Sperm-binding to the perivitelline membrane of chicken egg yolk as a functional test for sperm evaluation in dogs

During fertilization, spermatozoa interact with the zona pellucida (ZP) through the binding between the acrosome and proteins 2 and 3 (ZP2 and ZP3). The perivitelline membrane of chicken egg yolk is homologous to the mammalian ZP3, which allows the binding of sperm of several species. The aim of this study was to standardize and evaluate the efficiency of sperm-binding to the perivitelline membrane of chicken eggs as a functional method for canine semen evaluation. For this purpose, nine post-thaw sperm samples were used, which were divided into two aliquots: the first was kept in water bath at 37ºC (live sample) and the second was submitted to cold shock to induce cellular damage (dead sample). The two aliquots were mixed on five proportions, corresponding to 0%, 25%, 50%, 75%, and 100% of viable cells, and the binding test was performed by analyzing the number of spermatozoa bonded to the perivitelline membrane by means of computerized assessment of sperm motility (CASA) or conventional microscopy. Additionally, samples were submitted to sperm motility analysis, evaluation of plasmatic and acrosomal membrane integrity, and sperm mitochondrial activity. The sperm-binding test to the perivitelline membrane of chicken egg yolk was considered a feasible sperm analysis test for both fertilizing capacity and overall sperm attributes evaluation, mainly when the analysis is performed by a conventional microscope, which expands its practicality to the majority of canine reproduction laboratories.

The use of serum lactate has become increasingly common in Veterinary Medicine, since it is an excellent marker of tissue oxygenation, elevated in situations of tissue hypoxia, where anaerobic glycolysis is used as an energy source. Studies point out its use in dogs as a prognostic factor in several affections; however it is not commonly used in domestic felines due to the high stress threshold of the species, which may interfere with their values. The aim of the present study was to evaluate serum lactate values in domestic cats during physical restraint and after chemical restraint using the combination of 8mg/kg ketamine-S, 0.4 mg/kg midazolam, and 2 mg/kg Tramadol. To this end, 20 clinically healthy female cats without defined race, from six months to five years of age, were divided into two groups, GPR (n = 10) represented by felines that had only physical restraint and the GCR (n = 10) by felines that underwent physical restraint (TPR) and after 30 hours by chemical restraint (TCR). It was observed that in both groups (GPR and CPR) during physical restraint presented hyperlactatemia (GPR = 6.23 ± 0.9 mmol/L – GCR – Tpr = 6.66 ± 1.59 mmol/L) and mean values of 4.42 ± 1.28 mmol/L during chemical restraint (GCR – Tcr). It was concluded that physical restraint interferes with serum lactate values, with the reduction of such values with the use of chemical restraint in the feline species.

O plasma sanguíneo em pó (PSP), produto natural de indústria frigorífica, tem mostrado efeitos benéficos sobre o crescimento e desempenho de leitões desmamados precocemente. Atualmente, embora o circovírus suíno 2 (PCV2) tenha grande importância para a suinocultura, não há informações sobre o impacto do uso de PSP e a resposta imune ao PCV2 em infecções naturais. Este trabalho avaliou diferentes níveis de inclusão de PSP em dietas de leitões e as cargas virais de PCV2 correspondentes. Quatro níveis de inclusão de PSP foram testados em dois períodos consecutivos: 0, 2, 4 ou 6% durante o período 1 (14 aos 28 dias de idade) e 1, 2 ou 3% de PSP durante o período 2 (29 a 42 dias de idade). No período 3 (42 aos 56 dias de idade), todos os leitões foram alimentados com dieta isenta de PSP. Amostras de soro foram coletadas semanalmente e testadas para anticorpos anti-PCV2 e carga de DNA de PCV2. As concentrações de 6% e 3% de PSP fornecidas nas rações durante o período 1 e 2, respectivamente, influenciaram na carga viral de PCV2 de suínos naturalmente infectados.

Sie balu is an Acehnese dried meat preserved by the addition of salt, acid and dried. However, long processing and drying it under the sun can cause meat products contaminated by Salmonella. Irradiation can eliminate Salmonella and other pathogenic bacteria in food. This study aims to determine the presence of Salmonella in sie balu after irradiated with increasing doses and 2 to 4 months shelf life. Sie balu was made of 5 kg fresh beef, dried under the sun to dry, vacuum and irradiated at doses of 5, 7 and 9 kGy. Salmonella was detected using bacteriological and biochemical tests. Results showed sie balu contaminated by Salmonella paratyphi B and Salmonella cholerasuis. The irradiation and shelf life significantly affect (P0.05) the count of Salmonella in sie balu compared with unirradiated one. Irradiation doses of 5, 7 and 9 kGy do not affect the count of Salmonella in sie balu. Extending the shelf life up to 4 months can increas the count of Salmonella. This study concluded that irradiated sie balu at doses of 5, 7 and 9 kGy can be stored for up to two months

A study has been conducted to investigate the effect of two local herbs in Indonesia; Areca catechu nut and Sauropus androgynous leaf, combined with mineral block to treat gastro intestinal nematode in goats. A.catechu is well known to have anthelmintic property due to its alkaloid and tannin content, and S.androgynus also contains tannin, alkaloid and steroid that will enhance the anthelmintic property of those herbals. As many as 80 goats had been divided into 4 groups; 20 were treated with albendazole (Group A), 20 were given A.catechu and S.androgynus (Group B), 20 were given combination of A.catechu, S.androgynus, and mineral block (Group C), and 20 as negative control (Group D). The result revealed that herbal treatment showed significantly effective toward nematodes in gastro intestinal, demonstrated by declining egg worm of Group B and C. Body weight improvement and overall performance in Group C was higher compared to other groups. However, goats treated with albendazole showed slightly better results in the decreasing amount of egg worms found after treatment. Nevertheless, A.catechu and S.androgynus are potential anthelmintic herbal; combination with mineral block will enhance the anthelmintic activity. Therefore they are good options for treating helminthiasis in livestock.

One of the Acehnese traditional food preserved by the addition of salt, acid and dried was sie balu. The preservation method may lead to changes in the structure and nutritional content. Sie balu with the addition of vinegar, garlic and ginger can reduce the value of water activity (aW), pH and bacterial counts. This condition affects the physical properties and sensory of sie balu. Irradiation can control pathogenic microorganisms in meat without affecting the physical condition. Low-dose irradiation is able to maintain the desired sensory attributes nutritious food products. This study aims to determine the characteristics of the sensory and physic sie baluthat irradiated with increasing doses and the shelf life of 3-6 weeks. Sie balu was made of 10 kg fresh beef, dried in the sun to dry, vacuum and irradiated at doses of 5, 7 and 9 kGy. Sensory test conducted by taste sie balu by trained panelists and fill out the hedonicform. The physical properties based on pH and aW. The microbe total calculated used Total Plate Count.The results showed that the irradiation dose significantly (P 0.05) affected on microbe total, aroma, color, texture and taste of sie balu. Storage time does not affect the sensory properties. Irradiation dose and storage time significantly (P 0.05) on pH and aWsiebalu. The study concluded that irradiation can improve the physical properties, sensory and microorganisme reductionof sie balu.
