Preliminary study on designing the binder of sperm-1 synthetic vaccine using sequence-based methods and molecular docking

Objective: The main objective of this study is to design a synthetic vaccine from the binder of sperm-1 (BSP1). Materials and Methods: This study was carried out using bioinformatics-related techniques. BSP-1 has been chosen as one of the biomarkers of a ruminant's male fertility. We hypothesize that the BSP1 synthetic vaccines, which contain T-cell epitopes, can produce antibodies more effectively for the development of a sperm fertility detection kit. A sequence of BSP-1 peptides A0A0K1YXR5 from Bubalus bubalis (Domestic water buffalo) origin has been decided to be used to develop the peptide vaccine. Results: In this study, we succeeded in making synthetic vaccines from BSP-1 with a peptide sequence of LPEDSVPDEERVFPFTYRNRKHF. The three-dimensional theoretical prediction analy¬sis of the peptide binding pattern to its ligand, as well as the molecular docking, has also been revealed. Conclusions: A synthetic vaccine from the BSP-1 has been developed in this study with the amino acid sequence LPEDSVPDEERVFPFTYRNRKHF, which is buffer-soluble, and the three-dimensional theoretical prediction analysis of the peptide binding pattern of BSP-1 to its ligand, as well as molecular docking, has also been revealed. Objective: We investigated the effects of a source of selenium [inorganic or nano-selenium (nano-Se)] on female V-line rabbits with or without injection of ivermectin (IVM). Material and Methods: Eighty four rabbits (12 weeks old) were randomly divided into 4 groups of 21 rabbits each with the basal diet supplemented as per the following treatments: G1 (control): inorganic Se at 0.3 mg/kg diet with no IVM injection; G2: inorganic Se with IVM injection; G3: nano-Se with no IVM injection; and G4: nano-Se with IVM injection. IVM was injected subcutane¬ously at 0.2 mg/kg body weight starting when the does were 14 weeks old and repeated weekly for five consecutive weeks. Results: Replacement of inorganic Se with nano-Se improved body weight and total body weight gain, total feed intake, average feed conversion ratio, and reproductive performance (age at puberty, number of service/conception, conception rate, number of kits/litter, and lit¬ter weight at birth). Similarly, sexual activity of does, serum estrogen levels, and serum levels of antioxidants (glutathione reduced, catalase, and malondialdehyde) increased in nano-Se-sup¬plemented groups. Ivermectin treatment in inorganic Se-supplemented groups was detrimental to growth and reproductive performance, while these parameters improved in IVM-treated and nano-Se-supplemented groups. Objective: Most people in Matrouh Governorate consume camel milk as a treatment for many diseases in a raw state to obtain nutritive value. Raw dromedary camel milk can be contaminated by Escherichia coli through fecal matter at any point of milk handling; therefore, it may lose its value and safety specifications. This survey aimed to estimate the incidence of E. coli in fresh camel milk. Materials and Methods: 100 fresh camel milk samples (50 from markets and 50 from farms) were randomly collected from different districts in Matrouh Governorate, Egypt, over 4 months for the detection of E. coli incidence through conventional bacterial isolation, molecular investigation, and gene sequencing. Results: The prevalence rates of E. coli in the examined market and farm raw camel milk based on conventional methods were 24% and 8%, respectively, while those by molecular identification using phoA as an E. coli determinate gene were 4% and 6%, respectively. Moreover, E. coli phoA gene phylogenetic analysis revealed high sequence similarity to E. coli strain CP033158.1 in India and E. coli strain CP047594.1 in China. Antibiotic sensitivity of E. coli isolates showed high suscep¬tibility to norfloxacin (10 μg) and cefoperazone (75 μg). On the other hand, high resistance was found in rifamycin (30 μg) and cefoxitin (30 μg). Conclusion: The results indicate that market camel milk is more contaminated than the farms' own. Objective: The work aimed to assess the safety and quality of broiler meat in experimental liste¬riosis changes in storage. Materials and Methods: Ross Cobb 500 chickens (40) were divided into 4 groups of 10 animals each. Chickens from three experimental groups were infected by Listeria innocua, Listeria ivanovii, and Listeria monocytogenes. Meat samples were stored for 5 days at 0°C–4°C. Meat samples were kept in the refrigerator for 3, 4, and 5 days. Microbiological and laboratory indicators of meat freshness were found on these days as well. Results: After the slaughter of chickens with experimental listeriosis, pathological changes in mus¬cles and organs were noted against the background of fattening carcasses with a high slaughter yield. By bacterial contamination, 1 day after slaughter, the meat of chickens of the experimental groups (L. innocua, L. ivanovii, and L. monocytogenes) outperformed the control group by almost 1.9, 13.9, and 24.7 times, respectively (p < 0.05). The same trend is observed for the third, fourth, and fifth days of meat