O presente estudo teve como objetivo traçar o perfil de gonadotrofinas em 12 novilhas Nelore, a fim de testar a hipótese de que há declínio nas concentrações de FSH e elevação transitória nos níveis de LH durante a seleção folicular. A partir da ovulação (D0) foram colhidas amostras de sangue da veia jugular a cada 12 horas até o D5, para a dosagem de FSH e LH plasmáticos. Empregou-se o método de radioimunoensaio de duplo anticorpo. A sensibilidade do ensaio de LH foi de 0,02ng/ml e a de FSH de 0,005ng/ml. Os coeficientes de variação intra-ensaio foram 13,6% e 18,8%, respectivamente. Os dados (média±EPM) foram normalizados para o momento da divergência folicular e, posteriormente, analisados por ANOVA e por regressão linear, cúbica e quadrática. Também foi empregado o Teste t para comparação entre o ponto mais alto e o mais baixo da curva. Não se verificou efeito de tempo sobre as concentrações plasmáticas de FSH e de LH quando se utilizou análise de variância e de regressão. Entretanto, através do Teste t pontual, o FSH atingiu as menores concentrações plasmáticas 36 (0,40±0,05ng/ml) e 60 horas (0,42±0,04ng/ml) após a divergência, comparativamente às 36 horas anteriores ao desvio, quando as concentrações foram máximas (0,63±0,08ng/ml). Conclui-se, portanto, que há declínio nas concentrações plasmáticas de FSH, contudo, não foi comprovada elevação transitória nas concentrações de LH próximo ao momento do desvio folicular em fêmeas Nelore. | Present study aimed to evaluate gonadotropins profiles in 12 Nelore heifers, in order to test the hypothesis that FSH concentrations decrease and LH presents a transient increase during follicle selection. Blood samples from jugular vein were harvested twice daily starting at the time of ovulation (D0) until D5. Plasma samples were assayed for FSH and LH by double antibody radioimmunoassay method. LH and FSH assay sensitivity was 0,02ng/ml and 0,005ng/ml, respectively. The intraassay coefficient of variation was 13,6% and 18,8%, respectively. Data (mean±SEM) were normalized to follicle deviation and analyzed by ANOVA and by linear, cubic, and quadratic regressions. Comparisons between higher and lower FSH values were also performed by T-test. There was no effect of time in plasmatic FSH and LH circulating levels when variance analysis or regression analysis were performed. However, by T-test, FSH concentrations reached the lowest plasmatic levels 36 (0,40±0,05ng/ml) and 60 hours (0,42±0,04ng/ml) after follicular deviation, comparatively to 36 hours before deviation, when the concentrations were maximal (0,63±0,08ng/ml). In conclusion, there is a FSH decrease, although a transient LH elevation has not been confirmed encompassing follicle deviation in Nelore females.

