Purpose. Методы. Молекулярно-генетические методы (выделение геномной ДНК, полимеразная цепная реакция (ПЦР), электрофорез продуктов амплификации в полиакриламидном геле), полевые методы (фенологические наблюдения даты колошения и анализ признаков «высота растений», «длина колоса» и «длина остей», визуальные – оценка признака «цвет колоса»), статистические методы (определение средних значений агрономических признаков с помощью описательной статистики с использованием программы Excel из пакета Microsoft Office, дисперсионный анализ выполнен процедурой GLM (генеральная линейная модель) из специализированного программного комплекса AGROBASE 21). Результаты. В течение четырех лет (2010/11, 2011/12, 2012/13, 2013/14) 47 сортов пшеницы мягкой озимой были изучены по фенотипическим признакам и проанализированы с помощью 17 микросателлитных локусов. Выявлено 35 ассоциаций маркеров с датой колошения, 39 – с высотой растений, 33 – с размером остей, 20 – с цветом колоса и 8 – с размером колоса; ассоциации были стабильными и достоверными на протяжении 2–4 лет исследования. Выводы. Микросателитные маркеры, которые обнаружили значительные и стабильные в разные годы ассоциации с агрономическими признаками, могут быть полезными и пригодными для отбора с помощью маркеров (MAS) в рамках программ селекции пшеницы Украины. | Мета. Визначення асоціацій мікросателітних маркерів зі специфічними областями геному, які контролюють важливі агрономічні ознаки, на виборці сортів пшениці м’якої озимої селекції СГІ – НЦНС, зареєстрованих в Державному реєстрі сортів, придатних для поширення в Україні, протягом різних років. Методи. Молекулярно-генетичні методи (виділення геномної ДНК, полімеразна ланцюгова реакція (ПЛР), електрофорез продуктів ампліфікації в поліакриламідному гелі), польові методи (фенологічні спостереження дати колосіння та аналіз ознак «висота рослин», «довжина колосу» та «довжина остюків», візуальні – оцінка ознаки «колір колосу»), статистичні методи (визначення середніх значень агрономічних ознак за допомогою описової статистики з використанням програми Excel із пакету MicrosoftOffice, дисперсійний аналіз виконано процедурою GLM (генеральна лінійна модель) зі спеціалізованого програмного комплексу AGROBASE 21). Результати. Протягом чотирьох років (2010/11, 2011/12, 2012/13, 2013/14) 47 сортів пшениці м’якої озимої, були вивчені за фенотиповими ознаками та проаналізовані за 17 мікросателітними локусами. Виявлено 35 асоціацій маркерів з датою колосіння, 39 – з висотою рослин, 33 – з розміром остей, 20 – з кольором колосу і 8 – з розміром колосу; асоціації були стабільними та достовірними протягом 2–4 років дослідження. Висновки. Мікросателітні маркери, які виявили значні та стабільні в різні роки асоціації з агрономічними ознаками, можуть бути корисними й придатними для добору за допомогою маркерів (MAS) у рамках програм селекції пшениці України.

Field study, laboratory test, analytical approach and statistical evaluation. Results. Effect of sowing time on economic characters of common fenugreek (Trigonella foenum graecum L.) and blue fenugreek (Trigonella coerulea (Desr.) Ser.) was investigated. It was found that sowing time significantly influenced the individual productivity of plants and average yield of their fresh and dry matter. In particular, common and blue fenugreek plants formed not only the most developed vegetation but also higher productivity and fresh and dry matter yield, respectively, due to early-spring sowing. In case of summer sowing, the poorest yield of dry matter per unit was obtained for these species caused by a low fresh yield, though the moisture loss index was lower. Conclusions. Common fenugreek is promising for producing “mushroom grass” spices, and the conveyor supply of production in the Forest-Steep zone of Ukraine can be ensured by the first sowing time (the first or the second decades of April) and the se­cond sowing time (the third decade of April) as during this period more developed vegetation is formed that favored the higher yield of dry matter (1,3–1,5 t/ha).