storage. To keep chicken meat fresh for 5 days, only samples from the con¬trol group stayed fresh. Conclusion: According to the total bacterial contamination, the meat of chickens of the groups L. innocua and L. ivanovii was dangerous to human health after 5 and 4 days of storage, respec¬tively. From the first day after the chickens were killed, the meat of chickens that had been infected with L. monocytogenes did not meet the requirements (up to 100 CFU/gm) and was not safe to store or eat. Objective: The current broiler trial was planned to assess the effects of Kolin Plus™, a polyherbal formulation (PHF), on performance, protein and fat accretion, and serum L-carnitine (LC) levels in broilers fed a high-energy diet (HED). Materials and Methods: A total of 500 1-day-old Cobb 430 male chicks were assigned to 5 treat¬ment groups consisting of 10 replicates, with 10 birds in each replicate (n = 100). Group G1 was a negative control fed HED, and group G2, a positive control supplemented with synthetic choline chloride (SCC) 1,500 gm/ton in HED. Groups G3, G4, and G5 were supplemented with PHF in HED at 400, 500, and 750 gm/ton feed, respectively (PHF400, PHF500, and PHF750). Results: The PHF produced a dose-dependent numerical improvement in body weight, feed con¬version ratio, livability, and the European Production Index. There were no changes in carcass nitrogen and protein accretion, whereas a statistically significant decrease (p < 0.05) in carcass fat and fat accretion was observed in the SCC and PHF groups. Moreover, PHF showed a significant increase in serum LC levels. Conclusion: Kolin Plus™ improves performance parameters akin to SCC by improving fat metabo¬lism and mobilization by enhancing serum LC levels and restoring normal fat accretion. Objective: Many bacteria are involved in causing mastitis in dairy cows. Perfect identification of bacteria is crucial for the appropriate choice of drug for treatment. This study aims to find out the various bacteria that cause mastitis through the 16S ribosomal ribonucleic acid (16S rRNA) gene. Materials and Methods: A total of 150 mastitis somatic cell samples were tested with bacterial nested polymerase chain reaction (PCR) universal primers, targeting the 16S rRNA gene. The primers had both Gram-positive and Gram-negative bacterial specificities. Inflammatory cytokine interleukin (IL-10), IL-4, and interferon-gamma (IFNγ) expression genes were measured and compared in mastitis-free and mastitis-affected animals. Results: Based on the PCR, 70 (46.7%) samples showed positive results. The expression of the IL-10 gene was significantly higher (p < 0.001) in mastitis-affected cows than noninfected animals. Compared to cows diagnosed with clinical mastitis, the IL-4 and IFNγ genes were expressed more strongly in healthy cows (p < 0.0001). Conclusion: Mastitis has been linked to both Gram-positive and Gram-negative bacteria. These genes are strong predictors of mastitis in the states analyzed, as evidenced by the differential expression in mastitis and healthy conditions of the IL-4, IL-10, and IFNγ genes. Objective: The study was undertaken to assess the changes in viable bacterial counts and physicochemical parameters of water used during the live transportation of Pangasius catfish (Pangasianodon hypophthalmus). The correlations between the changing patterns of these parameters were also established. Materials and Methods: Water samples were collected every 2 h interval, plated onto agar plates for assesing viable bacterial counts. Physicochemical parameters, namely, water temperature, dissolved oxygen (DO), pH, and ammonia (NH3) concentration in the water were measured with a glass thermometer, DO test kit, pH test kit, and total NH3 measurement kit, respectively. Results: The viable bacterial counts increased significantly from 0 to 2 h in all the studied chan¬nels and remained almost similar up to the end of the supply channels. The water temperature was almost stable regardless of the supply channels and transportation period at around 30°C. The DO concentration and pH level decreased, and NH3 concentrations increased gradually in all the supply channels. The viable bacterial counts were inversely correlated to the DO levels and directly associated with the NH3 concentrations of the water used during the live transportation of Pangasius catfish. Conclusions: Gradual increase in viable bacterial counts and fluctuation in some vital physicochemical parameters with the duration of transportation indicated an unfavorable environment for the survival of Pangasius catfish. Objective: This study aimed to evaluate the combination of hairy eggplant (Solanum ferox) and bitter ginger (Zingiber zerumbet) on the production performance and hematological parameters of whiteleg shrimp (Litopenaeus vannamei). Materials and Methods: Four treatments were formulated in the test feed, where P1 (control + commercial vitamin C); P2 (120 ml/l combination of S. ferox and Z. zerumbet); P3 (100 ml/l); and P4 (80 ml/l) 6,000 post-larvae shrimp with an average initial weight of 0.2 gm were randomly stocked in four groups, with three replications per treatment, and 500 were stocked in