Cellular immune responses induced in vitro by Ehrlichia ruminantium secreted proteins and identification of vaccine candidate peptides

Secreted proteins are reported to induce cell-mediated immunity characterised by the production of interferon-gamma (IFN)-γ. In this study three open reading frames (ORFs) (Erum8060, Erum7760, Erum5000) encoding secreted proteins were selected from the Ehrlichia ruminantium (Welgevonden) genome sequence using bioinformatics tools to determine whether they induce a cellular immune response in vitro with mononuclear cells from needle and tick infected animals. The whole recombinant protein of the three ORFs as well as four adjacent fragments of the Erum5000 protein (Erum5000A, Erum5000B, Erum5000C, Erum5000D) were successfully expressed in a bacterial expression system which was confirmed by immunoblots using anti-His antibodies and sheep sera. These recombinant proteins were assayed with immune sheep and cattle peripheral blood mononuclear cells (PBMCs), spleen and lymph node (LN) cells to determine whether they induce recall cellular immune responses in vitro. Significant proliferative responses and IFN-γ production were evident for all recombinant proteins, especially Erum5000A, in both ruminant species tested. Thus overlapping peptides spanning Erum5000A were synthesised and peptides that induce proliferation of memory CD4+ and CD8+ T cells and production of IFN-γ were identified. These results illustrate that a Th1 type immune response was elicited and these recombinant proteins and peptides may therefore be promising candidates for development of a heartwater vaccine.

A total of 103 blood samples from pet dogs around Ipoh were screenedfor common blood protozoa. A total of 14 samples were found positive for Erhlichia canis and one sample was found positive for microfilaria of Dirofilaria immitis. Both these diseases are transmitted by vectors; ticks (Rhipicephalus sangiuneus) and mosquitoes respectively. In the hot and wet tropical environment where vectors are abundant, pet care, hygiene and regular screening will help veterinarians detect these infections early to facilitatetreatment.

A study was carried out to analyse the efficacy of neem leaf powder, administered in a capsule against coccidiosis in young goats as well as to identify the species of coccidia found in a selected private farm in Sungai Siput, Perak. A total of seven Eimeria spp were found, in faecal samples from the experimental goats, namely; E. arloingi,E. hirci, E. alijevi, E. christenseni, E. jolchijevi, E. ninakohlyakimovae and E. caprina at a rate of 40%, 23%, 14%, 7%, 5% and 2% respectively. Two types of treatment for coccidiosis, herbal and synthetic, were given to 24 young goats for a period of 8 weeks to evaluate the effectiveness of a herbal product, that is the neem leaf powder capsule, a product of the Veterinary Research Institute.Results show that there is no significant differences (p>0.01) between neem capsule treatment and a synthetic drug (sulphurbased) treatment in treating coccidiosis infection. This study shows that neem leaf powder can be an effective substitute for controlling coccidiosis in goats

Objective: To evaluate the incidence, etiology and progression of Pigmentary keratitis in dogs. Materialsand Methods: A total of 83 corneas from 55 dogs of different breeds, sex and age were selected for the study. Signalment, anamnesis, nature of discharge and duration of illness was collected from all the animals.The progression of pigmentation was assessed by dividing the cornea in to four quadrants. Pigmentation grading, extent of pigmentation and mean pigment density were calculated by dividing the cornea in to 24 sectors. Schirmer tear test (STT), fluorescein dye test (FDT), tonometry, and slitlamp biomicroscopy and Cornealimpression cytology were done. Results: Among the 55 animals, 51 dogs were Chinese Pug (92.7%) and the mean age was 33.13 ± 3.12 months. Among 55 animals, 28 were females (50.9%) and left cornea was affected in 44 animals (53.01%). The mean duration of the disease as noticed by the owner was 07.21 ± 0.65 months.Most of the owners were totally unaware about the condition of the eye. Among 83 corneas, 40 (35%) showed pigmentation in all the sectors. 29 animals (53%) wereaffected with keratoconjunctivitis sicca (KCS) followed by 13 animals (24%) affected with entropion. The mean value of random blood sugar was 107.84 ± 0.99 and the mean intraocular pressure in the animals under the study was 40.64 ± 2.38. The mean value of pigmentation grading,extent of pigmenta ion and mean pigment density was 32.59 ± 2.27, 15.67 ± 0.83 and 1.37 ± 0.07 respectively. The mean value of Schirmer tear test was 10.31 ± 0.58 andunder high power microscopy, Leishman’s stained corneal impression cytology revealed infiltration of neutrophils in all the slides. Conclusion: It was concludedthat Chinese pugs under the age of 3 years are mostly affected and females and left eye is mostly affected. All the animals with pigmentation is having KCS.