Определить влияние сроков сева на продуктивность разных видов пажитника в условиях Лесостепи Украи­ны. Методы. Полевой, лабораторный, аналитический и статистический. Результаты. Исследовано влияние сроков сева на хозяйственно-ценные признаки пажитника сенного (Trigonella foenum graecum L.) и пажитника голубого (Trigonella coerulea (Desr.) Ser.). Установлено, что на индивидуальную продуктивность растений и среднюю урожайность их зеленой и сухой массы существенно влиял срок сева. В частности, более развитую вегетативную массу и продуктивность и, как следствие, большую урожайность зеленой и сухой массы имели растения пажитника голубого и сенного при ранневесенних сроках сева. Самый маленький выход сухой массы этих видов с единицы площади получено при летних сроках сева из-за небольшой урожайности зеленой массы, несмотря на более низкий показатель потери влажности. Выводы. Для получения пряности «грибная трава» перспективным является использование пажитника сенного, а для конвейерного поступления продукции в Лесостепи Украины – применение 1-го (I–II декады апреля) и 2-го (III декада апреля) сроков сева, при которых формируется более развитая вегетативная масса и установлена большая урожайность сухой массы (1,3–1,4 т/га). | Мета. З’ясувати вплив строків сівби на продуктивність різних видів гуньби в умовах Лісостепу України. Методи. Польовий, лабораторний, аналітичний та статистичний. Результати. Досліджено вплив строків сівби на господарсько-цінні ознаки гуньби сінної (Trigonella foenum-graecum L.) та гуньби голубої (Trigonella coerulea (Desr.) Ser.). Встановлено, що на індивідуальну продуктивність рослин та середню врожайність їх зеленої та сухої маси істотно впливав строк сівби. Зокрема, розвиненішу вегетативну масу й продуктивність та, як наслідок, вищу врожайність зеленої та сухої маси мали рослини гуньби голубої та сінної за ранньовесняних строків сівби. Найменший вихід сухої маси цих видів з одиниці площі отримано за літніх строків сівби через невелику врожайність зеленої маси, незважаючи на нижчий показник втрати вологості. Висновки. Для отримання прянощів «грибна трава» перспективним є використання гуньби сінної, а для конвеєрного надходження продукції в Лісостепу України – застосування 1-го (I–II декади квітня) та 2-го (III декада квітня) строків сівби, за яких формується розвиненіша вегетативна маса та встановлено вищу врожайність сухої маси (1,3–1,4 т/га).

Developing technology for clonal micropropagation of peppermint (Mentha piperita L.) plants of Ukrainian breeding based on the complex of methods of isolated tissue and organ culture in vitro. Methods. During the experiment, such methods as isolated tissue and organ culture in vitro, clonal micropropagation, detached scion grafting, chemotherapy with adding of virucide Ribavirin to the nutrient medium, biometric and statistical ones were used. Results. The stepped procedure of sterilization that we have developed allows to receive 88–100% of sterile explants. For M. piperita L. introduction into culture and clonal micropropagation, Murashige and Skoog (MS) nut­rient medium appeared to be optimal supplemented with 6-benzylaminopurine (0.75 mg/l), adenine (0.05 mg/l), indole-3-acetic acid (IAA) (0.05 mg/l) and gibberellic acid (0.5 mg/l) on which the reproduction ratio on the 28th day ranged between 1:7 and 1:15. For recovery of plants from viral infection, virucide Ribavirin at concentration of 10 mg/l was added to the nutrient medium. The proposed nutrient medium for rhizogenesis, that contained IAA (0.5 mg/l) and indole butyric acid (IBA) (0.5 mg/l), allows to obtain the frequency of rhizogenesis up to 84–100%. Regenerated plants were adapted to the conditions in vivo on substrate peat : universal soil : perlite : sand in the ratio 2:1:1:1. The survival rate for peppermint varieties amounted to 96–100%. Conclusions. Biotechnological scheme was developed that permits to get healthy, purebred planting material and intensively propagate plants for supplying breeding programs of the Experimental Station for Medicinal Plants of the Institute of Agroecology and Environmental Management of the National Academy of Agrarian Sciences of Ukraine, among which such varieties as ‘Lebedyna pisnia’ and ‘Ukrainska pertseva’ were selected as the most promising for clonal micropropagation.

