Objective: The study investigated the effect of different stocking density (SD) rates on the welfare, growth, and hemato-biochemical parameters in broiler chickens. Materials and Methods: 106 broiler chicks of 10 days old were used and assigned into four groups: A, B, C, and D. The chicks of group A were reared in floor space containing one bird per square foot area (SD1.0). The chicks of groups B, C, and D were reared at 1.5, 2.0, and 2.5 birds per square foot area (SD1.5, SD2.0, and SD2.5). Welfare, body weight, and hemato-biochemical parameters were assessed and monitored by physical observation and laboratory methods. Results: The birds reared at SD2.0, and SD2.5 rates showed increased panting breathing. Wet feces adhered below the vent. There were a significant number of birds showing dirtiness of body and feathers. Birds reared in SD2.5 were familiar with moist litters and high ammonia smell. Foot-pad dermatitis, scratches, and blister formation were detected in the leg. The study revealed that the higher SD negatively correlated to the welfare behavior indicators. Live body weight was significantly (p < 0.05) decreased in birds reared at higher SD rates. Birds housed in SD1.0 and SD1.5 are optimum for body weight and improved feed conversion ratio. The hemato-biochemical parameters of birds reared at various SD rates did not differ. The total leucocyte count increased significantly, while total serum proteins decreased gradually as SD rates increased. Objective: In developing countries, such as Bangladesh, the live bird market (LBM) is a vital location for the trading of live poultry. The study was carried out in nine LBMs located around Bangladesh to ascertain the present regulations and procedures governing their operation. Additionally, the responsibilities and levels of engagement of the stakeholders were determined. Materials and Methods: The data were gathered through the use of a semi-structured interview guide. Thematic analysis was used to code the interview transcripts iteratively. Results: The findings indicated that the government was directly and indirectly involved in the leasing process of the markets. A market in this country is divided into numerous sectors, including LBM, fish market, vegetable market, and grocery stores. A markets hygienic condition is highly dependent on market authoritys decisions. In some markets, market officials conducted routine sanitary inspections. Veterinarians played a little role in the inspection procedure. Conclusion: There is no adequate, functional monitoring system to ensure that LBMs adhere to cleanliness and adequate and functional biosecurity. Biosecurity enhancements, effective cleaning programs, and regular monitoring by relevant authorities are critical for LBMs in Bangladesh. [J Adv Vet Anim Res 2021; 8(4.000): 671-678]

Objective: The purpose of this work is to develop and validate an appropriate solvent solution and quantitative thin layer chromatography (TLC) method for determining the aflatoxins content of chicken feeds and dietary grains. Materials and Methods: To obtain the optimal mobile phase, samples were extracted with methanol/water (3:1) + 5% sodium chloride and partitioned using several solvent systems using preparative TLC. Camag TLC scanner 3 was used to scan the TLC plates at 366 nm and quantify them using JustTLC software. The method was tested for linearity, specificity, accuracy, precision, sensitivity, and robustness in accordance with ICH recommendations, and then utilized to screen 132 Nigerian poultry/food samples for total aflatoxins (TAFs). Results: The best separation of aflatoxins was achieved using acetonitrile and dichloromethane (3:17) mobile phase over an average run time of 45 min, resulting in linear calibration curves (R2 > 0.99) in the concentration range limit of quantitation (LoQ) to 50 ng/spot with a limit of detection of [J Adv Vet Anim Res 2021; 8(4.000): 656-670]

Objective: The case study describes the cause of an increase in mortality rates among 35-day-old broilers that developed respiratory distress and bloody diarrhea on a farm in Malaysia. Materials and Methods: The organ samples were subjected to laboratory testing and postmortem inspection. Escherichia (E.) coli and Mycoplasma (M.) gallisepticum were detected using bacterial isolation and molecular diagnostics using polymerase chain reaction. Results: Chickens with the infection had widespread fibrin buildup in several organs and hemorrhages on the duodenal mucosa. Additional histology and laboratory analysis of organ samples revealed infection with M. gallisepticum, E. coli, and enteric Eimeria spp., all of which are consistent with complex chronic respiratory disease (CCRD) associated with coccidiosis. Tylosin tartrate 20% (w/w) (2.5 gm/l) was prescribed for 1 week along with a combination of the broad-spectrum bacteriostatic drug streptomycin (25 mg/kg) and coccidiostat (2 gm/5 l). Conclusion: CCRD and coccidiosis are both infectious diseases that can infect chicken flocks, resulting in production losses and carcass quality degradation. Early disease detection and proper treatment should be provided promptly, and tight farm