Methodical aspects of assessing grape breeding material Full text

Purpose. Developing methodical aspects of assessing grape breeding material in hybrid combinations for a number of indicators of adaptability and productivity. Methods. Breeding, hybridological, statistical ones. Results. Main requirements were determined for a new generation of table and technical grapes that was based on the set breeding task and patent searches, literary sources and Internet resources. 12 main indicators of adaptability, productivity and quality of products were analyzed, a high level of their display is required for pre-selection of grape seedlings for further study. Gradation from the lowest level of display of the above economic characters of grapes to the medium and high ones was developed. For their definition points-based system is used – the lowest level of display corresponds to one and three points, average – five, and high – seven and nine. As a result, studied plants can score 12 to 108 points. Variation in the plant evaluation over the years should not exceed 10 points. A seedling cannot be selected by the maximum level of displaying only one parameter, in the same manner as it shall be rejected with a minimum level of display of any of the said indicators. Seedlings that display a stable middle level of traits can be placed to the breeding nursery to study the level of display of the essential parameters and properties of the grafted culture. Conclusions. Criteria and basic parameters of genotypic values in hybrid combinations were determined. Plants that have been estimated in 60 points or higher may be considered as promising ones, that is those which require further studying. Valuable genotypes correspond to the level of 80–100, very valuable – of 101–108 points.

Purpose. Theoretical and experimental substantiation of introduction of “Romantica” roses originated by Meilland company under the conditions of the Right-Bank Forest-Steppe Zone of Ukraine. Methods. Biometrical, comparative-morphological, statistic ones. Results. Viability, growth and development indices as well as ornamental qualities (tolerance for winter conditions, frost hardiness, drought resistance, complex disease and pest resistance, habit maintenance, shoot formation, flowering, coloration, shape of flowers, petal number etc.) were analyzed for 12 rose cultivars of “Romantica” series planted on the collection and exhibition plots of the National Dendrological Park «Sofiivka» NAS of Ukraine. The investigations were conducted within 2012–2015. The most of the cultivars got the mark of introduction success 41–45 points of 50; so, the cultivars are totally promising for introduction into the conditions of the Right-Bank Forest-Steppe Zone of Ukraine. ‘Leonardo da Vinci’ and ‘Yves Piaget’ cultivars got point number 35–38 and they can be considered promising ones. The acclimatization number for the most of the investigated cultivars was 85–90 points of 100 that means good adaptation of the plants in the conditions of the Right-Bank Forest-Steppe Zone of Ukraine. The adaptation of cv. ‘Leonardo da Vinci’ and cv. ‘Yves Piaget’ roses (point number 75) can be considered satisfactory. The total evaluation of “Romantica” roses ornamental qualities made 76–87 points of 100; so, all the cultivars display high ornamental qualities and deserve to be introduced to the production. Conclusions. The garden roses of “Romantica” series originated by Meilland company being introduced into the conditions of the Right-Bank Forest-Steppe Zone of Ukraine are adapted well in the introduction region and they are promising for the subsequent introduction.

Adaptive capacity and breeding value of spring triticale varieties and lines was established. Methods of breeding and parentage of the best samples were analyzed. Comparison of yields under conditions in of different years of growing allowed to identify genotypes with high yield potential – ‘ЯТХ 38-14’, ‘ЯТХ 61-14’, ‘ЯТХ 62-14’, with stable yields – ‘ЯТХ 17-14’, ‘Boryviter kharkivskyi’, ‘ЯТХ 43-14’, ‘Gusar kharkivskyi’, drought resistance ones – ‘ЯТХ 37-14’, ‘ЯТХ 43-14’, ‘ЯТХ-64-14’. The genotypes ‘Lebіd kharkіvskyi’, ‘ЯТХ 38-14’, ‘ЯТХ 62-14’ appeared to be more plastic. These samples may be used of hybridization to improve yield. Conclusions. The most promising to production and breeding are the lines ‘ЯТХ 17-14’, ‘ЯТХ 64-14’, ‘ЯТХ 37-14’, ‘ЯТХ 43-14’ (their average yield 4,27–4,48 t/ha) and varieties ‘Zlit kharkivskyi’, ‘Boryviter kharkivskyi’, ‘Gusar kharkivskyi’, ‘Darkhliba kharkivskyi’, which have high adaptive capacity combined with high productivity. They are valuable for use as a starting material in breeding for adaptability.

Purpose. To define the role of fertilizers containing amino acids in the nutrition systems of highly productive varieties of winter wheat. Methods. Field studies, biochemical technique, analytical procedure, statistical evaluation. Results. In the process of investigations, sufficiently high activity of fertilizers containing amino acids was established when applying them for leaf-feeding. Fertilizers based on algae (Megafol, Megafol Protein, Terra-Sorb foliar) and animal (Izabion) hydrolysates appeared to be very effective for increasing yield of culture. It was found that in case of low doses of mineral nitrogen, fertilizers helped to increase both the yield and quality indicators of high-yielding wheat variety. Conclusions. It was defined that fertilizers that include amino acids are highly effective compositions containing plant-available nitrogen in organic form and can be promising for application as antistress agents and improving efficiency of macro- and microelements use by cultivated plants. They are essential components of modern technologies for cultivation of highly productive varieties and hybrids in crop production of Ukraine.

