemical episodes of respiratory tract disease has not been evaluated to our knowledge. The objective of the study was to determine the incidence of distal respiratory tract infection (DRTI) in foals on a sample of Thoroughbred breeding farms (n = 10) in Ontario. In a pilot study, clinical criteria commonly used to select foals for antimicrobial treatment (detection of abnormal lung sounds, plus nasal discharge, cough, fever, tachypnea, and/or lethargy) were found to segregate foals with and without endoscopically confirmed DRTI. Mucopurulent exudate and bronchial erythema were observed more frequently (P < 0.005), bronchial lavage total cell count and neutrophil concentration were significantly (P < 0.005) higher, and intracellular cocci were recovered significantly (P < 0.01) more often from bronchial lavage samples of affected foals (n = 8) than of controls (n = 8). These clinical criteria were used to identify cases in a cohort of Thoroughbred foals (n = 219) from May 1 to October 30, 1991. Case morbidity adjusted for clustering was 82 +/- 5% (95% confidence limits, 72 to 92%). Most (74%) episodes of clinical DRTI were detected in july and August, and equal numbers were detected before (53%) and after (47%) weaning of foals. Of 178 cases, 66 (48%) were selected at random for endoscopy and bronchial lavage. Grade-II pharyngeal lymphoid hyperplasia was observed commonly (60% of foals); auditory tube diverticulum (guttural pouch) discharge was observed in 18 of 86 (21%) foals, and guttural pouch infection was confirmed in 6 of 7 foals examined endoscopically. Endoscopically confirmed DRTI, defined as visual detection of bronchial exudate with microscopic detection of intracellular cocci and markedly high neutrophil count in bronchial lavage samples, was confirmed in 75 of 86 (87%) cases tested. These data indicate that DRTI might be reliably diagnosed by auscultation during a simple rebreathing exercise. The syndrome of DRTI was extremely common in Thoroughbred foals, characterized by marked inflammation of visible airways and cytologic evidence of bacterial infection. Risk factors for clinical (undifferentiated) DRTI were not identified in this study.

Female Sprague-Dawley rats were treated IM with 0, 2.5, 25, and 50 micrograms of clenbuterol HCl/kg of body weight/d for 21 days. In all treated rats, significant increase in body weight gain (P < 0.05) and improvement in feed conversion ratio (P < 0.05) were recorded. Hydrometra was observed in the uterus of treated rats, and histologically, it was possible to see dilatation of luminal glands and ovarian alterations. Clenbuterol treatment induced significant (P < 0.05) increase in uterine estrogen receptor concentration of rats treated with the 2 higher doses. Treatment apparently failed to enhance the rate of oxidative and conjugative biotransformations, except for glucuronidation of p-nitrophenol (P < 0.05). On the basis of the data obtained, we could affirm that high doses of clenbuterol affect the female reproductive system of rats inducing, almost in part, estrogen-like modifications, but probably by a different mechanism of action correlated to intense adrenergic stimulation.

Bovine leukemia virus (BLV) infection and culling of cows in a commercial dairy herd were evaluated to determine whether a relation existed between the 2 factors. Cattle from the study population, a Holstein dairy herd consisting of approximately 400 milking cows, were tested for antibodies to BLV, using the agar gel immunodiffusion test, semiannually for 2 years, annually for 2 years, and when cattle were culled. Complete records of BLV test results were available for 849 (79%) of the 1,078 cattle that had at least 1 test during the study period. Using the Cox hazard model, the cull hazard rates (culls/cow-months) were greater for BLV seropositive cows than for seronegative cows > 36 months old. Hence, among older dairy cows, BLV-infected cows were culled prematurely, compared with uninfected cows.

Six Salmonella enteritidis bacterin formulations differing in adjuvant content and whole-cell inactivation procedures were evaluated in egg-laying chickens. Chickens given S. enteritidis bacterins containing modified Freund's incomplete adjuvant had greater humoral immune responses to S. enteritidis than did birds given other bacterin formulations (P < 0.05). Better protection against infection by S enteritidis phage types 8, 13a, and 23 was obtained in birds vaccinated with bacterin 5. Bacterin 5 contained S. enteritidis cells inactivated by 20% acetone and modified Freund's incomplete adjuvant.

Pharmacokinetic determinants of danofloxacin (1.25 mg/kg of body weight, IV) and its penetration into the respiratory tract tissues were studied in sixteen 4- to 6-week-old calves. The disposition curve was best described by an open 3-compartment model. Mean elimination half-life was 7.4 hours and the steady-state volume of distribution was 4.3 L/kg. The large volume of distribution was confirmed by a rapid and high penetration of the drug into respiratory tract tissues and secretions. In all structures (lung tissue, bronchial mucosa, bronchial secretions, and nasal secretions), danofloxacin concentration peaked 1 hour after drug administration. The area under the curve ratio for concentrations in tissue or secretions to concentrations in plasma was approximately 5 for lung tissue, 3 for bronchial mucosa, 0.85 for bronchial secretions, and 0.42 for nasal secretions. Protein binding of danofloxacin was 49% in plasma, 31% in bronchial secretions, and 14% in nasal secretions, resulting in consistently higher free danoflaxacin concentrations in bronchial secretions than in plasma. Accumulation of danofloxacin within bronchial mucosa and the high concentration of free drug in bronchial secretions suggested that an active process may be involved in the transport of danofloxacin across the airway epithelium. The dose of danofloxacin administered provided drug concentrations above the minimal inhibitory concentration of common respiratory pathogens for up to 12 hours in bronchial mucosa, up to 8 hours in bronchial secretions, and up to 4 hours in nasal secretions.