Для оздоровления растений от вирусной инфекции в питательную среду добавляли вироцидRibavirinв концентрации 10 мг/л. Предложенная питательная среда для ризогенеза, содержащая по 0,5 мг/л индолил-3-уксусной кислоты и индолилмаслянной кислоты,усиливала частоту ризогенеза до 84–100%.Растения-регенеранты адаптировали к условиям in vivo на субстрате: торф : почва универсальная : перлит : песок в соотношении 2:1:1:1. Степень приживаемости растений сортов мяты перечнойсоставляет 96–100%. Выводы.Разработана биотехнологическая схема, которая позволяет получать оздоровленный и чистосортный посадочный материал и интенсивно размножать растения для обеспечения селекционных программ Опытной станции лекарственных растений Института агроэкологии и природопользования НААН Украины, среди которых выделяются сорта мяты перечной ‘Лебединая песня’ и ‘Украинская перечная’, как наиболее перспективные для клонального микроразмножения. | Мета. У процесі екс­перименту використовували метод культури ізольованих тканин і органів invitro, клональне мікророзмноження, живцювання, хемотерапію з додаванням у живильне середовище віроцида Ribavirin, біометричний та статистичний. Результати. Розроблена ступінчаста методика стерилізації зумовлює отримання 88–100% стерильних експлантатів. Для введення в культуру й клонального мікророзмноження м’яти перцевої оптимальним виявилось живильне середовище Мурашіге й Скуга (МС), доповнене 0,75 мг/л 6-бензиламінопурину, 0,1 мг/л аденіну, 0,05 мг/л індоліл-3-оцтової кислоти та 0,5 мг/л гіберелової кислоти, на якому коефіцієнт розмноження на 28 добу коливався в межах від 1:7 до 1:15. Для оздоровлення рослин від вірусної інфекції у живильне середовище додавали віроцид Ribavirin у концентрації 10 мг/л. Запропоноване живильне середовище для ризогенезу, що містить 0,5 мг/л індоліл-3-оцтової кислоти і 0,5 мг/л індолілмасляної кислоти, посилює частоту ризогенезу до 84–100%. Рослини-регенеранти адаптували до умов in vivo на субстраті: торф : ґрунт універсальний : перліт : пісок у співвідношенні 2:1:1:1. Ступінь приживлюваності рослин сортів м’яти перцевої становить 96–100%. Висновки. Розроблено біотехнологічну схему, яка дає можливість отримувати оздоровлений і чистосортний садивний матеріал та інтенсивно розмножувати рослини для забезпечення селекційних програм Дослідної станції лікарських рослин Інституту агроекології і природокористування НААН України, серед яких виділено сорти м’яти перцевої ‘Лебедина пісня’ та ‘Українська перцева’ як найперспективніші для клонального мікророзмноження.

Field, pot experiment, biometrical, gas analysis, analytical and statistical ones. Results. It is found that the modern high-yielding winter wheat varieties have a higher rate of flag leaf photosynthesis during generative period of development, particularly at grain filling period, improved ability to store assimilates in stem and effectively use them later for grain growth. The modern varieties form crop canopy with greater leaf area and chlorophyll indexes and larger photosynthetic capacity and maintain functional activity of photosynthetic apparatus longer at the end of the growing season. Conclusions. The superiority for grain productivity of modern high-yielding wheat varieties, originated from the Institute of Plant Physio­logy and Genetics, NAS of Ukraine, over varieties released before the “green revolution”, results from increased capacity and durability of functioning the crop photosynthetic apparatus, higher CO2 assimilation rate and optimization of source-sink relations in whole plant.