O objetivo do presente trabalho foi estudar as características em comum e as diferenças observadas na intoxicação induzida por 1 mg/kg de amitraz, IV, em cães e gatos. Os principais sinais clínicos observados em comum foram sedação, hipotermia, bradicardia, bradiarritmias, hipotensão, bradipnéia, midríase e hiperglicemia transitória, porém a intensidade destes sinais foi diferente entre as espécies. A hipotermia foi mais acentuada em gatos. Os cães foram mais sensíveis às alterações cardiorespiratórias apresentando diminuição mais significativa na freqüência cardíaca e respiratória, além de ocorrência de maior número de bradiarritmias. Os gatos apresentaram midríase mais prolongada do que os cães. Observou-se hiperglicemia e hipoinsulinemia transitórias e diminuição transitória dos níveis plasmáticos de cortisol em ambas espécies, porém os gatos apresentaram um pico de hiperglicemia maior e mais precoce do que os cães, e com relação aos níveis plasmáticos de cortisol, os cães apresentaram uma diminuição mais acentuada do que os gatos. O tempo médio de retorno da sedação foi mais prolongado em gatos. Estes resultados mostraram que a intoxicação por amitraz entre cães e gatos é muito similar, porém os gatos demonstraram maior sensibilidade à indução de hipotermia e hiperglicemia, além de midríase e um tempo médio de retorno da sedação mais prolongado, enquanto os cães apresentaram diminuição mais acentuada nos parâmetros cardiorespiratórios e dos níveis plasmáticos de cortisol do que os gatos. | The objective of the present work was to study the characteristics in common and the differences observed in the intoxication by amitraz 1 mg/kg, IV, in dogs and cats. The main clinical signs observed in common were sedation, hypothermia, bradycardia, bradyarrhytmia, hypotension, bradypnea, mydriasis and transitory hyperglycemia, however the intensity of these signs was different among the species. The hypothermia was more accentuated in cats than in dogs. The dogs were more sensitive to the cardiorespiratory alterations presenting more significant decrease in the heart and breathing frequency, besides occurrence of larger bradyarrhytmia number. The cats presented more prolong mydriasis than the dogs. It was observed hyperglycemia and transitory hypoinsulinemia and transitory decrease of the plasmatic levels of cortisol, however the cats presented a hyperglycemia pick more accentuated and more precocious than the dogs, and regarding the plasmatic levels of cortisol, the dogs presented a decrease more accentuated than the cats. In the medium time of return of the sedation smaller in cat. These results demonstrated that the amitraz intoxication between dogs and cats is very similar, however the cats showed more sensibility to the hypothermia and hyperglycemia induction, besides mydriasis and the medium time of return of sedation more prolong, while the dogs presented a decrease more accentuated in the cardiorespiratory parameters and of the plasmatic levels of cortisol than the cats.

Foram utilizados 20 gatos sem raça definida, com massa corpórea entre 2 e 4 quilogramas, hígidos, anestesiados com tiletamina e zolazepam (6,0 mg/kg). A artéria femoral foi canulada de forma asséptica e um manômetro acoplado a uma coluna de ar foi conectada ao cateter, a PAM foi aferida antes e a cada 15 minutos até o término do exame ultra-sonográfico. A fluxometria da artéria oftálmica externa foi obtida através do equipamento de ultra-sonografia dotado de eco-doppler, com transdutor setorial de 6,0 MHz, aplicado diretamente sobre a córnea. Cada olho teve o fluxo de sua artéria aferido pelo programa do próprio aparelho, sendo tomadas três amostras. O índice de resistência vascular foi calculado com base nos valores do fluxo sistólico e diastólico médio de cada vaso. Obteve-se um valor médio para a PAM de 144,9 ± 26,68 mmHg e uma velocidade de fluxo de 41,3 ± 14,28 cm/s para a artéria oftálmica externa no olho direito em sístole, 23,95 ± 11,46 cm/s em diástole e 42,75 ± 12,64 cm/s no olho esquerdo em sístole, 25,45 ± 9,61 cm/s em diástole, não existindo diferença significativa entre os olhos. O índice de resistência vascular calculado foi de 0,4175 para a artéria oftálmica externa no olho direito e 0,4015 para a do olho esquerdo, sem diferença significativa entre os olhos. Os resultados apontam para um mecanismo intrínseco de controle do fluxo, controlado por fatores locais na artéria oftálmica externa. Os valores do índice de resistência denotam uma artéria de baixo fluxo. Como conclusão, têm-se os valores de referência acima citados para a PAM e para o fluxo sangüíneo na artéria oftálmica externa em gatos sem raça definida anestesiados com tiletamina e zolazepam, e a informação de que não existe correlação entre esses valores. | There were used 20 mixed breed cats, both male and female, with body mass between 2 and 4 kilograms, that received anesthesia with tiletamine and zolazepam (6,0 mg/kg). The femoral artery was assepticale cannulated and a manometer connected to an air column was connected to the catheter. The MAP was surveyed before and every 15 minutes until the end of the ultrasound examination. The flow of the external ophthalmic artery was measured through the ultrasond equipment, with an echo-doppler and a sectorial transducer of 6.0 MHz, directly applied to the cornea. Each eye had the artery flow surveyed by the equipment's own device, 3 samples being taken. The vascular resistance index was calculated based on the mean values of systolic and diastolic flow of each vasses. An average value for the MAP of 144.9 ± 26.68 mmHg and a flow of 14.28 ± 41.3 cm/sec for the external ophthalmic artery in the right eye in systole, 23.95 ± 11.46 cm/sec in diastole and 42.75 ± 12.64 cm/sec in the left eye in systole, 25.45 ± 9.61 cm/sec in diastole, were verified, there being no significant difference between the eyes. The calculated index of vascular resistance was of 0.4175 ± 0.0774 for the external ophthalmic artery in the right eye and 0,4015 ± 0.0719 for the left eye, without any significant difference between the eyes. The results point to an intrinsic mechanism of flow control, controlled by local factors in the external ophthalmic artery. The values of the resistance index denote a low flow artery. Concluding, this study revealed the reference values, above cited, for the MAP and the blood flow in the external ophthalmic artery in mixed breed cats, anesthetized with tiletamine and zolazepam, and the information that there is no correlation between these values.

