Characterization and assessment of naturally mutant non-pathogenic O27 strain Escherichia coli and their potential use as poultry probiotics Full text

Objective: The purpose of the current study was to evaluate the molecular characteristics of naturally mutant non-pathogenic O27 strain of Escherichia coli and its efficacy as probiotic in broilers and determine the best age at which it can be administered. Materials and methods: A total of 24 virulence genes using 24 sets of primers were detected using the polymerase chain reaction technique. For probiotics experiments, 60 chicks (day 1 old) were divided into three groups, 20 per group, and reared for 4 weeks. The first group was considered as a negative control. The second group was treated orally with O27 strain at first day of life for three successive days and repeated at day 21. The third group was administered orally with O27 strain at day 10 old, and repeated at day 21 old. Results: The data revealed that type 1 fimbrial adhesion, salmochelin siderophore receptor, and sigma factor-binding protein were detected in O27 strain, but temperature-sensitive hemagglutin, hemolysin secretion gene, pyelonephritis-associated Pili gene, polysaccharide capsule synthesis gene, Shiga-toxin1 gene, Shiga-toxin2 gene, Brain microvascular endothelial cell invasion, E. coli attaching and effacing gene, heat-stable enterotoxin, heat-labile enterotoxin, east 1 toxin, colicin V, verotoxin type 2, necrotizing cytotoxic factor type 1, colonization factor antigen I, colonization factor antigen III, coli surface 2, coli surface 4, serine protease pic autransporter, vacuolating autotransporter toxin, and serine protease EspP precursor were not detected in O27 strain. Group 2 performance parameters were significantly better (p < 0.01) than groups 3 and 1. Hematological and biochemical parameters did not be influenced (p > 0.05) by the administration of O27 strain. Antibody titers of infectious bursal disease virus and Newcastle disease virus in groups 2 and 3 were improved as compared to group 1. Group 2 had significantly higher titers than group 3. Histopathologically, all groups showed normal histopathological pictures. However, jejunum in groups 2 and 3 showed more tall, intact, and densely packed microvilli and more crypt depth than the control group. Conclusion: The O27 strain of E. coli is non-pathogenic bacteria. Its effects on growth performances and enhancement of immunity in broilers match with the same impact of probiotics, and these candidates will fit to be a good probiotic in the future. The results revealed that the effects of O27 strain at the day 1 old of life for three successive days and repeated at day 21 old are better for improving the performance and immunity of the birds. Objective: This study aimed at evaluating the supporting and non-supporting loop colostomy techniques in dogs. Materials and Methods: In this study, 12 adult healthy mongrel dogs were used to carry out a short-term (21 days) temporary flank loop colostomy. The dogs were divided into two groups (n = 6). In the first group (Con group), conventional stoma fixation was operated without loop support. In the second group [subcutaneous silicone drain (SSD) group], an SSD was used as a supporting bridge. The stoma was fixed by a modified, interrupted seromuscular vertical mattress suture pattern. Both peristomal and stomal complications were recorded. Results: Results showed that a total of seven dogs (58%) suffered complications. There was a sig¬nificant tendency toward less peristomal (p = 0.012) and stomal complications (p = 0.001) in the SSD group. The intensity of pain was significantly (p < 0.05) different between the two groups in the first week post surgery. Comfort with stoma care was significantly (p = 0.015) better in the SSD group, experiencing the best results. Conclusion: It is concluded that an appropriate stoma location, using a soft supporting bridge, stoma fixation using modified interrupted neuromuscular vertical mattress sutures, and proper appliance fixation by adhesive tapes and a tight T-shirt may overcome the complications of short-term loop colostomy in dogs and introduce a satisfaction for stoma care. Objective: A clinico-epidemiological study was conducted at two veterinary hospitals in Dhaka to evaluate the prevalence of parvoviral enteritis (PVE) in dogs and feline panleukopenia (FPL) in cats, to detect factors associated with them, and to identify their common clinical signs. Materials and Methods: Clinico-epidemiological data were collected for a total of 88 dogs and 129 cats which were presented to the hospitals, entered into MS excel 2010, and then transferred to STATA-14 software for conducting descriptive and univariable statistical analyses (Fishers exact test). The maps showing the spatial distribution of PVE and FPL were produced by using QGIS software version 2.18.13. Results: PVE in dogs and FPL in cats had the highest prevalence compared to other diseases (34.1%; 95% confidence interval: 24.344.9; N = 88 and 20.2%; 95% confidence interval: 13.6 28.1; N = 129, respectively). Young age (43.8%), poor body condition (58.4%), and exotic breeds (44.2%) were significantly associated with the occurrence of PVE (p ≤ 0.05). Poor body condition (27.8%) and non-vaccination status (26.7%) were associated with FPL (p ≤ 0.05). Both PVE and FPL showed clinical signs of being off-feed, frequent vomiting, weakness, and moderate dehydration. Conclusion: The high prevalence of PVE and FPL indicates that they are common in dogs and cats in Dhaka, Bangladesh. The factors related to PVE are young age, exotic breed, and poor body condition score (BCS), and FPL