Several studies have explored the utilization of soil microorganisms, to address the environmental issues associated with glyphosate use and enhance crop yields. In our investigation, screening on Agar plate and broth medium  Luria Bertani was carried out after isolating bacterial strains from rhizospheric agricultural soil in Mascara,  Algeria, to biodegrade glyphosate, following that by testing the Plant Growth-Promoting Rhizobacteria and evaluate the effects of glyphosate on these proprieties. Our findings indicate that five bacterial strains exhibited growth in the presence of glyphosate concentrations up to 25 mg/ml, beyond this concentration the strains have developed tolerance. Following a partial examination of the 16S rRNA sequences, the bacterial strains were identified as belonging to the genus of Enterobacter. After 10 days of incubation with the glyphosate, Phosphate solubilization decreased in broth and agar Pikovskaya medium and the bacterial strains synthetized less of indole-3-acetic acid compared to the control, indicating the impact of glyphosate on these outcomes, high concentration of glyphosate inhibited nitrogen fixation, and various doses of glyphosate were found to restrict the growth of biofilms in these strains. The results of HPLC examination of secondary metabolites revealed that the primary degradation products of glyphosate in all strains were Sarcosine and Glycine. So, it seemed that the strain could both biodegrade glyphosate and use it for growth ,while also possessing rhizobacteria properties that promote plant development, enabling the use of the strains in the bioremediation of glyphosate-contaminated soils.

In this study, a strain of lactic acid bacteria Streptococcus salivarius was studied for its capacity to remove hexavalent chromium (Cr (VI)) from a liquid medium and to form biofilm. Both properties are useful for using the strain in bioremediation of metal-contaminated effluents. For biofilm formation capacity, three methods were used: the tube method (TM), the Congo red agar method (CRA) and adherence to polystyrene tissue culture plate method (TCP). S. salivarius, showed a positive-biofilm and a correlation between the three methods was noted. The bacterial surface hydrophobicity was studied using the microbial adhesion to solvents method (MATS). On AISI-316 L stainless steel, the strain with a hydrophobic surface showed a good adhesion on this support after 18 h incubation. The colonization of the supports and the biofilms formation by the bacterial cell was observed using scanning electron microscopy (SEM). The minimum inhibitory concentration (MIC) of Cr(VI) on S. salivarius was determined on MRS broth, it was relatively high and equal to 400mg/l. In addition, it displayed a remarkable capacity to reduce Cr(VI) concentration on the liquid medium containing initially 50 mg/l of Cr(VI) ; the percent removal rate was equal to approximately 42% after 72 h of incubation at 37 °C. In addition to its GRAS status, the obtained results suggested that S. salivarius could be successfully used in Cr(VI) bioremediation.

Better understanding the plastisphere is crucial as it has self-sufficient orga- nization and carries pathogens or organisms that may be involved in the pollutant adsorption and/or plastic degradation. To date, the plastisphere is mainly described at the taxonomic level and the functioning of its microbial communities still remains poorly documented. In this work, metagenomic and metaproteomic analyzes were performed on the plastisphere of polypropylene and polyethylene plastic debris sampled on a pebble beach from the Mediterranean Sea. Our results confirmed that the plastisphere was organized as self-sufficient eco- systems containing highly active primary producers, heterotrophs and predators such as nematode. Interestingly, the chemical composition of the polymer did not impact the structure of the microbial communities but rather influenced the functions expressed. Despite the fact that the presence of hydrocarbon-degrading bacteria was observed in the metagenomes, polymer degradation metabolisms were not detected at the protein level. Finally, hydrocarbon degrader (i.e., Alcanivorax) and pathogenic bacteria (i.e., Vibrionaceae) were observed in the plas- tispheres but were not very active as no proteins involved in polymer degradation or pathogeny were detected. This work brings new insights into the functioning of the microbial plastisphere developed on plastic marine debris.

[Departement_IRSTEA]Eaux [TR1_IRSTEA]BELCA | This study is a first attempt to investigate the impact of urban contamination on metal tolerance of heterotrophic river biofilms using a short-term test based on β-glucosidase activity. Tolerance levels to Cu, Cd, Zn, Ni and Pb were evaluated for biofilms collected at three sites along an urban gradient in the Seine river (France). Metallic pollution increased along the river, but concentrations remained low compared to environmental quality standards. Biofilm metal tolerance increased downstream from the urban area. Multivariate analysis confirmed the correlation between tolerance and contamination and between multi-metallic and physico-chemical gradients. Therefore, tolerance levels have to be interpreted in relation to the whole chemical and physical characteristics and not solely metal exposure. We conclude that community tolerance is a sensitive biological response to urban pressure and that mixtures of contaminants at levels lower than quality standards might have a significant impact on periphytic communities.

