Біохімічні показники насіння сочевиці залежно від елементів технології вирощування

Мета. Дослідити вплив строків сівби, мікродобрив та регуляторів росту на біохімічні показники насіння сочевиці. Методи. Польові, лабораторні. Результати. Наведено біохімічні показники якості насіння сочевиці залежно від строків сівби, дії мікродобрив, регуляторів росту та їх поєднання. На формування біохімічних показників насіння сочевиці впливають різні чинники. Вміст сирої клітковини в середньому за роки досліджень був 3,2–3,3% на контрольному варіанті, максимальні значення – 6,9% за застосування Реаком-СР-Бобові + Регоплант за 1-го строку сівби та 8,4% Квантум-Бобові за 2-го строку. На показник вмісту крохмалю в абсолютно сухій речовині насіння найбільше вплинуло застосування регулятора росту Стимпо – 56,5% у 1-й строк сівби та поєднання його з мікродобривом Квантум-Бобові – 54,3% у 2-й строк. Загалом за роки дослідження в насінні сочевиці сорту ‘Лінза’ вміст сирого протеїну був на рівні 26–28%. За комбінованого застосування мікродобрив і регуляторів росту 27,81% – Реаком-СР-Бобові + Стимпо за 1-го строку сівби та від 27,69% – Квантум-Бобові + Регоплант до 28,60% – Реаком-СР-Бобові за 2-го строку. Висновки. Встановлено максимальні показники вмісту сирої клітковини у варіантах з підживленням рослин у фазі бутонізації мікродобривом Квантум-Бобові – 4,5% за 1-го строку сівби і 10,8% за 2-го строку в 2016 р. Максимальний вміст крохмалю в насінні сочевиці в 2016 р. за обох строків сівби був у варіанті із підживленням у фазі бутонізації регулятором росту Стимпо – 58,0 та 55,8% відповідно. У 2017 р. кращі показники були у варіантах Квантум-Бобові та Квантум-Бобові + Регоплант – 55,1% за 1-го строку та у варіанті Реаком-СР-Бобові – 54,3% за 2-го строку. Вміст сирого протеїну в насінні сочевиці в 2016 р. у варіантах Реаком-СР-Бобові + Регоплант – 27,92% за 1-го строку сівби та Реаком-СР-Бобові + Стимпо – 29,93% за 2-го. У 2017 р. у варіанті Регоплант – 28,21% за 1-го та Квантум-Бобові – 28,39% за 2-го строку.

Purpose. Analysis of the process of hexaploid triticale breeding for winter hardiness by intraspecific hybridization method using systemic ecological tests under contrasting conditions. Methods. Dialectical investigation, field experiments, laboratory testing and statistical evaluation. Results. The results of breeding winter and alternate triticale varieties possessing a complex of valuable traits by the method of intraspecific hybridization of forms of different types of development using systemic ecological tests under contrasting conditions (Forest-Steppe – extremely arid Steppe) and at low temperatures were presented. During the years of research (1980–2005), 18 varieties were developed and transferred to the state testing, 17 of them were registered. Conclusions. Thus, the effective selection of highly productive genotypes with increased and high winter hardiness is possible from populations obtained by crossing hexaploid triticale of different types of development (winter triticale with spring and alternate ones) and contrasting level of frost and winter hardiness. During the period of 1980–2015, medium-tall and dwarf varieties of winter (‘Amfidyploid 256’, ‘Garne’, ‘Kharroza’, ‘Rarytet’, ‘Timofey’, etc.) and alternate (‘Nikanor’, ‘Yaroslava’, ‘Plastun Volynskyi’) triticales were developed. They are superior to the standard varieties of soft winter wheat for the critical temperature of freezing by -0.5...- 2.0 °C, characterized by increased (up to 9–12 t/ha) grain yield of various quality depending on the purpose of use.

