Pacific Ocean trawl samples, stomach contents of laboratory-raised fish as well as fish from the subtropical gyres were analyzed by Raman micro-spectroscopy (RMS) to identify polymer residues and any detectable persistent organic pollutants (POP). The goal was to access specific molecular information at the individual particle level in order to identify polymer debris in the natural environment. The identification process was aided by a laboratory generated automated fluorescence removal algorithm. Pacific Ocean trawl samples of plastic debris associated with fish collection sites were analyzed to determine the types of polymers commonly present. Subsequently, stomach contents of fish from these locations were analyzed for ingested polymer debris. Extraction of polymer debris from fish stomach using KOH versus ultrapure water were evaluated to determine the optimal method of extraction. Pulsed ultrasonic extraction in ultrapure water was determined to be the method of choice for extraction with minimal chemical intrusion. The Pacific Ocean trawl samples yielded primarily polyethylene (PE) and polypropylene (PP) particles >1 mm, PE being the most prevalent type. Additional microplastic residues (1 mm - 10 μm) extracted by filtration, included a polystyrene (PS) particle in addition to PE and PP. Flame retardant, deca-BDE was tentatively identified on some of the PP trawl particles. Polymer residues were also extracted from the stomachs of Atlantic and Pacific Ocean fish. Two types of polymer related debris were identified in the Atlantic Ocean fish: (1) polymer fragments and (2) fragments with combined polymer and fatty acid signatures. In terms of polymer fragments, only PE and PP were detected in the fish stomachs from both locations. A variety of particles were extracted from oceanic fish as potential plastic pieces based on optical examination. However, subsequent RMS examination identified them as various non-plastic fragments, highlighting the importance of chemical analysis in distinguishing between polymer and non-polymer residues.

This study incorporated stable isotope analyses with chemical analyses to determine the origin and migration of sulfur sources in East Asia, and these findings were compared with our decadal research from 2000 to 2001 and 2002 to 2003. The multiple sulfur isotope composition (32S, 33S and 34S) of the dissolved sulfate in precipitation was first measured from 2011 to 2013 in Seoul, South Korea. The δ34Snss values were −1.1‰ to 7.9‰ (avg. 3.6‰), strongly suggesting that sulfur derived from the combustion of Chinese coal is the predominant source of sulfate in the Seoul region. Low NO3/SO42− ratios in the precipitation samples indicated an insignificant effect of sulfur from vehicle exhaust. The seasonal variation of δ34Snss values appears to be caused by increasing biogenic sulfur activity during the spring and summer seasons. The some Δ33S values (0.13‰–0.16‰) measured in the three samples were sufficiently small; thus, whether these values can be attributed to mass-independent fractionation remains unclear. Measuring the Δ33S anomalies in dissolved sulfate provides valuable insights for identifying the sources of sulfur transferred from the stratosphere to the troposphere and upper troposphere.

As a pristine continent, Antarctica provides a good opportunity to study the spatial transport and temporal accumulation of environmental contaminants and the impacts of anthropogenic activities, both of which have given rise to ongoing public concern. In this research, an approach of coupling aquatic time-integrated passive sampling with chemical analysis and bioassays was used to assess pollution by hydrophobic organic contaminants in Antarctic waters. Passive samplers were deployed in waters of Fildes Peninsula, Antarctica, and their extracts were used for chemical analyses of sixty-six hydrophobic organic contaminants belonging to five groups [organophosphorus flame retardants (PFRs), phthalic acid esters (PAEs), polycyclic aromatic hydrocarbons (PAHs), organochlorine pesticides (OCPs), and polychlorinated biphenyls (PCBs)] and in vitro bioassays for endocrine disruption and genotoxicity. In total, twenty pollutants (six PFRs, one PAE, two PAHs, six OCPs, and five PCBs) were quantified, and six PFRs had concentrations that ranged from ND (not detected) to 44.37 ng L−1 in Antarctic waters. The concentrations detected in the waters were generally low and insufficient to have significant in vitro endocrine disruption potential or genotoxicity. The source and transport pathways of PFRs and PAE in Fildes Peninsula were studied, and multiple local sources (wastewater, air traffic, research stations, and animal feces) for different PFRs were proposed. A spatial and temporal analysis showed slight changes in the exposure of OCPs and PCBs in Antarctic waters. Furthermore, a comparison among a variety of Antarctic water sampling cases revealed that passive sampling can be a tool for aquatic time-integrated investigations in polar regions.

