Bioinformatic analysis of maize gene encoding starch branching enzyme SBEIIb

Purpose. Investigation of maize ae1 gene polymorphism by bioinformatic methods. Methods. Global and local alignment of the nucleotide and amino acid sequences, in silico translation and transcription, translates modeling, primers design, phylogenetic analysis. Results. 255 nucleotide sequences of maize аe1 gene, 500 amino acid sequences of homology translates of maize ae1 gene (SBEIIb enzyme homologs) and 100 mRNA expressed from the maize ae1 gene were analyzed to establish phylogenetic relationships. Polymorphism of maize ae1 gene different regions was investigated by bioinformatic methods. Modeling of the maize enzyme SBEIIb was performed. Conclusions. According to the results of amino acid sequences of SBEIIb enzyme homologs alignment, it was found that ae1 gene orthologs are present only in monocots, paralogs – in monocots, dicots, and other taxa, including algae and animals. Based on the results of alignment of plants mRNA from which enzyme SBEIIb is translated, maize ae1 gene orthologs and the nearest paralogs encoding starch branching enzymes with chloroplast localization were defined; this suggests a possible origin of ae1 gene due to duplication of the gene encoding the 1,4-alpha-glucan-branching enzyme 2 with chloroplast or amyloplast localization. In the maize ae1 gene structure, regions were found that include polymorphic sites not defined previously. For the polymorphic sites design primers were developed that allowed to differentiate the maize lines. It was determined that the detection of polymorphism in theory can influence the enzyme function and, as a result, change the concentration of amylopectin in maize grain.

Purpose. Studying variety samples of winter triticale of various ecological and geographical origin for revealing polymorphism of the culture for its susceptibility to pathogenic complex of Fusarium head blight and defining high-yielding and resistant to diseases varieties that later can be put into the production and breeding process. Methods. Field experiments, statistical evaluation. Results. It was defined that the development of Fusarium head blight in different variety samples of winter triticale depends on the hydrothermal conditions and genotype of the host-plant. Resistance of varieties and hybrids to the disease was partial, and no immune samples were found among the analyzed ones. The following varieties proved to be tolerant to Fusarium head blight: ‘Granat’, ‘Zorro’, ‘Obrii Myronivskyi’. The positive correlation between the development of Fusarium head blight and root rot of winter triticale was found. It was determined that in the pathogenic complex of Fusarium head blight the amount of the following pathogens is increasing: Fusarium sporotrichioides, F. аvenaceum and F. poae. Conclusions. The leading varieties that combine high yields and resistance to Fusarium head blight are as follows: ‘Obrii Myronivskyi’, ‘ADM 8’, ‘Yuvileine Volynske’, ‘Yukon’, ‘Zorro’, ‘Tsekad 90’, ‘Zerniatko’, ‘Legion’ and ‘Rarytet’. These varieties should be involved in the selection process to breed the hybrids with the augmented resistance to the disease and high yields.

Purpose. Review of the main achievements of the Wheat Breeding and Seed ProductionDepartment in the Plant Breeding and Genetic Institute – National Centre of Seed and Cultivar Investigation in the developing theoretical principles of breeding and creation of winter wheat varieties of different types during 100-year (1916–2016) period of breeding programs realization. Results. The main theoretical, methodical developments and breeding achievements of Wheat Breeding and Seed Production Department during 100-year (1916–2016) history have been considered. In the course of the Department activity, the research and methodology grounds of bread winter wheat breeding and seed production have been laid, 9 stages of breeding programs development have been accomplished. As a result, more than 130 varieties of different types have been created, 87 of them have been released in some periods or registered in the State registers of plants varieties of Ukraine and other countries and grown in the total sowing area about 220 million hectares.

Purpose. The analysis of allelic status of such key genes of carotenogenesis as gene of lycopene-ε-cyclase (lcyε) and gene of β-carotene hydroxylase (crtRB1) for DNA-markers in domestic maize lines and their hybrids. Methods. DNA isolation, PCR, gel electrophoresis. Results. Allelic status of key genes of carotenogenesis was investigated in eight maize inbred lines and their single crosses. Molecular genetic polymorphism in the studied sample of maize lines and hybrids has been detected in gene of β-carotene hydroxylase for marker crtRB1-3’TE. For this gene, codominant character of inheritance of alleles of parental lines in single crosses was confirmed. For markers of gene of lycopene-ε-cyclase lcyε-3’INDL and lcyε-SNP216, polymorphism in the group of investigated lines and hybrids has not been identified, genotypes included only one variant of alleles for each marker. For lines ‘DK253ZSZM’, ‘DK633/266zS,zM’, ‘DK366zS,zM’ and hybrids ‘DK296S×DK253ZSZM’, ‘DK272S×DK633/266zS,zM’ and ‘DK231S×DK366zS,zM’, the decrease of the activity of β-carotene hydroxylase owing to the mutation of gene crtRB1 under the influence of transposone element at the 3’-end, the inhibition of β-carotene transition into β-cryptoxanthin can be expected, that allows to predict β-carotene accumulation in grain. Conclusions. The study of allelic status of carotenegenesis gene of lycopene-ε-cyclase in maize showed no polymorphism for markers lcyε-3’INDL and lcyε-SNP216 in eight inbred lines and their single crosses, along with this, for marker lcyε-3’INDL in genomes of all studied samples the allele was identified to be favorable for the accumulation of β-carotene. For marker crtRB1-3’TE of gene of β-carotene hydroxylase, the studied breeding material was polymorphic. Allele of crtRB1 being favorable for the accumulation of β-carotene was identified in lines ‘DK253ZSZM’, ‘DK633/266zS,zM’, ‘DK366zS,zM’ and hybrids ‘DK296S×DK253ZSZM’, ‘DK272S×DK633/266zS,zM’ and ‘DK231S×DK366zS,zM’. Single crosses inherit maternal and paternal alleles of gene of β-carotene hydroxylase codominantly.

