In vitro and in vivo approaches for the measurement of oral bioavailability of lead (Pb) in contaminated soils: A review

We reviewed the published evidence of lead (Pb) contamination of urban soils, soil Pb risk to children through hand-to-mouth activity, reduction of soil Pb bioavailability due to soil amendments, and methods to assess bioaccessibility which correlate with bioavailability of soil Pb. Feeding tests have shown that urban soils may have much lower Pb bioavailability than previously assumed. Hence bioavailability of soil Pb is the important measure for protection of public health, not total soil Pb. Chemical extraction tests (Pb bioaccessibility) have been developed which are well correlated with the results of bioavailability tests; application of these tests can save money and time compared with feeding tests. Recent findings have revealed that fractional bioaccessibility (bioaccessible compared to total) of Pb in urban soils is only 5–10% of total soil Pb, far lower than the 60% as bioavailable as food-Pb presumed by U.S.-EPA (30% absolute bioavailability used in IEUBK model).

The feasibility of reducing children’s exposure to lead (Pb) polluted soil in New Orleans is tested. Childcare centers (median = 48 children) are often located in former residences. The extent of soil Pb was determined by selecting centers in both the core and outlying areas. The initial 558 mg/kg median soil Pb (range 14–3692 mg/kg) decreased to median 4.1 mg/kg (range 2.2–26.1 mg/kg) after intervention with geotextile covered by 15 cm of river alluvium. Pb loading decreased from a median of 4887 μg/m² (454 μg/ft²) range 603–56650 μg/m² (56–5263 μg/ft²) to a median of 398 μg/m² (37 μg/ft²) range 86–980 μg/m² (8–91 μg/ft²). Multi-Response Permutation Procedures indicate similar (P-values = 0.160–0.231) soil Pb at childcare centers compared to soil Pb of nearby residential communities. At ∼$100 per child, soil Pb and surface loading were reduced within hours, advancing an upstream intervention conceptualization about Pb exposure prevention.

We studied the source, concentration, spatial distribution and health risk of 16 polycyclic aromatic hydrocarbons (PAHs) in urban soils of Beijing. The total mass concentration of 16 PAHs ranged from 93 to 13 141 μg kg⁻¹ with a mean of 1228 μg kg⁻¹. The contour map of soil PAH concentrations showed that the industrial zone, the historical Hutong district and the university district of Beijing have significantly higher concentrations than those in remainder of the city. The results of sources identification suggested that the primary sources of PAHs were vehicle exhaust and coal combustion and the secondary source was the atmospheric deposition of long-range transported PAHs. The incremental lifetime cancer risks (ILCRs) of exposing to PAHs in the urban soils of Beijing for adult were 1.77 × 10⁻⁶ and 2.48 × 10⁻⁵, respectively under normal and extreme conditions. For child, they were 8.87 × 10⁻⁷ and 6.72 × 10⁻⁶, respectively under normal and extreme conditions.

A composted biosolid from wastewater treatment was added to soils of two public parks of Sevilla, and successive samples were taken during one year. In one of the parks, a second addition of biosolid was carried out after the first year. The soil contents in metals (pseudo-total) and their plant-available and oral bio-accessible fractions were significantly altered when the soils were amended with biosolid. Increase of the bio-accessible metal contents represents a deterioration of the environmental quality of recreational areas, where hand-to-mouth transfer of pollutants to children is likely to occur, although part of the metals added might be leached by rainfall or irrigation. The limits established in several countries for metal contents of soils in recreational areas are often exceeded after application of the biosolid. A careful study of the metal contents of recycled wastes is thus recommended before being used for green area maintenance.

The oral bioaccessibility and the human health risks of As, Hg and other metals (Cu, Pb, Zn, Ni, Co, Cd, Cr, Mn, V and Fe) in urban street dusts from different land use districts in Nanjing (a mega-city), China were investigated. Both the total contents and the oral bioaccessibility estimated by the Simple Bioaccessibility Extraction Test (SBET) of the studied elements varied with street dusts from different land use districts. Cd, Zn, Mn, Pb, Hg and As showed high bioaccessibility. SBET-extractable contents of elements were significantly correlated with their total contents and the dust properties (pH, organic matter contents). The carcinogenic risk probability for As and Cr to children and adults were under the acceptable level (<1 × 10⁻⁴). Hazard Quotient values for single elements and Hazard Index values for all studied elements suggested potential non-carcinogenic health risk to children, but not to adults.