Coastal areas are the transition zones between ocean and land where intracellular antibiotic resistance genes (iARGs) and extracellular antibiotic resistance genes (eARGs) could spread among marine organisms, and between humans and marine organisms. However, little attention has been paid to the combined research on iARGs and eARGs in marine environment. In this context, we collected water and sediment samples from the coastal areas of the Bohai Bay in China and performed molecular and chemical analyses. The results of quantitative real-time PCR (qPCR) showed that the relative abundance of eARGs was up to 4.3 ± 1.3 × 10−1 gene copies/16S rRNA copies in the water samples and 2.6 ± 0.3 × 10−3 gene copies/16S rRNA copies in the sediment samples. Also, the abundance of eARGs was significantly higher than that of iARGs. Furthermore, the average abundances of antibiotic resistance genes (ARGs, include iARGs and eARGs) were the highest in both the water and sediment samples from the estuaries. The results of liquid chromatography-mass spectrometry showed that the concentrations of antibiotics in estuaries and areas near the mariculture site were higher than that in the other sites. The class 1 integron gene (int1) and sul1 in the intracellular DNA were significantly correlated in the water samples. Moreover, significant correlation between int1 and sul2 in the extracellular DNA was also found in the sediment samples. The combination of sulfamerazine and tetracycline as well as the combination of sulfamethazine and dissolved oxygen can both explain the abundance of ARGs, implying the combined effects of multiple stresses on ARGs.

Genotoxic compounds, as common contaminants of the air environment, are of interest in air pollution monitoring. There are several methods to determine the level of these contaminants in different localities, many of which may be difficult to access with the use of conventional active and passive samplers. In the present study, the needles Pinus mugo Turra and Picea abies were used to monitor sampling localities in Austria, Slovakia, and the Czech Republic. Needles were extracted and chemical analysis and the genotoxicity bioassay SOS chromotest were used to obtain complex information about the chemical mixture of pollutants present and their genotoxic effects. The SOS chromotest method was optimized by using a CPRG chromogenic substrate to reduce the false positive genotoxic effect of needle extracts. Pinus mugo Turra and Picea abies were identified as suitable passive sampling matrices for long-term air monitoring using the same plants sampled at the same time. The presented study brings an innovative method for the fast screening and identification of localities loaded by genotoxic active air contaminants.

Drinking water safety is continuously threatened by the emergence of numerous toxic organic pollutants (TOPs) in environmental waters. In this study, an approach integrating in vitro bioassays and chemical analyses was performed to explore toxicological profiles of representative source and drinking water from waterworks of the Yangtze River (Yz), Taihu Lake (Th), and the Huaihe River (Hh) basins in eastern China. Overall, 34 of 96 TOPs were detected in all water samples, with higher concentrations in both source and drinking water samples of Hh, and pollutant profiles also differed across different river basins. Non-specific bioassays indicated that source water samples of Hh waterworks showed higher genotoxicity and mutagenicity than samples of Yz and Th. An EROD assay demonstrated dioxin-like toxicity which was detected in 5 of 7 source water samples, with toxin concentration levels ranging from 62.40 to 115.51 picograms TCDD equivalents per liter of water (eq./L). PAHs and PCBs were not the main contributors to observed dioxin-like toxicity in detected samples. All source water samples induced estrogenic activities of 8.00–129.00 nanograms 17β-estradiol eq./L, and estrogens, including 17α-ethinylestradiol and estriol, contributed 40.38–84.15% of the observed activities in examined samples. While drinking water treatments efficiently removed TOPs and their toxic effects, and estrogenic activity was still observed in drinking water samples of Hh. Altogether, this study indicated that the representative source water in eastern China, especially that found in Hh, may negatively affect human health, a finding that demonstrates an urgent requirement for advanced drinking water treatments.