Purpose. Studying and identifying winter wheat varieties that are resistant to causal agents of major diseases and pests. Methods. Laboratory analysis, field study. Results. On artificial infection backgrounds of causal agents such varieties as ‘Smuhlianka’, ‘Svitanok Myronivskyi’, ‘Berehynia Myronіvska’, ‘Horlytsia Myronіvska’ have shown high level of resistance to brown rust; ‘Svitanok Myronivskyi’, ‘Berehynia Myronіvska’ – to powdery mildew; ‘Smuhlianka’ – to covered smut. Varieties ‘Voloshkova’, ‘Yuviliar Myronivskyi’, ‘Myrliena’, ‘Oberih Myronivskyi’, ‘Kolos Myronivschyny’, ‘Lehenda Myronivska’ had medium resistance to Septoria leaf blotch; ‘Smuhlianka’, ‘Myrliena’, ‘Oberih Myronivskyi’, ‘Berehynia Myronіvska’, ‘Horlytsia Myronіvska’, ‘Myronivska storichna’ – to Fusarium head blight; ‘Myronіvska 65’, ‘Smuhlianka’, ‘Lehenda Myronivska’, ‘Berehynia Myronіvska’ – to root rots. Among the varieties studied, there were those with group resistance to diseases: ‘Voloshkova’, ‘Myrliena’, ‘Yuviliar Myronivskyi’, ‘Oberih Myronivskyi’, ‘Bohdana’, ‘Myronivska storichna’, ‘Ekonomka’, ‘Svitanok Myronivskyi’, ‘Berehynia Myronіvska’, ‘Horlytsia Myronіvska’, ‘Smuhlianka’. Varieties bred at the V. M. Remeslo Myronivka Institute of Wheat are distinguished by pest resistance. During autumn tillering phase of winter wheat the smallest number of large cereal aphids was observed in varieties ‘Smuhlianka’ and ‘Myronivska storichna’, leafhoppers – in varieties ‘Smuhlianka’, ‘Myrliena’, ‘Yuviliar Myronivskyi’. The slight population of thrips in the phase of earing was marked in the variety ‘Kolos Myronivschyny’, in the milk-ripe stage the smallest number of larvae per ear was detected in varieties ‘Smuhlianka’, ‘Voloshkova’, ‘Kolos Myronivschyny’. Varieties ‘Lehenda Myronivska’ and ‘Smuhlianka’ were proved to be resistant to damages of stems by sawflies. It should be noted that ‘Smuhlianka’ variety shows resistance to aphids, leafhoppers and grain sawflies. Conclusions. It was established that winter wheat varieties bred at the V. M. Remeslo Myronivka Instutite of Wheat are characterized by resistance to certain causal agents, some of them – to their group. Varieties that are resistant to the most common and harmful phytophages were identified.

Purpose. To develop an algorithm of creation of new highly productive clonal varieties of Salicaceae family representatives with improved agronomic characters that can be used at energy plantations Methods. Field investigations, laboratory analysis, analytical approach, selection method. Results. Flow chart of selection process with representatives of Salicaceae family included the following stages: the Ist stage – creation of primary families (F0); the IInd stage – creation of the second families, or branches; the 222rd stage – interfamily (individually-familial) selection; IVth stage – familial-group selection; the Vth stage – clonal selection (selection of ramets). Conclusions. Breeding process with the use of complex of advanced methods of selection (individual, familial, group and clonal ones) allows to get model populations with high-frequency of valuable alleles and genotypes and with the high level of genetic variety. Due to a negative multi-stage individual intrafamilial and interfamily selection, cleaning of artificial populations from undesirable forms takes place without the substantial decrease of genetic variety level.

Purpose. To analyse biochemical and economic characters of new tomato varieties adapted to the conditions of the south of Ukraine. Methods. Field study, laboratory testing, statistical analysis. Results. Intraspecific hybridization method followed by individual selection was used to create such new tomato varieties as ‘Naddnіprianskyi 1’, ‘Inguletskyi’, ‘Kimmeriiets’, ‘Sarmat’, ‘Taim’, ‘Legin’, ‘Kumach’ adapted to the conditions of the south, which are characterized by high yielding (69,2–78,0 t/ha), fruit quality (content of soluble dry matter 5,65–5,92%), drought (7,2–8,1 points) and heat (70,4–88,2%) resistance. All varieties are of intensive type, they require advanced agricultural technology, irrigation. Conclusions. New tomato varieties, namely ‘Naddnіprianskyi 1’, ‘Inguletskyi’, ‘Kimmeriiets’, ‘Sarmat’, ‘Taim’, ‘Legin’, ‘Kumach’ were entered in the State register of plant varieties suitable for dissemination in Ukraine. They are recommended for outdoor growing in the Steppe and Forest-Steppe zones of Ukraine.