are poor BCS and non-vaccination. Specific measures such as care during younger age, good nutrition, and routine vaccination are needed to prevent and control PVE and FPL in Dhakas dog and cat population. Objective: This study was carried out to study the influence of gender, neuter status, and training method on police dog narcotics olfaction performance, behavior, and welfare. Materials and Methods: A total of 120 German Shepherds aged 13 years were used for this study. The dogs were separated into two experiments. The first experiment (32 dogs and 28 bitches) was used to study the influence of gender on olfaction and smell to narcotics in police dog perfor¬mance and training methods with behavioral problems and welfare. The second experiment (30 dogs and 30 bitches) was used to study the influence of sexual status (entire or neutered) on the smelling of narcotics in police dog performance by comparing with intact dogs and bitches. Results: We found that there were significant differences in sex in training to detect narcotics. Male German Shepherds were found to be significantly more trainable than females. Neutering causes a difference in trainability in male and female dogs. Gonadectomy had adverse effects on training. The intact male and female German Shepherds were found to be significantly more train¬able than the neutered ones, and the reward-based method was found to be significantly more trainable than punishment. Dog training methods incorporated by punishment result in pain, suf¬fering, emotional instability, symptoms of depression, aggression, unwanted barking, growling at other people, not under control all time, less trainability, increased problematic behavior, and decreased dog welfare. Conclusion: Reward-based method is associated with lower lousy behavior and dogs with good behavior, such as, attachment attention behavior, dogs under the control of handler all times, higher trainability, less problematic behavior, and increased dog welfare. Objective: Mastitis is considered as an economically important disease of dairy buffaloes in Asia. This study examined the mastitis milk and nasal swab samples for the detection and genotyping of methicillin-resistant Staphylococcus aureus (MRSA) in water buffaloes. Materials and Methods: Staphylococcus aureus was identified based on biochemical tests and Polymerase Chain Reaction (PCR) detection of nuc gene, whereas MRSA on mecA gene. The disc diffusion test was used to determine the antibiotic resistance and staphylococcal cassette chro¬mosome mec (SCCmec), spa, and multilocus sequence typing for the genotyping of isolates. Results: Staphylococcus aureus was detected on 39/93 milk (41.94%) and 27/384 nasal swab (7.03%) samples. However, only nine isolates (23.08%) harbored the mecA gene from milk sam¬ples and three isolates (11.11%) from the nasal carriage. All MRSA isolates exhibited resistance to cefoxitin and penicillin, whereas 50% were found resistant to clindamycin. All these isolates were found susceptible to sulfa-trimethoprim and chloramphenicol, whereas the majority of the isolates were susceptible to gentamicin, ciprofloxacin, tetracycline, and rifampicin. The SCCmec types of the MRSA isolates were type IVc (50.00%), type II (8.33%), type I (8.33%), and non-typea¬ble (33.33%). The spa types and sequence type (ST) identified were t019 (ST30), t701 (ST1649), t311 (ST5), t657 (ST1148), t015 (ST508), t1939 (ST12), t800 (ST9), t091 (ST2454), t138 (ST5991), and t1642 (ST5992). Conclusion: Milk and nasal swab samples from dairy water buffaloes were found positive for MRSA. The MRSA isolates were still susceptible to most antibiotics tested. Moreover, the gen¬otypes of some MRSA isolates were found similar to some human MRSA strains, suggesting a possible human to animal transmission. [J Adv Vet Anim Res 2020; 7(3.000): 397-406]

Objective: This study aims to investigate the callipyge gene (CLPG) polymorphism in sheep of Edilbay, Volgograd, and Kalmyk breeds. Materials and Methods: The analysis was performed by the polymerase chain reactionrestric¬tion fragment length polymorphisms method. The objects of the study were Edilbay fat-tailed sheep (n = 500) at the breeding plant Volgograd-Edilbay (Volgograd region), Volgograd fine-wool sheep (n = 500) at the breeding plant Romashkovskiy (Volgograd region), and Kalmyk fat-tailed sheep (n = 500) at the breeding plant Kirovsky (the Republic of Kalmykia, Yashkul rayon). To conduct the research, tissue samples of 1 cm² from sheep of Kalmyk and Edilbay breeds were taken from the auricle. Results: The allelic CLPG gene variants have been determined and genotypes of representative sampling of the three breeds of livestock grown in the steppe zone of Russia. The presented results of the CLPG gene polymorphism in these sheep breeds grown in Russia were obtained for the first time. The research study has revealed that in terms of the CLPG gene, the Edilbay, Volgograd, and Kalmyk sheep breeds have only a homozygous form. Objective: The study obtained data on the potential of local feed ingredients, both in quantity, quality, and continuity and to observe the absorption power and ability of livestock to use feed designed in vitro. Materials and Methods: The method uses a survey method of potential sources of animal feed and calculates the carrying capacity of ruminants and nutritional analysis of feed ingredients with proximate analysis. Feed formulations were made based on proximate analysis results of four formulas (P1, P2, P3, and P4) and tested in vitro. Results: First, Nganjuk district has the highest of the local food potential with the production of agricultural waste and agricultural