Leptin inhibits hepatocyte growth factor-induced ductal morphogenesis of bovine mammary epithelial cells

We examined the effect of stroma-derived factors, hepatocyte growth factor (HGF) and leptin, on morphological differentiation of bovine mammary epithelial cells (BMEC) in collagen gel three-dimensional culture in vitro. BMEC treated with HGF, but not leptin, formed duct-like organoids. The formation of organoids by HGF was enhanced by treatment with a mixture of insulin, cortisol and prolactin, while BMEC treated with the mixture alone did not produce the organoid. In contrast, the formation of organoids by HGF was dose-dependently inhibited by simultaneous addition of leptin, regardless of the presence or absence of the hormone mixture. These results suggest that stroma-derived factors intricately regulate mammary epithelial morphogenesis.

An R664X nonsense mutant AE1 is responsible for dominant hereditary spherocytosis in cattle and is degraded by the proteasomal endoplasmic reticulum-associated degradation. The present study demonstrated that R664X AE1 translated in vitro had the trypsin-sensitve site identical to that of the wild-type AE1. The P661S/R664X mutant containing a possible N-glycosylation site at Asnsup(660) showed an increase in size by 3 kDa both in the cell-free translation system and in transfected HEK293 cells. Moreover, steady state levels of R664X and P661S/R664X in HEK293 cells were markedly increased in the presence of a proteasome inhibitior. These findings indicate that the truncated C-terminal region of R664X AE1 has lumenal localization in the endoplasmic reticulum and is not accessible to proteasomal machineries in the cytosol.

In our previous study, it was demonstrated that the administration of anion salts, which slightly lower the dietary cation-anion difference (DCAD), in the prepartum period is safe and effective for preventing milk fever in multiparous cows. In the present study, several clinico-pathological constituents in serum and urine, which might be related to milk fever, were analyzed using stored samples from the previous study to identify clinico-pathological parameters for easily evaluating the efficacy of lowering DCAD and to further investigate the mechanism by which lowering DCAD prevents milk fever. Among the parameters analyzed in the present study, inorganic phosphorus (iP) was involved in milk fever because the serum concentration and urinary excretion of iP were significantly higher in the group of primiparous cows (heifer group), which did not develop hypocalcemia, than those in other groups of multiparous cows. Serum chloride concentrations in the heifer group and the group of multiparous cows fed anion salts (anion group) tended to remain higher than those in other control groups of multiparous cows suggesting that serum chloride concentration may be utilized for evaluating the status of metabolic acidosis and the efficacy of lowerng DCAD in dairy cows fed anion salts. In addition, plasma estradiol-17beta concentration in the heifer group tended to be lower at parturition compared with that in other multiparous groups suggesting that estrogen known as a potent inhibitor of bone resorption may be involved in developing milk fever.

Hantaviruses are causative agents of some severe human illnesses, including hemorrhagic fever with renal syndrome (HFRS) and hantavirus pulmonary syndrome (HPS). The viruses are maintained by rodent hosts, and humans acquire infection by inhaling virus-contaminated excreta from infected animals. To examine the epidemiology of hantavirus infections in Japan and Far East Russia, we conducted epidemiological surveys in these regions. In Japan, anti-hantavirus antibodies were found in four rodent species, Clethrionomys rufocanus, Rattus norvegicus, R. rattus, and Apodemus speciosus. Although no new HFRS cases have been officially reported over the past 20 years in Japan, one member of the Japan Ground Self-Defense Force did test positive for hantavirus antibody. Repeated surveys in Far East Russia have revealed that two distinct hantavirus types cause severe HFRS in this region. Hantavirus sequences identified from A. peninsulae, fetal HFRS cases in Vladivostok, and Amur virus are highly similar to each other (92% identity) , but they are less similar (-84% identity) to the prototypical Hantaan virus, which is carried by A. agrarius. Phylogenetic analysis also indicates that Amur and A. peninsulae -associated viruses are distinct from Hantaan virus, suggesting that A. peninsulae is the reservoir animal for Amur virus, which causes severe HFRS. From HFRS patients in the Khabarovsk region, we identified viruses with nucleotide sequences that are more similar to Far East virus (96%identity) than to the Hantaan (88-89%identity) or Amur (81-83% identity) viruses. Phylogenetic analysis also indicates that the viruses from Khabarovsk HFRS patients are closely related to the Far East virus, and distinct from Amur virus.