biosecurity should be implemented to prevent chicken mortality on the farm, as was achieved successfully. Objectives: The experiment was conducted to determine the effect of dietary energy and protein level growth performances of selected indigenous Ethiopian sheep breeds. Materials and Methods: Fifty intact ram lambs, 25 from each breed with 12 months of age and a mean initial body weight (IBW) of 19.31 ± 1.7 kg, were employed for this experiment. Animals were distributed randomly into five dietary treatments, i.e., minimum Energy and Protein (mEmP), medium energy and protein (MEMP), medium Energy and high Protein (MEHP), high energy and medium protein (HEMP), and high Energy and high Protein (HEHP) diets in randomized complete block design with 2 * 5 factorial arrangements. The minimum, medium, and high energy diets were 2.388, 2.866, and 3.344 Mcal/kg dry matter (DM) with the corresponding 10%, 16%, and 20% crude protein (CP) diets, respectively. The diets were formulated in a total mixed ration from wheat bran (WB), maize grain, peanut cake, and pasture hay feed ingredients. Diet offer was at the rate of 3% of lambs live weight and revised biweekly as per the attained body weight changes. Digestibility trial was conducted for 7 days of actual fecal data collection, followed by 90 days of feeding trial. Results: The animals fed on the MEHP diet had a maximum DM and nutrient intakes (CP and organic matter) and the best final body weight (FBW), total gain, gain rate, average daily gains, and feed conversion efficiency (31.3, 12.9 kg, 41.2%, 143.3 gm, and 23.13, respectively), followed by HEMP, HEHP, MEMP, and mEmP diets. Digestibility of DM and nutrients linearly followed similar trends (p < 0.01). Hararghe sheep was heavier (p < 0.01) by 4.3 and 3.1 kg in its FBW and total gain and more efficient in nutrients utilization (22.57 vs. 18.18) as compared to Afar sheep (AS). Conclusion: It is concluded that MEHP and MEMP are superior and optimum diets for sheep breeds, and Hararghe sheep is carried out better than AS in most growth performance parameters. Objective: A phylogenetic study was carried out on the avian influenza virus (AIV) isolated from a disease outbreak in Sidenreng Rappang Regency, South Sulawesi, Indonesia, in 2018. Material and Methods: Oropharyngeal swabs and organ samples were obtained from ducks that showed clinical symptoms: torticollis, fascial edema, neurological disorders, the corneas appear cloudy, and death occurs less than 1 day after symptoms appear. In this study, isolate A/duck/ Sidenreng Rappang/07180110-11/2018 from duck was sequenced and characterized. Results: It was found that each gene segment of the virus has the highest nucleotide homology to the Indonesian highly pathogenic avian influenza (HPAI) H5N1 clade 2.3.2.1c. Multiple alignments of the sample Hemagglutinin (HA) gene with the avian influenza references virus showed that the pattern of amino acid arrangement in the cleavage site PQRERRRK-RGLF is the characteristic of the HPAI virus. In addition, the HA gene contained Q222 (glutamine) and G224 (glycine), signify¬ing a high affinity to avian receptor binding specificity (SA α2,3 Gal). Furthermore, there was no genetic reassortment of this virus based on the phylogenetic analysis of HA, NA, PB1, PB2, PA, NP, M, and NS genes. Objective: Chicken infectious anemia virus (CIAV) is an economically important emerging infec¬tion of poultry as it causes immunosuppression and reduces egg production. Although it is world¬wide distributed and first reported (single case) in Bangladesh in 2002, no epidemiological and serological investigations have been conducted. The current study aimed to conduct a serologi¬cal investigation on the prevalence of CIAV infection in broiler breeder and layer farms in some selected areas of Bangladesh. Materials and Methods: A total number of 460 sera samples were randomly collected from unvac¬cinated broiler breeder and layer flocks, of which 276 were from 11 broiler breeder farms and 184 from 12 layer farms. The sera samples were subjected to a commercially available enzyme-linked immunosorbent assay kit to observe antibodies induced by CIAV. Results: Results demonstrated that the overall prevalence of CIAV was 83.6% among a total of 460 samples. In broiler breeder birds, the prevalence was 89.9%, whereas it was 78.3% in layer birds. A higher number of female birds was found to be seropositive than male birds. However, chickens of all age groups were found to be susceptible to the virus. Conclusions: These results indicate the presence of CIAV in Bangladesh, which may be the sequel of naturally occurring either vertical or horizontal infection in all bird flocks tested without clinical symptoms of the disease. A further epidemiological investigation will be required, followed by molecular isolation and characterization of the virus for suitable vaccine candidate selection and/ or preparation. Objective: Present research aims to isolate, identify, and determine the virulence of the Streptococcus agalactiae (group B Streptococcus; GBS), isolated from popped eye disease affected Tilapia and Vietnamese Koi (V. Koi) fishes. Materials