Установлено, что новые высокоинтенсивные сорта озимой пшеницы имеют более высокую интенсивность фотосинтеза флагового листа в течение генеративного периода развития, особенно во время налива зерна, улучшенную способность запасать ассимиляты в стебле и эффективно использовать их в дальнейшем для налива колоса. Новые сорта формируют посевы с большими значениями хлорофильного индекса и хлорофильного фотосинтетического потенциала и более длительным сохранением функциональной активности фотосинтетического аппарата в конце вегетации. Выводы. Преимущество по зерновой продуктивностью новейших высокоинтенсивных сортов пшеницы, оригинатором которых является Институт физиологии растений и генетики НАН Украины, перед сортами, созданными до начала «зеленой революции», обеспечивается повышением мощности и продолжительности функционирования фотосинтетического аппарата посева, более высокой интенсивностью фотосинтеза и оптимизацией донорно-акцепторных отношений в системе целого растения. | Мета. Порівняльний аналіз показників фотосинтетичного апарату сортів озимої пшениці сучасної і більш ранньої селекції у зв’язку з їх продуктивністю. Методи. Польовий, вегетаційний, фітометричний, газометричний, аналітичний, статистичний. Результати. Встановлено, що нові високоінтенсивні сорти озимої пшениці мають вищу інтенсивність фото­синтезу прапорцевого листка протягом генеративного періоду розвитку, особливо під час наливу зерна, поліпшену здатність запасати асиміляти в стеблі та ефективно використовувати їх в подальшому для наливання колоса. Нові сорти формують посіви з більшими значеннями хлорофільного індексу й хлорофільного фотосинтетичного потенціалу та тривалішим збереженням функціональної активності фотосинтетичного апарату в кінці вегетації. Висновки. Перевага за зерновою продуктивністю новітніх високоінтенсивних сортів пшениці, оригінатором яких є Інститут фізіології рослин і генетики НАН України, порівняно з сортами, створеними до початку «зеленої революції», забезпечується підвищенням потужності й тривалості функціонування фотосинтетичного апарату посіву, вищою інтенсивністю фотосинтезу та оптимізацією донорно-акцепторних відносин у системі цілої рослини.

Based on this, assumption can be made about the possibility to evaluate a sample for the presence of granules larger than 35 µm that can significantly reduce the number of measurements and speed up analysis.

Установлен значительный полиморфизм тритикале озимого по размеру гранул крахмала. Выявлено, что наименьший средний размер крахмальных зерен наблюдался у образцов с наименьшим максимальным размером гранул, низкой частотой гранул с размерами более 35 мкм и, соответственно, минимальной дисперсией. На основании этого можно сделать допущение о возможности оценивания образца по наличию гранул размером более 35 мкм, что значительно уменьшит количество измерений и ускорит анализ. | Мета. Визначити можливість диференціації зразків за розміром крохмальних зерен та виявлення зразків з мінімальним розміром гранул, що має значну позитивну кореляцію з ефективністю трансформації крохмалю в етанол. Розмір гранул крохмалю у сорту пшениці озимої м’якої ‘Цвіт Калини’ становив 24,2 мкм, що було в межах значень, типових для сортів та ліній тритикале озимого. У переважної більшості гранул розмір варіював від 15 до 35 мкм, але розподіл їх за розміром був нерівномірним у досліджуваних ліній. Висновки. Встановлено значний поліморфізм тритикале озимого за розміром гранул крохмалю. Виявлено, що найменший середній розмір крохмальних зерен спостерігався у зразків з найменшим максимальним розміром гранул, низькою частотою трапляння гранул з розмірами понад 35 мкм та, відповідно, мінімальною дисперсією. Це дає підставу зробити припущення про можливість оцінювання зразка за наявністю гранул розміром понад 35 мкм, що значно зменшить кількість вимірювань та прискорить аналіз.

The article presents the results of studies to determine the parameters of the polymerase chain reaction (PCR) in order to develop a multiplex system for identification of the structural elements of the design of transgenic gene cp 4 epsps, which provides tole­rance to glyphosate. For amplicon target DNA sequences, the following values of temperature conditions of PCR were determined: step 1 (initial denaturation) 95 °C – 3 min; step 2 (specific reaction products accumulation): denaturation 95 °C – 45 s; hybridization of primers 55 °C – 50 s; elongation 72 °C – 1 min; number of cycles – 40; step 3 (final elongation) 72 °C – 6 min. A series of PCR were carried out for the purpose of selecting the optimal amount of DNA matrix for efficient estimate of transgenic sugar beet plants for the presence of specific sequences. Conclusions. To identify transgenic glyphosate-tolerant sugar beet, it is advisable to determine 35S promoter and gene cp 4 epsps in individual genotypes. It was found that during the selection of temperature parameters of multiplex reaction a 5 °C rise in primer hybridization temperature did not affect the identification of gene als that allowed to include specific primers for determination of this sequence as an internal control. Based on the results of test multiplex reactions, concentrations of dNTPs and Mg2+ ions were determined that allowed to exclude the possibility of non-specific fragments and false-negative results. The optimum amount of matrix DNA (100–150 ng) for an efficient estimate of transgenic sugar beet plants for the presence of specific sequences was determined. Obtained results allowed to develop a multiplex test system for identification of transgenic glyphosate-to­lerant sugar beet which can be used for simultaneous determination of the 35S promoter, cp 4 epsps gene and als gene as an internal reaction control.