Purpose. To conduct in vitro screening of different genotypes of winter triticale for resistance to salinity in the shoot apical meristem culture. Methods. Plant tissue culture in vitro, in vitro breeding, statistical analysis. Results. It was found that the increase of sodium chloride concentration from 0.6 to 1.5% resulted in inhibition of the callus culture growth in all genotypes that was indicative of the toxic effect of the stress factor. It turns out that 1.2% sodium chloride concentration allowed to differentiate triticale genotypes for salt tolerance. The line ‘38/1296’ appeared to be the most resistant to salinity stress because under breeding conditions calli of this genotype were characterized by higher morphogenetic potential, had the highest crude mass increase, and plants-regenerants were obtained only from explants of this line after cultivation on the medium containing 1.5% sodium chloride. The ‘ADM 11’ variety was the most sensitive to saline stress as mass necrosis and lack of regenerative ability in its calli were observed under breeding conditions. In the studied forms, genotypic dependence of morphogenesis processes in vitro culture was registered. From the induced calli, plants-regenerants were obtained, and their completion of growing, root development and transfer to in vivo conditions were optimized. Conclusions. Genotypic response to salinity stress in the culture of shoot apical meristems of winter triticale was expressed by various crude mass increase and different morphogenetic potential on exposure to a stress factor. The line ‘38/1296’ can be used as a valuable material for further breeding of winter triticale. The culture of shoot apical meristems is recommended to apply as a test system for screening of triticale genotypes for resistance to salinity stress

Purpose. To investigate features of commercial head quality formation in varieties of cutting lettuce (Lactuca sativa var. capitata L.) for different growing methods. To study the influence of growing methods for cutting lettuce plants on the weight of commercial heads, indices of yield and quality of freshly harvested products. Methods. Field study, laboratory testing and statistical evaluation. Results. The results of studying peculiarities of head formation in such cutting lettuce varieties as ‘Godar’ (control), ‘Smuhlianka’, ‘Olzhych’ and ‘Dyvohrai’ were shown. Based on the results of phenological observations, the duration of interstage periods of growth and development of cutting lettuce plants was determined for the different growing methods. The model of cutting lettuce of capitata variety for different growing methods was presented in terms of biochemical parameters (content of dry matter, vitamin C, nitrates, amount of sugars). The influence of growing methods for cutting lettuce plants on the commercial head weight, indices of yield and quality of freshly harvested products was studied. Obtained results confirmed that in early ripening varieties the heads were smaller (150 g) and not dense, in late ripening varieties they were large and dense, weight was 150–500 g. Freshly harvested commercial products of head lettuce in case of seedling cultivation was characterized by a slight increase in dry matter content by 0.15–0.29%. The corresponding pattern was observed for the total sugar content. Its amount was slightly higher: 1.3–1.9% in case of direct sowing and 1.6–1.9% – for seedlings cultivation. The content of ascorbic acid (vitamin C, mg/100 g) was the highest in the ‘Dyvohrai’ variety despite of the method of growing. The largest quantity of nitrates in lettuce heads was in the inner stem (480 mg/kg), while in the middle of the head it was reduced to 110 mg/kg. Conclusions. Freshly harvested heads of cutting lettuce were analyzed for the content of such basic biochemical indices as dry matter, vitamin C, protein, nitrates, amount of sugars. Biochemical characteristics of freshly harvested products of heading lettuce in case of direct sowing and non-seedling methods of growing were within the error being almost identical with minor deviations. According to the study results, a typical model of capitate lettuce variety was designed which will have practical application in qualification examination of plant varieties.

Purpose. To establish correlation dependence of influence of such factors as seeding rate, sowing time and seeding methods on yield indicators, as well as individual response of winter wheat varieties to them in case of the ecological system of seed production, that would allow to form elements of intensive varietal technology and obtain the genetically determined productivity potential of the variety in the future. Methods. Weight measuring was used to determine seed yield; the reliability of the experiment results was identified by statistical evaluation, analysis of variance and regression procedure. Results. During the period of investigation, the weather conditions differed significantly both for the temperature regime and the amount of precipitation, but the stability of grain and seed yield over years was the main requirement to varieties, that allowed to study the intergenotypic correlation relationships of crop yield indices and the dynamics of their changes under the effect of ecological factors. In the course of investigation, winter wheat yield indices were fixed annually from 684 plots, which were grouped and analyzed for factors of influence in order to ensure the complete certainty. Conclusions. Variants of investigation were defined for the certified seed yield and environmental factors (sowing time and seeding method, seeding rate), that had strong positive correlation relationships. Seeding rate and seeding method had the greatest impact on 1000 kernel weight. For tillering coefficient, varieties depending on the variant of the investigation had a negative close correlation relationship, which was greatly affected by seeding rate. For correlation relationships, winter wheat varieties ‘Bohdana’ and ‘Slavna’ appeared to be the most stable, ‘Chorniava’ and ‘Astarta’ were the most plastic.