Middle carpal cartilage explants from 4 horses with mild osteoarthritis involving that joint were maintained in tissue culture to test the effects of a polysulfated glycosaminoglycan (PSGAG) on proteoglycan synthesis and degradation. Cultures were exposed to 0.025 or 25 mg of PSGAG/ml for 48 hours, after which the medium was replaced with medium containing similar doses of PSGAG and 35S. Subsequently, the sulfated proteoglycan content of the medium and extracts of the explants was measured. Gel filtration chromatography was used to estimate the size and to purify the principal, large proteoglycan monomer, which was further characterized by digestion, using glycosidic enzymes. In a second experiment, explants were incubated with 35S for 48 hours, and were subsequently exposed to the same concentrations of the PSGAG for an additional 48 hours. The amount of remaining labeled proteoglycan was determined for culture medium and cartilage extracts. Gel filtration chromatography was used to assess the hydrodynamic size of the large proteoglycan monomer. Aliquots of proteoglycans from the second experiment were incubated in high-molecular weight hyaluronate and chromatographed to assess reaggregation. Polysulfated glycosaminoglycan caused a significant (P < 0.04) decrease in sulfated proteoglycan synthesis by cartilage explants. Radioactive proteoglycan content in explants labeled prior to exposure to PSGAG were similar. Large proteoglycan monomer size was similar in both experiments (median partition coefficient [K(AV)] = 0.40), and was not influenced by PSGAG treatment. Prelabeled explants exposed to hyaluronate and chromatographed under associative conditions had similar proportions of the radiolabel eluting as proteoglycan aggregate. Enzymatic digestion of newly synthesized large monomer revealed a mild dose-dependent increase in the proportion of keratan sulfate substitution on core protein. It was concluded that PSGAG in vitro, at the dosages evaluated, caused a decrease in proteoglycan synthesis, had little effect on labeled proteoglycan degradation, did not influence the size of large monomer, and caused a modest increase in the concentration of keratan sulfate in proteoglycans synthesized by osteoarthritic equine chondrocytes.

Both ovaries from 88 bovine fetuses in the fifth month or later of gestation were studied histologically to determine the prevalence, origin, and time of appearance of atretic corpora lutea (ACL). Ovaries from 36 (41%) fetuses had ACL; fetuses < 6 months of gestation did not have ACL. Six fetuses had more than 25 ACL, but there was no apparent relationship between fetal age and number of ACL. Formation of ACL involved disintegration of the stratum granulosum of secondary follicles, concomitant with proliferation and invasion by vascularized elements of the theca. Fully developed ACL consisted of a large primary oocyte surrounded by a prominent zona pellucida and encased in a well-vascularized, largely thecal, fibrocellular wall. They measured approximately 0.5 to 1.0 mm in diameter. Empty, collapsed zona pellucidas were seen in many of the degenerating ACL.

Saline (0.9% NaCl) solution, flunixin meglumine (1.1 mg/kg), prednisolone sodium succinate (1.1 mg/kg), U74389F (1.5 mg/kg), and dimethyl sulfoxide (0.5 g/kg) were each administered IV to 5 neonatal calves 15 minutes after the start of a 3-hour infusion of Escherichia coli lipopolysaccharide (LPS; 2 micrograms/kg/ hr). Four additional calves were given a 3-hour IV infusion of saline solution alone. Only flunixin significantly suppressed eicosanoid production and mitigated clinical signs associated with endotoxemia. Prednisolone provided partial protection against LPS-induced hypotension and lacticemia. Pronounced hypoglycemia and lacticemia were observed in U74389F-treated calves; LPS-induced hypotension and hypoglycemia were marked in dimethyl sulfoxide-treated calves.

Two 2.5-cm2 full-thickness skin wounds were created surgically over the lateral aspect of the cannon bone of each limb of 6 horses (n = 48 wounds). Dressings evaluated were a nonadherent gauze pad (group 1); a synthetic semiocclusive dressing, (group 2); equine amnion (group 3); and a synthetic fully occlusive dressing (group 4). Wounds were assessed subjectively at each dressing change, and total wound area, area of granulation tissue, and area of epithelium in each wound were determined by computerized digital analysis of photographs of the wounds. Complete healing time (wound covered by epithelium) also was determined for each wound. Statistical comparisons were made, using Kruskal-Wallis analysis and a Mann-Whitney U test. Median time to complete healing was: group 1, 53 days; group 2, 71 days; group 3, 63 days; and group 4, 113 days. Time to complete healing was significantly longer for wounds of group-4 horses than all other groups, and wounds of group-1 horses healed faster than did those of group-2 horses (P < 0.05). Wounds in group-4 horses required significantly (P less than or equal to 0.05) more excisions of granulation tissue (median, 11.5 times) than did those in group-1 (median, 3.5), group-2 (median, 5.5) or group-3 (median, 2.5) horses. Epithelial tissue was detected later in wounds of group-4 horses (median, 27 days) than in wounds of horses in groups 1, 2 or 3 (median, 17 days); however, this difference was not statistically significant. Significant differences were not found for percentage of healing attributable to wound contraction or epithelialization. Use of synthetic semiocclusive and fully occlusive dressings resulted in significantly (P < 0.05) prolonged healing and production of excess wound exudate, compared with control wounds. In this model, occlusion of wounds was not beneficial for healing of full-thickness skin wounds of the distal portion of the limbs of horses.