Purpose. Comparative studies of the photosynthetic apparatus of winter wheat varieties of modern and earlier breeding in the relation to their productivity. Methods. Field, pot experiment, biometrical, gas analysis, analytical and statistical ones. Results. It is found that the modern high-yielding winter wheat varieties have a higher rate of flag leaf photosynthesis during generative period of development, particularly at grain filling period, improved ability to store assimilates in stem and effectively use them later for grain growth. The modern varieties form crop canopy with greater leaf area and chlorophyll indexes and larger photosynthetic capacity and maintain functional activity of photosynthetic apparatus longer at the end of the growing season. Conclusions. The superiority for grain productivity of modern high-yielding wheat varieties, originated from the Institute of Plant Physiology and Genetics, NAS of Ukraine, over varieties released before the “green revolution”, results from increased capacity and durability of functioning the crop photosynthetic apparatus, higher CO2 assimilation rate and optimization of source-sink relations in whole plant.

Purpose. Investigation of maize ae1 gene polymorphism by bioinformatic methods. Methods. Global and local alignment of the nucleotide and amino acid sequences, in silico translation and transcription, translates modeling, primers design, phylogenetic analysis. Results. 255 nucleotide sequences of maize аe1 gene, 500 amino acid sequences of homology translates of maize ae1 gene (SBEIIb enzyme homologs) and 100 mRNA expressed from the maize ae1 gene were analyzed to establish phylogenetic relationships. Polymorphism of maize ae1 gene different regions was investigated by bioinformatic methods. Modeling of the maize enzyme SBEIIb was performed. Conclusions. According to the results of amino acid sequences of SBEIIb enzyme homologs alignment, it was found that ae1 gene orthologs are present only in monocots, paralogs – in monocots, dicots, and other taxa, including algae and animals. Based on the results of alignment of plants mRNA from which enzyme SBEIIb is translated, maize ae1 gene orthologs and the nearest paralogs encoding starch branching enzymes with chloroplast localization were defined; this suggests a possible origin of ae1 gene due to duplication of the gene encoding the 1,4-alpha-glucan-branching enzyme 2 with chloroplast or amyloplast localization. For the polymorphic sites design primers were developed that allowed to differentiate the maize lines. It was determined that the detection of polymorphism in theory can influence the enzyme function and, as a result, change the concentration of amylopectin in maize grain.

Purpose. Investigation of maize ae1 gene polymorphism by bioinformatic methods. Methods. Global and local alignment of the nucleotide and amino acid sequences, in silico translation and transcription, translates modeling, primers design, phylogenetic analysis. Results. 255 nucleotide sequences of maize аe1 gene, 500 amino acid sequences of homology translates of maize ae1 gene (SBEIIb enzyme homologs) and 100 mRNA expressed from the maize ae1 gene were analyzed to establish phylogenetic relationships. Polymorphism of maize ae1 gene different regions was investigated by bioinformatic methods. Modeling of the maize enzyme SBEIIb was performed. Conclusions. According to the results of amino acid sequences of SBEIIb enzyme homologs alignment, it was found that ae1 gene orthologs are present only in monocots, paralogs – in monocots, dicots, and other taxa, including algae and animals. Based on the results of alignment of plants mRNA from which enzyme SBEIIb is translated, maize ae1 gene orthologs and the nearest paralogs encoding starch branching enzymes with chloroplast localization were defined; this suggests a possible origin of ae1 gene due to duplication of the gene encoding the 1,4-alpha-glucan-branching enzyme 2 with chloroplast or amyloplast localization. Purpose. Improvement of a range of strawberry varieties based on comprehensive study and introduction of new Ukrainian and West European cultivars. Methods. Cultivars and elite forms were studied using the methods of primary variety studying and state variety testing at three-year strawberry plantations of the Institute of Horticulture of the National Academy of Sciences of Ukraine (the city of Kyiv) and its variety testing stations. Results. It was found that more frequent spring and summer droughts as well as black frosts in winter adversely affected the strawberry productivity. The best varieties were selected among new ones for the complex of traits, they included ‘Elegance’ from the Great Britain, ‘Gercules’ from Ukraine (both are dessert grapes), ‘Elsanta’ from Holland (for universal use), ‘Alba’ from Italia (as raw material). The highly adaptive and productive ‘Istochnik’ and ‘Festyvalna romashka’ varieties (as raw material) are recommended for the north regions. Conclusions. In the context of profound climatic change, high-profitable cultivation of mid-ripening varieties and a strawberry as such in the Northern Forest-Steppe zone is expedient under controlled conditions – in high film tunnels, in hot-houses and under perennial irrigation that already has become the European tendency.