Os efeitos embriotóxicos da exposição diária pré-natal a 0,0, 0,7, 3,0 ou 15,0mg/kg do extrato aquoso da I. carnea nos dias 5 a 21 de gestação foram estudados. Foram avaliados a performance reprodutiva materna, anormalidades esqueléticas e viscerais e malformações. Além disso, após o tratamento foram encontrados achados anatomopatológicos. Em relação às ratas mães, nossos resultados mostraram que a exposição às diferentes doses não afetou o peso corporal, ganho de peso, consumos de água e ração e performance reprodutiva. Apesar disso, apresentaram vacuolização citoplasmática de forma dose-dependente em fígado, rins, tireóide e glândula adrenal. Exames fetais não mostraram anormalidades externas ou malformações, sendo somente encontradas evidências de anormalidades esqueléticas e viscerais após altas doses do extrato. Foi observada redução dos centros de ossificação. Os presentes dados mostram que a ingestão prenatal do extrato de I. carnea induz embriotoxicidade. Estes efeitos são atribuídos à ação na homeostase maternal ou diretamente na concepção. | The embryotoxic effects of prenatal daily exposure to 0.0, 0.7, 3.0 or 15.0 mg/kg of the aqueous extract (AQE) from Ipomoea carnea (I. carnea) dried leaves on gestational days 521 were studied in rats. Maternal reproductive performance, skeletal and visceral abnormalities, and malformations were evaluated. Moreover, anatomopathological findings in dams following the treatment were recorded. Regarding the dams, our results show that body weight, weight gain, food and water consumption, and reproductive performance were all unaffected by exposure to the different doses of the AQE. Nonetheless, dams treated with AQE presented a dose-dependent cytoplasmic vacuolation in the liver, kidneys, thyroid and adrenal glands. Fetal examination did not show external abnormalities or malformations. Evidences of several skeletal and visceral abnormalities were found, particularly after the higher dose of AQE. A reduced ossification centers were also detected. The present data show that prenatal ingestion of the I. carnea AQE in rats induces embryotoxicity. These effects are attributed to an active principle from I. carnea acting on maternal homeostasis, or directly in the conception.

The helminth community infecting Miniopterus natalensis was studied at two localities, the De Hoop Nature Reserve (DHNR) (n = 57), Western Cape Province and Pretoria (n = 12), Gauteng Province, South Africa. Hosts from the DHNR had formed part of an earlier, unrelated study and were all pregnant females. A single hymenolepidid cestode species, the nematodes Molinostrongylus ornatus and Litomosa chiropterorum together with nematodes of the subfamily Capillariinae were present at both study sites, while a single digenean, Allassogonoporus sp., was only found in hosts from the DHNR. The prevalence of helminth infections was high at both localities, 68.4 % in the DHNR and 77.7 % in Pretoria, whereas the mean intensity of infection was low at the DHNR (3.76 3.15), but higher in Pretoria (10.4 9.9). Molinostrongylus ornatus and, to a lesser extent L. chiropterorum, were the main contributors to the higher intensities in Pretoria. The species richness ranged from 0 to 4 at both localities.