Purpose. To analyse the qualitative and quantitative composition of volatile organic substances (VOS) in the petals of six species of dog-rose in the collection of M. M. Gryshko National Botanic Garden of the NAS of Ukraine for further breeding. Methods. Volatile organic substances were obtained by steam distillation. VOS was determined using gas chromatography (Chromatograph Agilent Technologies 6890). Results. The qualitative and quantitative composition of volatile organic substances in the petals in such species as Rosa L. (R. roullettii HCh (Correvon), R. multiflora Thunb., R. pimpinellifolia L., R. canina L., R. centifolia L., R. rugosa Thunb.) was investigated. According to the results of the research, 105 VOS were discovered in the dog-rose petals, among which 11 were not identified. The dog-rose petals of identified VOC contained 16 components which share exceeds 5%, 31 – was ranging from 1% to 5%, 47 – was less than 1%. They belonged to different groups including monoterpenoids, sesquiterpenoids, sesquiterpene alcohols, saturated unbranched hydrocarbons etc. The largest number of components were identified in the petals of R. roulettii (49) and R. centifolia (45), while the number of identified components in species R. multiflora (33), R. rugosa (31) and R. canina (30) was relatively smaller. All studied dog-rose species were characterized by the presence of such saturated unbranched hydrocarbons as decan, tetradecane, pentadecane, hexadecane, heptadecane, octadecane, nanodecane, heneicosane, tricosane, tetracosane, pentacosane, heptocosane, hentriacontane. Triterpene hydrocarbon squalene is the important component of the VOS complex in dog-rose pen tals. b-phenylethyl alcohol to be one of the main components of the rose essential oils was found during investigation in the following four dog-rose species as R. centifolia (0.61%), R. pimpinellifolia (3.56%), R. rugosa (4.24%), and R. multiflora (5.43%). Significant content of dihydro-b-ionol (18.46%), dihydro-b-ionone (0.69%), thiaspiran A (1.35%), and thiaspiran B (2.17%) were revealed in the petals of R. roulettii, which also have an influence on the aromatic bouquet of the rose oil. Conclusions. For the first time, 105 volatile organic substances were found in the petals of six species of dog-rose, 11 of which were not identified. It was found out that the largest number of components were identified in the petals of R. roulettii (49) and R. centifolia (45). The number of VOS identified in species R. multiflora (33), R. rugosa (31), and R. canina (30) was relatively smaller. Component composition of VOS in the petals of studied dog-rose species is an important constituent in breeding investigations of prospective producers for the various purposes of use.

Мета. Серед описаних у фаховій літературі ідентифікованих генів стійкості проти збудника бурої іржі виділити чужорідні, інтрогресовані у вид Triticum aestivum L. джерела, встановити їх походження та перспективи використання в селекційній практиці. Результати. Пшениця озима м’яка як основна зернова культура належить до групи рослин, яких найдавніше вирощують у контрольованих умовах. Протягом періоду вегетації вона зазнає згубного впливу збудників хвороб, тому пошук джерел стійкості проти них є першочерговим завданням селекції. Бура іржа – одна з найпоширеніших і шкодочинних хвороб пшениці. Вона призводить до значних втрат урожаю та погіршення якості зерна. Популяція збудника Puccinia reconditа вирізняється неабиякою адаптивною здатністю. Висока варіабельність вірулентності гриба призводить до накопичення патотипів, здатних долати гени стійкості пшениці. Найбільш екологічно безпечним методом контролювання захворювання є створення стійких сортів. Ефективність селекції на стійкість проти бурої іржі можна покращити, використовуючи різні Lr-гени стійкості. На цей час у геному пшениці та її родичів ідентифіковано й охарактеризовано за хромосомною локалізацією та ефективністю понад 90 (Lr) генів стійкості проти цього збудника. Виявлено, що майже всі ефективні на території України гени стійкості проти збудника бурої іржі, окрім Lr10 та Lr23, є чужорідними, перенесеними в Triticum aestivum від інших видів: Aegilops speltoides – гени Lr28, Lr35, Lr36, Lr47, Lr51, Lr66; Aegilops tauschii – Lr1, Lr21, Lr22а, Lr32, Lr39, Lr42; Triticum timopheevii – Lr18 та Lr50; Thinopyrum elongatum – Lr19, Lr29, Lr24; Secale cereale – Lr25, Lr26 та Lr45; Aegilops umbellulata – Lr9, Lr76; Triticum speltа – Lr44, Lr65, Lr71; Triticum dicoccoides – Lr53, Lr64; Aegilops triuncialis – Lr58, LrTr; Tr. timopheevii spp. viticulosum – LrTt1; Aegilops ventricosa – Lr37; Aegilops kotschyi – Lr54; Elymus trachycaulis – Lr55; Aegilops sharonensis – Lr56; Aegilops geniculate – Lr57; Aegilops peregrine – Lr59; Triticum turgidum – Lr61; Aegilops neglecta – Lr62; Triticum monococcum – Lr63. Висновки. Залучення до схрещувань культурних та диких видів родичів пшениці дасть змогу отримати неоднорідний за стійкістю проти збудника бурої іржі селекційний матеріал.