Merrill) as growing season length in F2 and assessing hybrid combinations to identify more quick-ripening phenotypes as compared to parents. Methods. Laboratory test, mathematico-statistical evaluation. Results. In most crossbreeding combinations, when parents differed by growing season length, late ripeness was dominated in F2, in one combination – early ripeness, in two combinations, when parents scarcely differed by growing season length, complementary effect was observed for this index. It was found that ‘Anzhelika’/‘Mageva’ combination generated the highest number of more quick-ripening forms than any of the parents (13.1%), a smaller number was identified in ‘Legenda’/‘Vizhion’ (6.4%) and ‘Anzhelika’/‘Gentleman’ (4.0%), and barely noticeable number was observed in ‘Legenda’/‘Yelena’ combination (1.3%). Conclusions. Найбільше форм, скоростигліших за одну з батьківських, виділено в комбінації ‘Анжеліка’/‘Магева’ (13,1%), менше – в ‘Легенда’/‘Віжіон’ (6,4%), ‘Анжеліка’/‘Джентльмен’ (4,0%) і зовсім незначну їх кількість – 1,3% виявлено в ‘Легенда’/‘Єлена’. Висновки. В комбінаціях схрещування ‘Легенда’/‘Віжіон’, ‘Легенда’/‘Корадо’, ‘Легенда’/‘Устя’, ‘Легенда’/‘Єлена’, ‘Юг-30’/‘Джентльмен’, ‘№ 894’/‘Віжіон’, ‘Анжеліка’/‘Аннушка’, ‘№ 894’/‘Аннушка’, ‘Легенда’/‘Аннушка’, ‘№ 441’/‘Джентльмен’, ‘№ 441’/‘Віжіон’, ‘№ 441’/‘Аннушка’, ‘Анжеліка’/‘Джентльмен’, ‘Анжеліка’/‘Прип’ять’, де батьківські форми значно й мало відрізнялися за тривалістю періоду вегетації, в F2 виявлено домінування пізньостиглості. В комбінаціях схрещування ‘Устя’/‘Віжіон’ і ‘Юг-30’/‘Віжіон’, де батьківські форми практично не відрізнялися за періодом вегетації, в F2 спостерігався комплементарний ефект за цим показником.

Purpose. To develop a conceptually new method for determination of varietal purity (typicality), hybridity, steri­lity of seed lots. Methods of molecular biology (genomic DNA extraction, PCR with SSR markers application, capillary electrophoresis), genetic, statistical, mathematical analysis. Results. Разработать принципиально новый метод определения сортовой чистоты (типичности), гибридности, стерильности партий семян. Методы молекулярной биологии (экстракция геномной ДНК, ПЦР с применением SSR-маркеров, капиллярный электрофорез), генетический и статистически-математический анализ. Результаты. Разработан новый метод определения сортовых качеств партий семян, состоящий из следующих этапов: одновременное выделение ДНК из репрезентативной совокупности семян оцениваемого образца; ПЦР и дальнейший анализ продуктов амплификации путем определения качественного и количественного состава аллелей маркерных SSR-последовательностей; расчет сортовых показателей качества партии семян с использованием экспериментально полученных соотношений аллелей. Выводы. Разработанный метод определения сортовых качеств партий семян позволяет значительно сократить расход материалов, времени и труда при выполнении анализа. Последовательное качественное и количественное определение аллелей в совокупном образце партии семян является принципиально новым подходом для установления сортовой чистоты (типичности), гибридности, стерильности. | Мета. Розроблено новий метод визначення сортових якостей партій насіння, що складається з таких етапів: одночасне виділення ДНК із репрезентативної сукупності насінин оцінюваного зразка; ПЛР та подальший аналіз продуктів ампліфікації шляхом визначення якісного та кількісного складу алелів маркерних SSR-послідовностей; розрахунок сортових показників якості партії насіння з використанням експериментально отриманих співвідношень алелів. Висновки. Розроблений метод визначення сор­тових якостей партій насіння дає змогу значно скоротити витрати матеріалів, часу та праці під час виконання аналізу. Послідовне якісне та кількісне визначення алелів у сукупному зразку партії насіння є принципово новим підходом для встановлення сортової чистоти (типовості), гібридності, стерильності.