This article summarizes the different types of free radicals, antioxidants and the effect of oxidative stress on the activities of glutathione peroxidase and glucose 6-phosphate dehydrogenase in bovine liver diseases. A growing body of evidence suggests that the formation of reactive oxygen species is a common occurrence associated with most if not all disease processes. The overall importance of reactive oxygen species to the progression and severity of various disease states varies greatly depending on the conditions and whether the disease is acute or chronic. Free radical researches in animals are in progress and further investigations are needed to establish the involvement of reactive oxygen species in diseases affecting different animal species and the pathology they produce.

In order to evaluate the present epidemiological situation of Trichinella infection in wild animals in Hokkaido, Japan, red foxes (Vulpes vulpes), raccoon dogs (Nyctereutes procyonoides), brown bears (Ursus arctos), martens (Martes melampus), rodents and insectivores captured in Hokkaido were examined for muscle larvae by the artificial digestion method from 2000 to 2006. Foxes (44/319, 13.8%), raccoon dogs(6/77, 7.8%) and brown bears (4/126, 3.2%) were found to be infected with Trichinella larvae and all other animal species evaluated were negative. Multiplex PCR and DNA sequencing revealed that larvae from a fox captured in Otofuke, in south-eastern Hokkaido, were T. nativa, and larvae from 27 animals including 21 foxes, 2 raccoon dogs and 4 brown bears captured in western Hokkaido were Trichinella T9.

Squamous cell carcinoma is a malignant tumour of keratinocytes. There are several factors associated with the development of squamous cell carcinoma, including prolonged exposure to ultraviolet light, lack of a pigmented skin, lack of hair or a very sparse hair coat at the affected sites. A retrospective study of squamous cell carcinoma from 1978 to 2002 in farm animals was made. The samples were obtained at the Regional Diagnostic Laboratory of the Veterinary School in Pelotas, Southern Brazil. The distribution of the neoplasm among the species was as follows: 30 in cattle, 7 in sheep and 13 in horses. Epidemiological data as breed, sex, age, and anatomical location of the tumors were statistically analyzed. The histological classification of the tumors graded from well differentiated to poorly differentiated. A multifactorial etiology of squamous cell carcinoma in farm animals is suggested although more retrospective and prospective studies are needed, to determine the role of each factor in the pathogenesis of the neoplasm. | O carcinoma de células escamosas (CCE) é um tumor maligno dos queratinócitos. O desenvolvimento desse tipo de tumor está associado a muitos fatores, incluindo a exposição prolongada à luz ultravioleta, falta de pigmento na epiderme, perda de pêlos ou cobertura de pêlos muito esparsa nos locais afetados. Realizou-se um estudo retrospectivo dos casos de carcinoma de células escamosas em animais de produção (bovinos, ovinos e eqüinos) de 1978 a 2002. O material foi obtido nos arquivos do Laboratório Regional de Diagnóstico da Faculdade de Veterinária da Universidade Federal de Pelotas. Os 50 tumores do tipo carcinoma de células escamosas estavam distribuídos nas diferentes espécies (bovina: 30; ovina: sete e eqüina: 13). Dados como raça, sexo, idade e localização corpórea, dentro de cada espécie, foram tabulados. Classificaram-se os tumores segundo seu grau de malignidade em bem diferenciados a pobremente diferenciados, através de avaliação histológica. A etiologia multifatorial dos cce em animais de produção requer mais estudos retrospectivos e prospectivos, para determinar o papel de cada fator na patogênese desse neoplasma.