The environmental pollution associated with mining and metallurgical activities reaches its greatest extent in several Andean cities and villages. Many locations in this area have accumulated through centuries a large amount of mining wastes, often disregarding the magnitude of this situation. However, in these naturally mineralized regions, there is little information available stating the exact role of mining and metallurgical industries in urban pollution. In this study, we demonstrated that the various metallic elements present in indoor dust (As, Cd, Cu, Pb, Sb, Sn, Zn) had a common origin and this contamination was increased by the proximity to the mines. Lead dust concentration was found at concerning levels for public health. In addition, wrong behaviors such as carrying mining workwear home contributed to this indoor dust pollution. Consequently, the constant exposure of the population could represent a potential health hazard for vulnerable groups, especially children.

Paired indoor and outdoor concentrations of fine and coarse particulate matter (PM), PM2.5 reflectance [black carbon(BC)], and nitrogen dioxide (NO₂) were determined for sixteen weeks in 2008 at four elementary schools (two in high and two in low traffic density zones) in a U.S.–Mexico border community to aid a binational health effects study. Strong spatial heterogeneity was observed for all outdoor pollutant concentrations. Concentrations of all pollutants, except coarse PM, were higher in high traffic zones than in the respective low traffic zones. Black carbon and NO₂ appear to be better traffic indicators than fine PM. Indoor air pollution was found to be well associated with outdoor air pollution, although differences existed due to uncontrollable factors involving student activities and building/ventilation configurations. Results of this study indicate substantial spatial variability of pollutants in the region, suggesting that children’s exposures to these pollutants vary based on the location of their school.

In the present study, the risk to humans by consuming European sea bass (Dicentrarchus labrax), captured at three sites along a Hg contamination gradient, was evaluated by comparing muscle and kidney total Hg (T-Hg) levels with the European regulations for marketed fish. Moreover, T-Hg and organic Hg (O-Hg) levels in muscle were compared with the Provisional Tolerable Weekly Intake (PTWI) and the Reference Dose (RfD). Although T-Hg levels in muscle were below the European value allowable for marketed fish, kidney’s levels were higher than the set value, stressing the importance of redefining the concept of edible tissue and which tissues should be considered. Mercury weekly ingestion in the contaminated areas was higher than the PTWI, and O-Hg daily ingestion rates were higher than the RfD in all sampling sites. Thus, populations consuming sea bass from the contaminated sites may be at risk, with particular relevance for children and pregnant women.

This study determined the concentrations of major and trace elements in shellfish (oysters, clams and mussels) and conducted an assessment of the health risks due to the consumption of contaminated seafood. Samples were collected at 34 sites along Todos os Santos Bay, Brazil. The elements were determined by ICP OES and Hg by Direct Mercury Analysis. Relatively high concentrations of trace elements (As, Zn, Se and Cu) were found in seafood tissues. Potential daily intake of As, Co, Se, Zn and Cu associated to shellfish consumption suggested relevant non-carcinogenic risk for all studied locations. Copper was the element that posed the greatest non-carcinogenic risk, while Pb posed the highest carcinogenic risk. Health risks for humans were greatest from the consumption of mussels. Contaminated shellfish offer the greatest risk for children, subsistence fishers and subsistence shellfish consumers.

The biocide triclosan (TCS, 5-chloro-2-(2,4-dichlorophenoxy)phenol) is commonly used in several personal care products, textiles, and children’s toys. Because the removal of TCS by wastewater treatment plants is incomplete, its environmental fate is to be discharged into freshwater ecosystems, where its ecological impact is largely unknown. The aim of this study was to determine the effect of TCS on the antioxidant enzymatic chain of the freshwater mollusk zebra mussel (Dreissena polymorpha). We measured the activity of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx), as well as the phase II detoxifying enzyme glutathione S-transferase (GST) in zebra mussel specimens exposed to 1Â nM, 2Â nM, and 3Â nM TCS in vivo. The mussels were exposed for 96Â h, and the enzyme activities were measured every 24Â h. We measured clear activation of GST alone at all three dose levels, which shows a poor induction of the antioxidant enzymatic chain by TCS. CAT and SOD were activated only at 3Â nM, while GPx values overlapped the baseline levels.