industries, reaching 802,341.94 tons/year. Second, the most top carrying capacity analysis reached in Tulungagung district, which reached 62,534 ST/year or 43% of the total population of ruminants. Third, the results of the study of the quality of local feed ingredients indicate that each type of feed material has the right and proper nutrition given to ruminants. Fourth, the in vitro testing included showing P1 feed that had a very significant effect (p < 0.01) on dry matter digestibility and digestibility of organic matter of cows, respectively, 74.69% and 73.39%. Conclusion: The in vitro technique of making animal feed can be developed in the areas that have the potential to produce agricultural waste and agricultural industries to increase the carrying capacity of livestock. Objective: The objective of this work was to study the normal musculotendinous structures of the ostrich foot. Materials and methods: Ten African apparent healthy adult female ostriches (Struthio camelus) were slaughtered, and the pelvic limbs were separated from the ankle joint. The different biomed¬ical scanning techniques including radiography, computed tomography (CT), magnetic resonance imaging, and ultrasonography were achieved. Then, the ostrich feet were freshly dissected. Results: The radiographs and CT images showed the bony components of the ostrich foot that revealed the presence of long tarsometatarsus and phalanges of the only developed third and fourth toes. The third digit was the longest and possessed four phalanges, whereas the shorter fourth toe contained five phalanges. The ostrich foot consisted mainly of tendons in addition to several small associated muscles. The extensor structures were the extensor digitorum longus tendon, Mm. extensor proprius digiti III, and extensor brevis digiti III and IV. On the other hand, the flexor structures were the flexor digitorum longus, flexor hallucis longus, flexor perforans et perforatus digiti III, and flexor perforatus digiti III and IV tendons. Furthermore, fibularis lon-gus tendon and two muscles (Mm. abductor digiti IV and lumbricalis) were related to the flexor tendons. Conclusion: The combination between the dissecting anatomy and the different biomedical scan¬ning techniques was of value in describing the normal anatomical course of the musculotendi¬nous structures of the ostrich foot, which aids in the diagnosis of any clinical abnormalities in these structures. Objective: This study was carried out to determine the frequency and prevalence of clinical conditions and their treatment, especially antibiotics in dogs and cats. Materials and Methods: A period of 12-month retrospective study was conducted at the Teaching Veterinary Hospital, Chattogram Veterinary and Animal Sciences University, Chittagong, Bangladesh from July 2018 to June 2019. A total of 849 cases, including 488(57.5%) and 361 (42.5%) dogs and cats respectively, were in account to espy the clinical conditions. Season, age, sex, and breed were the parameters to analyze the prevalence of those clinical conditions. Results: From the study, it was found that the endoparasitic infestation was highly frequent in both dog and cat (55% in dogs and 59% in cats). The endoparasitic infestation was highly prevalent in cats (91.53%) significantly (p = 0.003), which were ≤1 year of age. On the other hand, the ectoparasitic infestation was found prevalent significantly (p = 0.06) in the winter than any season and dewormed dogs (p = 0.03). Prevalence of canine parvovirus infection in dogs and wound in cats were substantially higher (p < 0.001 and p=0.05 respectively) in the winter whereas the prevalence of myiasis in dogs was prominent in the rainy season significantly (p = 0.01). The mostly used antibiotic was ceftriaxone (9.5% in dogs and 4% in cats). Conclusion: Different endoparasitic, ectoparasitic, and infectious diseases found prone to infect pet animals, mainly dogs, and cats. Objective: Sheep farming is an important source of livelihood in the southern Khyber Pakhtun Khwa province of Pakistan where they are reared under extensive system (ES). This system has limitations of improper feeding, rearing, and management which negatively affect their productivity. The pres¬ent study compares different rearing systems and their stresses on fertility, behavior, and growth rate potential of two sheep breeds of Pakistan. Materials and Methods: Eighty ewes were selected; forty each from Damani and Balkhi breeds reared in semi-intensive system (SIS) and ES at Paharpur district, Dera Ismail Khan. Blood samples were collected in triplicate on days 0, 14, and 28 of May, and the molecular and behavioral stress, growth, and fertility rates were recorded. Results: The highest growth and fertility rate were observed in both the breeds in SIS than in ES (p < 0.01). The values of stress indicators (cortisol, heat shock protein (HSP-70)) and behavioral stress parameters were found lower in Damani breed as compared to Balkhi breed in both rearing systems (p < 0.01). All the molecular stress and behavioral stress parameters were negatively associated with metabolic hormones (T3 and T4) (p < 0.01). A positive correlation was observed among all the molecular stress and behavioral stress parameters (p < 0.01). Similarly, T3 and T4 were positively interlinked to one another (p < 0.01). Conclusion: It was concluded that both Damani and Balkhi sheep showed lower stress levels and better fertility and growth parameter in SIS than ES system of rearing. Local Damani breed exhibited well tolerant ability and genetic adaptation to the local environment as compared to Balkhi breed. [J Adv Vet Anim Res 2020; 7(1.000): 170-176]