Avaliaram-se os efeitos do fornecimento de diferentes frações inspiradas de oxigênio (FiO2) sobre a dinâmica respiratória em cães submetidos a infusão contínua de propofol e mantidos em ventilação espontânea. Oito cães foram submetidos a cinco anestesias diferenciando uma da outra pela FiO2 fornecida. Formaram-se cinco grupos denominados G100 (FiO2 = 1), G80 (FiO2 = 0,8), G60 (FiO2 = 0,6), G40 (FiO2 = 0,4) e G20 (FiO2 = 0,21). Os animais foram induzidos à anestesia com propofol na dose necessária para intubação, e ato contínuo iniciou-se a infusão do fármaco e o fornecimento de oxigênio conforme a FiO2 determinada para cada grupo. As primeiras mensurações foram efetuadas 30 minutos após o início da infusão do anestésico (M0), em seguida em intervalos de 15 minutos (M15, M30, M45 e M60). Os dados numéricos obtidos foram submetidos a Análise de Variância, seguida pelo teste Tukey (p<0,05). Os valores registrados de saturação de oxihemoglobina (SpO2) e SaO2 foram significativamente menores para G20, enquanto que a pressão parcial de oxigênio no sangue arterial (PaO2) variou conforme a fração de oxigênio fornecida. Para a pressão parcial de dióxido de carbono no sangue arterial (PaCO2) foram registradas diferenças em M30, no qual G100 apresentou médias superiores a G20. Conclui-se que o fornecimento de oxigênio a 100%, 80% e 21% devem ser evitados, pois proporciona prejuízos significativos ao sistema respiratório de cães anestesiados com infusão contínua de propofol na dose de 0,7mg/kg/min sob ventilação espontânea. | The effects of several inspired oxygen fractions (FiO2) on the respiratory dynamics in spontaneously breathing dogs submitted to continuous infusion of propofol were evaluated. Eight adult mongrel dogs were used. Each animal underwent five anesthesias, in each procedure the patient was allowed to breath a different FiO2, thereby resulting in five groups, namely: G100 (FiO2 = 1), G80 (FiO2 = 0.8), G60 (FiO2 =0.6), G40 (FiO2 = 0.4), and G20 (FiO2 = 0.21). To induce anesthesia, propofol was given until the animals allowed orotracheal intubation, followed by immediate continuous infusion of drug. The initial measurement (M0) was recorded thirty minutes after the infusion of propofol has begun. Additional recordings were performed at 15 minute intervals for 60 minutes (M15, M30, M45 and M60). Numeric data were submitted to Analysis of Variance followed by Tukey Test (p<0.05). We recorded significantly lower values of SpO2 and SaO2 at G20, whereas PaO2 varied according to the changes in oxygen. Regarding PaCO2, the mean of G100 was greater than G20 at M30. However, spontaneously breathing dogs anesthetized with propofol at the rate of 0.7mg/kg/min should not be provided with 100%, 80%, and 21% oxygen owing to the significant compromise of respiratory system.