The study addresses the topic of suitable matrices for chemical analysis in fish monitoring and discusses the effects of data normalization in the context of the European Water Framework Directive (WFD). Differences between species are considered by comparing three frequently monitored species of different trophic levels, i.e., chub (Squalius cephalus, n = 28), (bream, Abramis brama, n = 11), and perch (Perca fluviatilis, n = 19) sampled in the German Danube. The WFD priority substances dioxins, furans and dioxin-like polychlorinated biphenyls (PCDD/F + dl-PCB), polybrominated diphenyl ethers (PBDE), α-hexabromocyclododecane (α-HBCDD), hexachlorobenzene (HCB), mercury (Hg), and perfluorooctane sulfonic acid (PFOS) as well as non-dioxin-like (ndl)-PCB were analyzed separately in fillet and carcass and whole body concentrations were calculated. Hg was analyzed in individual fish fillets and carcasses, all other substances were determined in pool samples, which were compiled on the basis of fish size (3 chub pools, 1 bream pool, 2 perch pools). The data were normalized to 5% lipid weight (or 26% dry mass in the case of Hg and PFOS) for comparison between matrices and species.Hg concentrations were generally higher in fillet than in whole fish (mean whole fish-to-fillet ratio: 0.7) whereas all other substances were mostly higher in whole fish. In the case of lipophilic substances these differences leveled after lipid normalization.Significant correlations (p ≤ .05) were detected between Hg and fish weight and age. Hg concentrations varied least among younger fish. PCDD/F, dl-PCB, ndl-PCB, PBDE, α-HBCDD and HCB correlated significantly (p ≤ .05) with lipid concentrations. Fillet-to-whole fish conversion equations and/or conversion factors were derived for all substances except α-HCBDD. Although more data also for individual fish would be desirable the results are nevertheless a step on the way to translate fillet concentrations of priority substances to whole fish concentrations.

Productive coastal and estuarine habitats can be degraded by contaminants including persistent organic pollutants (POPs) such as PCBs, dioxins, and organochlorine insecticides to the extent of official designation as contaminated sites. Top-predatory wildlife may continue to use such sites as the habitat often appears suitable, and thus bioaccumulate POPs and other contaminants with potential consequences on their health and fitness. Victoria and Esquimalt harbours are located on southern Vancouver Island, British Columbia (BC) and are federally designated contaminated sites due mainly to past heavy industrial activities, such as from shipyards and sawmills. We collected scat samples from river otters (Lontra canadensis) throughout an annual cycle, and combined chemical analysis with DNA genotyping to examine whether the harbour areas constituted a contaminant-induced ecological trap for otters. We confirmed spatial habitat use by radio telemetry of a subsample of otters. Fifteen percent of otter scat contained PCB concentrations exceeding levels considered to have adverse effects on the reproduction of mink (Neovison vison), and there were significant positive correlations between concentrations of PCBs and of thyroid (T3) and sex (progesterone) hormones in fecal samples. Radio telemetry data revealed that otters did not show directional movement away from the harbours, indicating their inability to recognize the contaminated site as a degraded habitat. However, analysis and modeling of the DNA genotyping data provided no evidence that the harbour otters formed a sink population and therefore were in an ecological trap. Despite the highly POP-contaminated habitat, river otters did not appear to be adversely impacted at the population level. Our study demonstrates the value of combining chemical and biological technologies with ecological theory to investigate practical conservation problems.

Forensic chemistry assessments documented the presence of Macondo (MC252) oil from the Deepwater Horizon (DWH) spill in offshore water samples collected under Natural Resource Damage Assessment (NRDA) protocols. In ocean depths, oiled water was sampled, observed, photographed, and tracked in dissolved oxygen (DO) and fluorometry profiles. Chemical analyses, sensor records, and observations confirmed the shifting, rising oil plume above the wellhead while smaller, less buoyant droplets were entrapped in a layer at ~1000–1400 m and advected up to 412 km southwest. Near-surface oil samples showed substantial dissolution weathering from oil droplets rising through the water column, as well as enhanced evaporative losses of lighter n-alkanes and aromatic hydrocarbons. Dispersant effects from surface applications and injected at the wellhead were seen in oil profiles as enhanced weathering patterns (increased dissolution), thus implying dispersants were a functionally effective mediation treatment. Forensic assessment methods are detailed in the Supplemental information (SI).

Chemical analyses and toxicity testing using six marine species were used to characterize the hazard of produced waters (PW) to marine life from twelve Australian offshore platforms. Hazard data were used in conjunction with platform-specific plume discharge dilution and species sensitivity distribution modeling to estimate cumulative risks by calculating the multiple substance potentially affected fraction of species in the local marine environment. Results provided two independent lines of evidence demonstrating that cumulative risks to marine life from these discharges meet intended 95% species protection goals at the edge of the mixing zone. A limited number of PW constituents (hydrocarbons, sulphide and ammonia) appeared to dictate risk thereby informing management and providing a rationale for more targeted analyses in future monitoring studies. Based on these findings a tiered framework is proposed to foster consistent screening and potential refinement of cumulative risk evaluations for PW discharges.