Purpose. To develop an algorithm of creation of new highly productive clonal varieties of Salicaceae family representatives with improved agronomic characters that can be used at energy plantations Methods. Field investigations, laboratory analysis, analytical approach, selection method. Results. Flow chart of selection process with representatives of Salicaceae family included the following stages: the Ist stage – creation of primary families (F0); the IInd stage – creation of the second families, or branches; the 222rd stage – interfamily (individually-familial) selection; IVth stage – familial-group selection; the Vth stage – clonal selection (selection of ramets). Conclusions. Breeding process with the use of complex of advanced methods of selection (individual, familial, group and clonal ones) allows to get model populations with high-frequency of valuable alleles and genotypes and with the high level of genetic variety. Due to a negative multi-stage individual intrafamilial and interfamily selection, cleaning of artificial populations from undesirable forms takes place without the substantial decrease of genetic variety level.

Medicinal plants: traditions and study prospects (devoted to the 100th anniversary of establishing the Experimental Station of Medicinal Plants)

Historical review was done concerning the formation and development of scientific research of the Experimental Station of Medicinal Plants over the period 1916–2016. The main stages of experimenting in the direction of “medicinal plant cultivation’’ in Ukraine were covered. Role and importance of the Experimental Station of Medicinal Plants in the scientific and methodological support of such scientific directions as breeding, seed production, cultivation technologies, pharmacognosy in medicinal plant cultivation were shown. For the needs of the branch, over 100 species of introduced medicinal plants were studied at the Station and agricultural techniques of their growing were developed, in addition, more than 50 varieties of medicinal plants were created, a number of machines and devices for growing, harvesting and initial treatment of raw materials and seeds of the most important medicinal plants was designed. Other scientific achievements of the Station were mentioned and promising areas of medicinal plant research in Ukraine were outlined.

Purpose. Improvement of a range of strawberry varieties based on comprehensive study and introduction of new Ukrainian and West European cultivars. Methods. Cultivars and elite forms were studied using the methods of primary variety studying and state variety testing at three-year strawberry plantations of the Institute of Horticulture of the National Academy of Sciences of Ukraine (the city of Kyiv) and its variety testing stations. Results. It was found that more frequent spring and summer droughts as well as black frosts in winter adversely affected the strawberry productivity. The best varieties were selected among new ones for the complex of traits, they included ‘Elegance’ from the Great Britain, ‘Gercules’ from Ukraine (both are dessert grapes), ‘Elsanta’ from Holland (for universal use), ‘Alba’ from Italia (as raw material). The highly adaptive and productive ‘Istochnik’ and ‘Festyvalna romashka’ varieties (as raw material) are recommended for the north regions. Conclusions. In the context of profound climatic change, high-profitable cultivation of mid-ripening varieties and a strawberry as such in the Northern Forest-Steppe zone is expedient under controlled conditions – in high film tunnels, in hot-houses and under perennial irrigation that already has become the European tendency.

Purpose. Determination of sowing time effect on productivity of various fenugreek species in the Forest-Steppe zone of Ukraine. Methods. Field study, laboratory test, analytical approach and statistical evaluation. Results. Effect of sowing time on economic characters of common fenugreek (Trigonella foenum graecum L.) and blue fenugreek (Trigonella coerulea (Desr.) Ser.) was investigated. It was found that sowing time significantly influenced the individual productivity of plants and average yield of their fresh and dry matter. In particular, common and blue fenugreek plants formed not only the most developed vegetation but also higher productivity and fresh and dry matter yield, respectively, due to early-spring sowing. In case of summer sowing, the poorest yield of dry matter per unit was obtained for these species caused by a low fresh yield, though the moisture loss index was lower. Conclusions. Common fenugreek is promising for producing “mushroom grass” spices, and the conveyor supply of production in the Forest-Steep zone of Ukraine can be ensured by the first sowing time (the first or the second decades of April) and the second sowing time (the third decade of April) as during this period more developed vegetation is formed that favored the higher yield of dry matter (1,3–1,5 t/ha).

Purpose. Selecting spring wheat lines with increased adaptive capacity. Methods. Field experiments, statistical evaluation. Results. Lines of spring soft wheat with better adaptive capacity were identified (‘Erythrospermum 13-39’, ‘Lutescens 11-16’, ‘Lutescens 05-24’, ‘Lutescens 10-36’) and spring durum wheat (‘Melanopus 10-02’ (biotype II), ‘Melanopus 10-02’, ‘Melanopus 10-03’, ‘Hordeiforme 12-12’). Spring soft wheat varieties ‘Zlata’, ‘Dubravka’, ‘Oksamyt Myronivskyi’, ‘Bozhena’ and spring durum wheat varieties ‘MIP Raiduzhna’, ‘Magdalena’ with increased adaptive capacity were passed for State variety testing. Conclusions. With regard to the analysis of parameters of yield capacity and its variability under the influence of changing environmental factors, soft and durum spring wheat lines with increased adaptive capacity were identified. The best spring wheat lines for general adaptive capacity were passed for State variety testing. Soft spring wheat variety ‘Zlata’ and durum spring wheat variety ‘MIP Raiduzhna’, in 2014, and such soft spring wheat varieties as ‘Dubravka’, ‘Oksamyt Myronivskyi’, ‘Bozhena’ and spring durum wheat variety ‘Magdalena’, in 2015, were passed for State variety testing.