Purpose. Reporting peculiarities of new forms of edible corn and prospects for their use in food production that have been created jointly by scientists from the Plant Production Institute nd. a. V. Ya. Yuryev of NAAS and Ustymivka Experimental Station for Plant Production on the base of the environmentally friendly method that uses biochemical effects of natural mutations of corn grain endosperm structure. Methods. Field study, laboratory analysis, mathematical and statistical evaluation. Results. Original series of inbred lines as well as promising varieties and hybrids of sugar and waxy corns were obtained. Varieties and hybrids are universally usable for obtaining both fresh and canned products, characterized by good taste, provide yields of up to 10 tons per hectare of commercial products and aimed to grow in various soil and climatic conditions. Brief economic and morphological characteristics of corn varieties and hybrids created jointly by scientists from the Plant Production Institute nd. a. V. Ya. Yuryev of NAAS and Ustymivka Experimental Station for Plant Production were given. Conclusions. The use of biochemical effect of natural mutations of endosperm structure is cost-effective and environmentally friendly method for creating lines, varieties and hybrids of corn for direct consumption and processing into food products. ‘Biliavka’, ‘Nika’ varieties and ‘Binom F1’, ‘Solo F1’, ‘Twister F1’ hybrids created with the help of this method are promising for commercial use

Purpose. To create a multiplex system for identification glyphosate-tolerant sugar beet by using PCR. Methods. Molecular genetic analysis. Results. The article presents the results of studies to determine the parameters of the polymerase chain reaction (PCR) in order to develop a multiplex system for identification of the structural elements of the design of transgenic gene cp 4 epsps, which provides tolerance to glyphosate. For amplicon target DNA sequences, the following values of temperature conditions of PCR were determined: step 1 (initial denaturation) 95 °C – 3 min; step 2 (specific reaction products accumulation): denaturation 95 °C – 45 s; hybridization of primers 55 °C – 50 s; elongation 72 °C – 1 min; number of cycles – 40; step 3 (final elongation) 72 °C – 6 min. A series of PCR were carried out for the purpose of selecting the optimal amount of DNA matrix for efficient estimate of transgenic sugar beet plants for the presence of specific sequences. Conclusions. To identify transgenic glyphosate-tolerant sugar beet, it is advisable to determine 35S promoter and gene cp 4 epsps in individual genotypes. It was found that during the selection of temperature parameters of multiplex reaction a 5 °C rise in primer hybridization temperature did not affect the identification of gene als that allowed to include specific primers for determination of this sequence as an internal control. Based on the results of test multiplex reactions, concentrations of dNTPs and Mg2+ ions were determined that allowed to exclude the possibility of non-specific fragments and false-negative results. The optimum amount of matrix DNA (100–150 ng) for an efficient estimate of transgenic sugar beet plants for the presence of specific sequences was determined. Obtained results allowed to develop a multiplex test system for identification of transgenic glyphosate-tolerant sugar beet which can be used for simultaneous determination of the 35S promoter, cp 4 epsps gene and als gene as an internal reaction control.