Foot-and-mouth disease (FMD) is an economically important disease of cloven-hoofed animals that is primarily controlled by vaccination of susceptible animals and movement restrictions for animals and animal-derived products in South Africa. Vaccination using aluminium hydroxide gel-saponin (AS) adjuvanted vaccines containing the South African Territories (SAT) serotypes has been shown to be effective both in ensuring that disease does not spread from the endemic to the free zone and in controlling outbreaks in the free zone. Various vaccine formulations containing antigens derived from the SAT serotypes were tested in cattle that were challenged 1 year later. Both the AS and ISA 206B vaccines adjuvanted with saponin protected cattle against virulent virus challenge. The oilbased ISA 206B-adjuvanted vaccine with and without stimulators was evaluated in a field trial and both elicited antibody responses that lasted for 1 year. Furthermore, the ISA 206 adjuvanted FMD vaccine protected groups of cattle against homologous virus challenge at very low payloads, while pigs vaccinated with an emergency ISA 206B-based FMD vaccine containing the SAT 1 vaccine strains were protected against the heterologous SAT 1 outbreak strain.

Spirocerca lupi (Spirurida: Spirocercidae) is a cosmopolitan parasite, principally of domestic dogs and dung beetles are its main intermediate hosts. In South Africa there has recently been growing concern over the upsurge of reported cases of clinical spirocercosis in dogs, while little is known or understood about the dynamics of the host-parasite associations between dung beetles and this nematode. We determined and compared the prevalence of infection in dung beetles between rural, urban and periurban areas of Tshwane (Pretoria) Metropole. Dung beetles were sampled during April and October 2006, at various localities in each of these areas. Localities were selected on the basis of being focal areas of high infection with S. lupi in dogs. Pig, dog and cow dung-baited pitfall traps were used for sampling the beetles. Trap contents were collected 48 h after the traps had been set and only dung beetles were collected from the traps. In total, 453 specimens belonging to 18 species were collected from 63 pitfall traps in all three areas. The numbers of species that were collected varied among the three areas. Dung beetles, irrespective of species (18) and numbers (447), predominantly preferred pig dung. The prevalence of dung beetles infected with the larvae of S. lupi varied considerably in the three areas. In the urban area 13.5 % of the dung beetles dissected were infected, while the prevalence of S. lupi in dung beetles in the rural area was 2.3 %. All the dung beetles that were infected with this nematode showed a preference for omnivore (pig and dog) dung.

Foot-and-mouth disease (FMD) is an acute, highly contagious viral infection of domestic and wild cloven-hoofed animals. The virus is a single-stranded RNA virus that has a high rate of nucleotide mutation and amino acid substitution. In southern Africa the South African Territories (SAT) 1-3 serotypes of FMD virus are maintained by large numbers of African buffaloes (Syncerus caffer), which provide a potential source of infection for domestic livestock and wild animals. During February 2001, an outbreak of SAT-2 was recorded in cattle in the FMD control zone of South Africa, adjacent to the Kruger National Park (KNP). They had not been vaccinated against the disease since they form the buffer between the vaccination and free zones but in the face of the outbreak, they were vaccinated as part of the control measures to contain the disease. The virus was, however, isolated from some of them on several occasions up to May 2001. These isolates were characterized to determine the rate of genetic change in the main antigenic determinant, the 1D/2A gene. Nucleotide substitutions at 12 different sites were identified of which five led to amino acid changes. Three of these occurred in known antigenic sites, viz. the GH-loop and C-terminal part of the protein, and two of these have previously been shown to be subject to positive selection. Likelihood models indicated that the ratio of non-synonymous to synonymous changes among the outbreak sequences recovered from cattle was four times higher than among comparable sequences isolated from wildlife, suggesting that the virus may be under greater selective pressure during rapid transmission events.

The South African Association for Marine Biological Research, incorporating Sea World, Oceanographic Research Institute and the Dolphinarium relocated from their old premises of 46 years on Durban’s beach front in March and April 2004 to the new Point Development’s marine theme park, uShaka Marine World in Durban, South Africa. Netting of fishes from the large exhibits could cause physical damage to struggling fishes, increasing the risk of opportunistic secondary bacterial infections (Inoue, dos Santos Neto & Morales 2004) which could not be treated in time for the opening of uShaka Marine World on 30 April 2004.