and Methods: A total of 330 fish samples were collected, of which Tilapia (n = 180) and V. Koi (n = 150), were collected from 35 affected ponds of four selected districts of Bangladesh. Isolation of the bacterium was done using different culture media (Nutrient broth, Plate count agar, Tryptic Soy Agar, and Blood agar), and identification by using various biochemical tests (con¬ventional and using API 20 Strep kit) and polymerase chain reaction (PCR) using primers against 16S rRNA gene of S. agalactiae. Antibiotic susceptibility of the bacteria was performed using seven different antibiotics disc (Tetracycline, Oxytetracycline, Chlortetracycline, Streptomycin, Ciprofloxacin, Gentamicin, and Neomycin). Virulence of the isolated S. agalactiae was determined by infecting healthy Tilapia and V. Koi fishes through experimental infection. Results: Isolated bacteria were found Gram-positive paired and chained cocci, β-hemolytic and non-motile. Findings of biochemical and serological tests indicate that the isolated bacterium belongs to Group B Streptococcus of Lancefield classification. PCR result also confirmed that the bacteria were S. agalactiae. The bacterial isolates possessed resistance property against all the seven antibiotics used in this study. The isolated GBS was found highly virulent and showed 80%90% mortality for Tilapia and V. Koi fishes in experimental infection within 16 days of post-infection. Conclusion: From the findings of this study, it may be concluded that isolated GBS from the Tilapia and V. Koi fishes were highly virulent and possessed multidrug-resistance properties. Objective: The study was carried out for molecular characterization and antibiotic resistance anal¬ysis of Escherichia coli isolated from different turkey farms in Dinajpur, Bangladesh. Materials and methods: A total of 45 samples comprising feces (n = 23) and cloacal swabs (n = 22) were collected randomly from turkeys. The samples were subjected to isolation and identification of E. coli by cultural and biochemical characteristics, followed by polymerase chain reaction and sequencing. An antibiogram of the isolated E. coli isolates was carried out by following the Kirby Bauer disk diffusion method. Results: Out of the 45 samples, 28 (62.21%) were positive for E. coli, of which 16 (35.55%) fecal samples were positive and 12 (26.66%) cloacal swabs were positive. The antibiotic sensitivity analysis revealed that all the E. coli isolates were 100% sensitive to levofloxacin, norfloxacin, neo¬mycin, gentamicin, and nitrofurantoin. On the other hand, all the isolates were 100% resistant to amoxicillin, azithromycin, erythromycin, tetracycline, bacitracin, cephalexin, nalidixic acid, van¬comycin, methicilin, piperacillin, pefloxacin, novobiocin, cefepime, trimethoprim, netilmicin, and aztreonam. Objectives: Probiotics are well documented for their health benefits by developing a balanced intestinal microbiota and boosting immunity. The present study was conducted to determine the effects of a probiotic preparation EnzimsporinTM (consisting of spore-forming bacteria Bacillus subtilis B-2998D, B-3057D, and Bacillus licheniformis B-2999D) on the biochemical, hematological, immunological parameters, intestinal microbiota, and growth dynamics of sheep and lambs. Materials and methods: Enzimsporin was fed to lambs and sheep at different doses to determine the bacterias probiotic effects. Sheep were divided into three groups (six each), which received 0, 1, and 3 gm of Enzimsporin/per head/day, respectively, and two groups of lambs (10 each), who received 0 gm and 1 gm of Enzimsporin/per head/day for 30 days in addition to their regular ration. On day 30, blood samples were collected, followed by the determination of biochemical, hematological, and natural resistance indicators. Fecal samples were examined to determine the intestinal microflora, and animals were weighed daily to determine their growth dynamics. Results: Supplementation of probiotics (EnzimsporinTM) improved the lambs body weight gain by 18.8%. Analysis of the clinical parameters showed improvements in the levels of total protein, globulins, and urea by 5.3%, 10.8%, and 6.2%, respectively, in the blood of probiotic-supplemented lambs. Similarly, an increment in the total protein, albumins, and globulins was observed in the sheep with EnzimsporinTM supplementation. The decrease in bilirubin and cholesterol levels in the blood and increased bactericidal and phagocytic index in the sheep and lambs with probiotic supplementation indicated a positive influence of EnzimsporinTM on the liver function and natural resistance. Furthermore, an increase in Lactobacillus and Bifidobacterium and a decrease in the Escherichia coli, Enterococcus, and Yeast in the fecal contents of experimental sheep and lambs indicated the potentiality of EnzimsporinTM on maintaining good gut health. Conclusion: Spore-forming bacteria B. subtilis B-2998D, B-3057D, and B. licheniformis B-2999D can be used in feeding sheep and lambs of 2 months of age to increase body weight gain, improve intestinal microbiota, strengthen the immune system, and maintain normal metabolic processes.