Objetivo do estudo foi avaliar a cinética da resposta inflamatória induzida experimentalmente com BCG em peixes modernos pertencentes ao gênero Centropomus sp. Os animais foram experimentalmente inoculados com BCG por via intramuscular na região do pedúnculo caudal sendo realizada a coleta do material nos tempos experimentais de 1, 3, 7, 14, 21 e 33 dias pós-inoculação. A fase aguda da resposta inflamatória se mostrou na forma de infiltrado composto predominantemente por células mononucleares, edema intersticial e necrose de tecido muscular. À medida que o processo se desenvolve, observa-se a organização do foco lesional na forma de um granuloma formado por células epitelióides. Essas células, ao exame imunohistoquímico apresentaram positividade a proteína S100 e citoqueratina, indicando características macrofágicas e secretória, além de apresentarem ao exame ultra-estrutural a presença de desmossomos entre as células adjacentes. Não houve participação efetiva de células gigantes e pigmentares, sugerindo ser uma característica relacionada à espécie. | This study set out to evaluate inflammatory response kinetics after experimental inoculation with BCG in modern Centropomus sp. fish. BCG was applied through intramuscular injection in the caudal peduncular region, and samples were collected for analyses at days 1, 3, 7, 14, 21, and 33 post-injection. Acute phase inflammatory infiltrate was characterized by predominant mononuclear cells, intersticial edema, and muscular tissue necrosis. As the inflammatory response evolved, the lesion organized into a granuloma formed by epithelioid cells. These epithelioid cells were positive for the S100 protein and cytokeratin at histochemical analysis, which demonstrates macrophage and secretory activity, besides ultra-structural analysis showed desmosomic junctions within adjacent cells. It didn't have an expressive participation of giant cells and pigment containing cells in the inflammatory reaction, suggesting that it's a specie-specific characteristic.

O objetivo do presente trabalho foi caracterizar a população de cães portadores de ruptura do ligamento cruzado cranial (RLCC) para fatores considerados de risco como raça, idade, sexo e peso corporal. A base de dados consistiu de informações resgatadas de prontuários de 323 cães com diagnóstico de ruptura de ligamento cruzado cranial atendidos no Serviço de Cirurgia de Pequenos Animais do Hospital Veterinário da Faculdade de Medicina Veterinária e Zootecnia da Universidade de São Paulo em um período de 7 anos (1999 a 2005). Os resultados mostraram que as raças de maior prevalência foram o Leonberger (100%), Cane Corso (66,7%), Dogue de Bordeau (50%), Starffordshire Terrier (40%) e o Chow Chow (36%), estando ainda o Rottweiler (11,6%) e o Labrador Retriever (8,1%) em 10º e 13º lugares, respectivamente. As idades de maior freqüência foram 2, 4, 3, 7 e 6 anos (média = 5,58 anos). Fêmeas (59,14%) foram mais acometidas do que machos (40,86%) e animais inteiros (76,15%) foram mais representativos do que animais castrados (17,76%). Quanto ao peso, entre 6 e 15 kg (32,82%) seguido por animais entre 36 e 45 kg (17,03%). Pode-se observar que os dados em nosso meio tendem a se assemelhar com a literatura estrangeira: ocorrência da RLCC em animais mais jovens de raças de grande porte e gigantes, o Rottweiler como uma raça em comum a fêmea é mais representativa do que o macho. | The aim of this research was to characterize in the canine population the risk factors for cranial cruciate ligament rupture, which are breed, age, sex and body weight. The data base consisted of collected information from medical records of 323 dogs bearing cranial cruciate ligament rupture attended during a period of 7 years (1999 to 2005) in the Small Animal Surgical Service of the Veterinary Hospital of the Veterinary Medicine Faculty at São Paulo University. The results showed that the most prevalent breed were Leonberger (100%), Cane Corso (66,7%), Dogue de Bordeau (50%), Starffordshire Terrier (40%) e o Chow Chow (36%). Also, Rottweiller (11,16%) and Labrador Retriever (8,1%) occupied the 10º e 13º positions, respectively. The ages 2, 4, 3, 7 and 6 (mean = 5,58) years old had the highest prevalence of cranial cruciate ligament rupture. Female (59,14%) were more represented than males (40,86%) and sexually intact animals (76,15%) were more represented than neutered dogs (17,76%). The group f dogs weighing between 6 and 15 kg and between 36 and 45 kg showed the highest incidence. In this research was observed that the database of the risk factors for the dogs bearing cranial cruciate ligament rupture examined were similar to the that seen in the foreign literature. There is a tendency for rupture of ligament in an earlier age, large breeds and giant dogs. The Rottweiler was a common breed among our study and that of the foreign literature. Also, female were more represented than male dogs.