Ovary plays a vitalrole in the reproductive biology andbiotechnology of female animals. In thisstudy, both the right and left ovaries ofthe Black Bengal goat were collected fromthe slaughter houses of different Thanasin the Mymensingh district. For each ofthe specimens, gross parameters such asweight, length and width were recorded.Then they were processed and stained withH&E for histomorphometry. This studyrevealed that the right ovary (0.53 ± 0.02g) was heavier than the left (0.52 ± 0.02 g).The length of the right ovary (1.26 ± 0.04cm) was lower than the left (1.28 ± 0.02cm) but the width of the right (0.94 ± 0.02cm) was greater than the left (0.90 ± 0.03cm). The diameter of ovarian follicles inthe cortex was measured as primordial 39.6± 6.61 µm, primary single layer 54.0 ± 4.06µm, primary multi-layer 147.6 ± 11.04 µm,secondary with C-shaped antrum 449.5 ±75.71 µm and graafian 1.3 ± 0.20 mm. Inthe graffian follicle, the thickness of thegranulosa cell layer was 79.2 ± 11.04 µm,theca interna 75.76 ± 6.82 µm, theca externa130.07 ± 12.53 µm and the oocyte diameterwas 109.8 ± 5.75 µm. In 2014, a total of 16 riverterrapins (Batagur affinis), locally calledtuntung sungai, died due to variouscauses and a post mortem was carriedout. Complete diagnostic evaluationof parasitological and bacteriologicalfindings were recorded. Significantparasitological findings consisted of seventerrapins showing presence of Ascarid(Sulcascaris sulcata) and Strongyle(Oesophagostomum sp.) worms in the gutcontents. Strongyle and strongyloides eggswere detected using McMaster’s method onfaeces of 12 terrapins. Bacterial culturesfrom organs indicated the presence ofE. Coli, Staphylococcus epidermidis,Staphylococcus chromogenes, Aeromonashydrophila (4+) dan E.Coli (4+). Thisindicates that common parasites andbacteria are important in the conservationprogrammes for river terrapins andmeasures to curb the infection is highlyrecommended. Continuous data collectionwill enable its management and assessmentin control programmes for a more effectiveconservation programmes.

Trypanosomiasis positive cases were reported in Sahom Farm Retreatin Gopeng, Perak; with multispecies livestock animals. Nzi and Vavoua traps were applied to survey the population of biting flies; stable flies (Muscidae: Stomoxyinae) and horse flies (Tabanidae)as the vector for surra. Results indicated the presence of Trypanosomiasis infection diagnosed by buffy coat examination, thinblood stained smears and serological test (Surra Sero K-Set test) and identification of its insect vectors. The presence of bothbiting flies provides the missing link between the occurrence of the disease and host or environmental factors precipitatingthe disease. Besides trypanosomiasis in cattle, other parasitic infections were also recorded with heavy infections for liver fluke (Fasciola gigantica ova) and coccidia oocysts. Therefore, some control measures are recommended to eradicate the vectors and to treat infected animals in order to prevent the dissemination ofthe trypanosmiasis.

The objectives of the present study were to determine the infection rate of equine piroplasmosis (EP) in horses and ponies in Kelantan,Malaysia and compare the microscopic examination with competitive enzymelinked immunosorbent assay (cELISA) test as methods for diagnosis of EP. 306 blood samples were randomly collected from equids including 148 horses and 158 ponies in various districts of Kelantan, from September 2013 to March 2014. Based on microscopic examination of the staining blood smears, the infection rates ofTheileria equi, Babesia caballi and of both infections in horses were 19.59%, 25% and 8.78% respectively, whereas in ponies theinfection rates were 14.55%, 19.62%, and 5.69% respectively. Based on cELISA test, the infection rates of T. equi, B. caballi and of both infections in horses were 50.67%, 62.16% and 33.10% respectively,whereas in ponies, the infection rates were 51.89%, 63.92% and 35.44% respectively. No significant difference were observed between equids species associated with a seroprevalence of T. equi, B. caballi andof both infections (P≤ 0.05). According to the Kappa value there was no compatibility between microscopic examination and cELISA on the diagnosis of T. equi, B. caballi and of both infections which were 0.235, 0.013 and 0.080 respectively. In conclusion, the current results for this research work indicate that equine piroplasmosis is widespread in Kelantan, Malaysia and cELISA test is more efficientthan microscopic examination for diagnosis of EP.

A 2-year-old (63 days) pregnant female dachshund dog was presented to the Teaching Veterinary Clinical Complex (TVCC) with complaint of a progressively growing swelling in the caudal ventral abdominal area. History, physical examination, radiography and ultrasonography confirmed an inguinal hystrocoele. A ventral midline incisionon the mass was made to relieve the foetuses and reduction of contents, and herniorrhaphy was performed. Sutures were removed on 7th post-operative day and the animal had an uneventful recovery.

A total of thirty-eight Mafriwal cattle were selected from a localcattle herd of a government cattle farm; of which 36 animals were sub-fertile Mafriwal female dams and two bulls which were considered as control animals (one male Mafriwal and one male Jersey). Two markers were used in the detection of Y chromosome in the sub-fertile female animal which are testis specific proteins Y-encoded (TSPY) and amelogenin (AMLX/AMLY) genes. The genes were amplified using PCR. The DNA bands from a normal male for TSPY gene size was approximately 260 bp while AMLX/ AMLY gene were approximately 341 and 467 bp. The examination of all samples showed that the sub-fertile cow revealedonly 467 bp while three fragments were detected in the control group; 260 bp (testis specific protein, Y-encoded gene), 341 and 467 bp (Amelogenin gene). The results showed that the sex chromosomeanomalies associated with Y chromosome did not occur in this group. These two sex markers can be used for the diagnosis of Y chromosome abnormality in a sub-fertile cow through polymerase chain reactionwhich is a rapid and reliable method for use in breeding herds.