Field study, comparative analysis, generalization. Results. The collection of roses at M. M. Hryshko National Botanical Garden (the city of Kyiv) was analyzed. A comparison was drawn between their gene pool and the world collection of roses. After collection screening, 22 old varieties were found at M. M. Hryshko National Botanical Garden and described. Their decorative pro­perties, economic characters and winter hardiness were eva­luated, the final result ranged from 81 to 100 points. Evalua­tion of winter hardiness that was based on the records of shoot damage degree in the field during the winter showed that in Kyiv conditions 13 cultivars were exceptionally resistant to frost, the other 9 appeared to be frost-sensitive. Conclusions. As follows from the analysis of decorative va­lues, economic characters and winter hardiness, 12 varieties of old roses such as ‘Cardinal de Richelieu’, ‘Commandant Beaurepaire’, ‘La Reine’, ‘Louise de Odier’, ‘Madame Plantier’, ‘Maiden’s Blush’, ‘Marie Baumann’, ‘Mousseuse Rouge’, ‘Persian Yellow’, ‘Poppius’, ‘Reine des Violettes’, ‘Versicolor’ were found to be perfectly suitable for landscaping.

Оценить результаты интродукции старинных сортов роз на основании анализа декоративных свойств и хозяйственно-ценных признаков. Методы.Полевые, сравнительный анализ, обобщение. Результаты. Проанализирован коллекционный фонд роз Национального ботанического сада имени Н. Н. Гришко (г. Киев). Проведено сравнение генофонда с мировой коллекцией роз. В результате скрининга коллекции в Национальном ботаническом саду имени Н. Н. Гришко выявлено 22 старинных сорта и составлено их описание. Проведена оценка декоративности, хозяйственно-ценных признаков и зимостойкости, которая в итоге составляла от 81 до 100 баллов. Оценка зимостойкости, которая базируется на учете степени зимнего повреждении побегов в полевых условиях, показала, что в условиях Киева оказались полностью зимостойкими 13 сортов, остальные девять оказались недостаточно зимостойкими. Выводы. В результате анализа оценок декоративности, хозяйственно-ценных признаков и зимостойкости было установлено, что для внедрения в ландшафтное строительство подходят 12 сортов старинных роз: ‘Cardinal de Richelieu’, ‘Commandant Beaurepaire’, ‘La Reine’, ‘Louise de Odier’, ‘Madame Plantier’, ‘Maiden’s Blush’, ‘Marie Baumann’, ‘Mousseuse Rouge’, ‘Persian Yellow’, ‘Poppius’, ‘Reine des Violettes’, ‘Versicolor’. | Мета. Оцінити результати інтродукції старовинних сортів троянд на основі аналізу їхніх декоративних властивос­тей та господарсько-цінних ознак.  Методи. Польові, порівняльний аналіз, узагальнення. Результати. Проаналізовано колекційний фонд троянд Національного ботанічного саду імені М. М. Гришка (м. Київ). Здійснено порівняння генофонду зі світовою колекцією троянд. Внаслідок скринінгу колекції в Національному ботанічному саду імені М. М. Гришка було виявлено 22 старовинні сорти та складено їхній опис. Проведено оцінку декоративності, господарсько-цінних ознак та зимостійкості, яка в підсумку становила від 81 до 100 балів. Оцінка зимостійкості, яка базується на обліку ступеня зимового пошкодження пагонів у польових умовах, свідчить, що в умовах Києва виявилися повністю зимостійкими 13 сортів, решта дев’ять були недостатньо зимостійкими. Висновки. Внаслідок аналізу оцінок декоративності, господарсько-цінних ознак та зимостійкості було встановлено, що для впровадження в ландшафтне будівництво є придатними 12 сортів старовинних троянд: ‘Cardinal de Richelieu’, ‘Commandant Beaurepaire’, ‘La Reine’, ‘Louise de Odier’, ‘Madame Plantier’, ‘Maiden’s Blush’, ‘Marie Baumann’, ‘Mousseuse Rouge’, ‘Persian Yellow’, ‘Poppius’, ‘Reine des Violettes’, ‘Versicolor’.