each pond with a total of 12 pounds. Results: Based on the results, there were significant differences in production performance (survival, absolute weight growth, specific growth rate, and feed conversion ratio). Biologically, the best performance was found in the P3 treatment (100 ml/l). In this treatment, the total number of hemocytes and the number of hyaline hemocytes were much higher, and this was not the case in the control treatment (P1), where the number of semi-granular and granular cells was significantly higher than the treatment group (p < 0.05). Conclusions: This study confirmed that supplementation of 100 ml/l of S. ferox and Z. zerumbet could improve the production performance and hemato-immunological parameters of whiteleg shrimp, with functional potential to be developed in phytobiotic-based commercial diets for shrimp. Objectives: The objective of this study was to evaluate the persistence of oxytetracycline (OTC) residue in common fish species (Catla, Rui, Mrigal, and Pangas) available in local fish markets and the corresponding transport water of the Mymensingh region. Materials and Methods: Live fish and corresponding transport water samples were analyzed by thin layer chromatography (TLC) and high-performance liquid chromatography for qualitative and quantitative detection of OTC residue, respectively. A total of 240 fish samples and 60 water samples were randomly collected from three local fish markets during the summer and winter seasons. Results: OTC residues were detected in 18 samples (13 fish and 5 water samples) in the summer and 8 samples (only fish samples) in the winter. The overall percentage of antibiotic residue positive in fish samples was 5.42%, and in water samples, it was 8.33%. In fish, OTC concentrations of TLC-positive samples ranged from 34.7 to 56.85 parts per billion (ppb) in Catla, 23.45–35.37 in Rui, 11.02–26.80 in Mrigal, and 10.80–77.55 in Pangas during summer. The concentrations were 18.5 ppb in Catla, 15–16.09 in Rui, 10–14.63 in Mrigal, and 21.02–40.11 in Pangas during the win¬ter season. On the other hand, the range of OTC concentrations of TLC-positive samples for water was 12.9–59.18 ppb during summer and below the detection level during winter. The highest prevalence of antibiotic residues among fish samples was found in Pangas (16.67%). The highest percentage of samples (15% in the fish sample and 30% in the water sample) found to be positive were collected from Mechua Bazar. The comparison between the summer and winter seasons showed that the percentage of positive antibiotic residue in the summer season (10.38% for fish and 16.67% for water) is higher than that of the winter season (6.67% for fish samples only). This variation indicates that fish transporters use more antibiotics during the summer than in the winter season. The difference between the means of fish species and water samples was not statistically significant (p > 0.05). In addition, no samples exceeded the maximum residue limits (MRL) of OTC (100 ppb) in fish set by the European Commission. Conclusion: Although the concentrations of antibiotic residues in fish edible tissues are below MRL values, the presence of antibiotic residues in transported water may lead to the development of antimicrobial resistance bacteria that are detrimental to humans, animals, and aquatic animals. Objective: Staphylococcus aureus (S. aureus) has evolved as one of the most significant bacteria causing food poisoning outbreaks worldwide. This study was carried out to investigate the prevalence, antibiotic sensitivity, virulence, and enterotoxin production of S. aureus in raw milk of cow from small-scale production units and house-raised animals in Damietta governorate, Egypt. Material and Methods: The samples were examined bacteriologically, and antimicrobial sensitivity testing was carried out. Moreover, isolates were characterized by the molecular detection of antimicrobial resistance, virulence, and enterotoxin genes. Results: Out of 300 milk samples examined, S. aureus was isolated from 50 samples (16.7%). Antibiotic sensitivity testing revealed that isolates were resistant to β-lactams (32%), tetracycline (16%), and norfloxacin (16%); however, they showed considerable sensitivity to ceftaroline and amikacin (72%). Multidrug-resistance (MDR) has been observed in eight isolates (16%), with a MDR index (0.5) in all of them. Of the total S. aureus isolates obtained, methicillin-resistant S. aureus (MRSA) has been confirmed molecularly in 16/50 (32%) and was found to carry mecA and coa genes, while virulence genes; hlg (11/16, 68.75%) and tsst (6/16, 37.5%) were amplified at a lower percentage, and they showed a significant moderate negative correlation (r = −0.59, p-value > 0.05). Antibiotic resistance genes have been detected in resistant isolates relevant to their phenotypic resistance: blaZ (100%), tetK (50%), and norA (50%). Fifty percent of MRSA isolates carried the seb enterotoxin gene. Conclusion: High detection rate of MRSA and MDR isolates from milk necessitates the prompt implementation of efficient antimicrobial stewardship guidelines, especially at neglected small-scale production units.