The problematic of microplastics pollution in the marine environment is tightly linked to their colonization by a wide diversity of microorganisms, the so-called plastisphere. The composition of the plastisphere relies on a complex combination of multiple factors including the surrounding environment, the time of incubation along with the polymer type, making it difficult to understand how the biofilm evolves during the microplastic lifetime over the oceans. To better define bacterial community assembly processes on plastics, we performed a 5 months spatio-temporal survey of the plastisphere in an oyster farming area in the Bay of Brest (France). We deployed three types of plastic pellets in two positions in the foreshore and in the water column. Plastic-associated biofilm composition in all these conditions was monitored using 16 S rRNA metabarcoding and compared to free-living and attached bacterial members of seawater. We observed that bacterial families associated to plastic pellets were significantly distinct from the ones found in seawater, with a significant prevalence of filamentous Cyanobacteria on plastics. No convergence towards a unique plastisphere was detected between polymers exposed in the intertidal and subtidal area, emphasizing the central role of the surrounding environment on constantly shaping the plastisphere community diversity. However, we could define a bulk of early-colonizers of marine biofilms such as Alteromonas, Pseudoalteromonas or Vibrio. These early-colonizers could reach high abundances in floating microplastics collected in field-sampling studies, suggesting the plastic-associated biofilms could remain at early development stages across large oceanic scales. Our study raises the hypothesis that most members of the plastisphere, including putative pathogens, could result of opportunistic colonization processes and unlikely long-term transport.

Microplastics and antibiotics widely coexist in the aquatic environment, especially in mariculture regions. However, antibiotics adsorbed on microplastics and their role in the colonization of microorganisms on microplastics are poorly understood. Therefore, in-situ aging experiments were conducted to investigate the impact of antibiotics and microplastics co-occurrence on microorganisms and assess their potential risks to human health. Results showed that antibiotics were adsorbed selectively on microplastics, with 29 investigated antibiotics (n = 40) detected in surrounding water but only 6 investigated antibiotics were adsorbed on microplastics. The concentration of antibiotics accumulated on microplastics was controlled by microplastic types and environmental conditions. For example, aged polypropylene (PP) had more developed pore structures resulting in higher adsorption of antibiotics than other microplastic types. High-throughput sequencing showed higher diversity and distinct composition of microorganisms attached to the microplastics than the surrounding water. Opportunistic pathogenic bacteria such as Mycobacterium possessed positive relationships with tetracycline and doxycycline on aged microplastics, which showed adsorbed antibiotics on aged microplastics could benefit some specific pathogens colonized on the microplastics and spread into unaffected ecosystems, marine organisms even humans. The health risk quotient (HQ) implied the potential human health risk of consuming commercial seafood polluted by antibiotics and microplastic loaded with antibiotics. This study revealed the interaction of antibiotics and microorganisms with aged microplastics in aquaculture systems, providing a novel insight into their synergistic effects on ecological and human health.

Titanium dioxide nanoparticles (TiO₂ NPs) easily combine with other pollutants such as heavy metals because of their excellent physiochemical properties. However, how such an interaction may affect the binding behavior of metals onto biofilms remains largely unclear. This study, examined the effects of TiO₂ NPs on Cd²⁺ accumulation and toxicity for natural periphytic biofilms were examined. The adsorption kinetics showed that adding 0.1 and 1 mg/L TiO₂–NPs increased the Cd²⁺ adsorption of biofilms at equilibrium by 23.5% and 35.8%, respectively. However, adding 10 mg/L TiO₂ NPs increased the Cd²⁺ adsorption of biofilms at equilibrium by only 1.9%. The adsorption isotherms indicate that the presence of TiO₂ NPs considerably increased the Cd²⁺ adsorption capacity of the biofilms; however, this effect became less prominent at high TiO₂ NP concentrations. The optimum pH for Cd²⁺ adsorption increased with increasing Cd²⁺ and TiO₂ NP contents. At low concentrations, the coexistence of Cd²⁺ and TiO₂ NPs may facilitate their respective accumulation by stimulating the secretion of extracellular polymeric substances and enhancing the microbial activity of the biofilm. The presence of TiO₂ NPs increases the surface binding energy between Cd²⁺ and functional groups such as carboxyl groups, enhancing the Cd²⁺ accumulation on the biofilm.
