In this study, we aimed to identify the toxic effects of chlorpyrifos exposure on the tissues of common carp. For this purpose, we evaluated histopathological changes in the brain, gills, liver, kidney, testis, and ovaries after 21 days of chlorpyrifos exposure. Activation of 8-OHdG, cleaved caspase-3, and iNOS were assesed by immunofluorescence assay in chlorpyrifos-exposed brain and liver tissue. Additionally, we measured the expression levels of caspase-3, caspase-8, iNOS, MT1, CYP1A, and CYP3A genes in chlorpyrifos-exposed brain tissue, as well as the expression levels of FSH and LH genes in chlorpyrifos-exposed ovaries, using qRT-PCR. We observed severe histopathological lesions, including inflammation, degeneration, necrosis, and hemorrhage, in the evaluated tissues of common carp after both high and low levels of exposure to chlorpyrifos. We detected strong and diffuse signs of immunofluorescence reaction for 8-OHdG, iNOS, and cleaved caspase-3 in the chlorpyrifos-exposed brain and liver tissues. Furthermore, we found that chlorpyrifos exposure significantly upregulated the expressions of caspase-3, caspase-8, iNOS, and MT1, and also moderately upregulated CYP1A and CYP3A in the brain tissue of exposed carp. We also noted downregulation of FSH and LH gene expressions in chlorpyrifos-exposed ovary tissues. Based on our results, chlorpyrifos toxication caused crucial histopathological lesions in vital organs, induced oxidative stress, inflammation, and apoptosis in liver and brain tissues, and triggered reproductive sterility in common carp. Therefore, we can propose that chlorpyrifos toxication is highly dangerous to the health of common carp. Moreover, chlorpyrifos pollution in the water could threaten the common carp population. Use of chlorpyrifos should be restricted, and aquatic systems should be monitored for chlorpyrifos pollution.

To develop a test for assessing the immunomodulatory effects of chemical pollutants on fish, we evaluated the effects of dexamethasone on the natural host–pathogen interaction between common carp (Cyprinus carpio) and Aeromonas salmonicida. Carp were exposed to 1mgL−1 dexamethasone for the entire experimental period. One week after the exposure test started, the exposed fish, as well as unexposed fish, were bath-infected with A. salmonicida. One hundred percent mortality was observed in bacteria-infected fish exposed to dexamethasone, whereas no infection-associated mortality was observed in infected fish in the absence of dexamethasone exposure. In a separate experiment, dexamethasone exposure significantly suppressed hemolytic complement activity in bacteria-infected fish. These results clearly indicate that exposure to a high concentration of dexamethasone suppressed the carp immune system and caused subsequent mortality. Thus, this proposed test method is likely to be useful for evaluating the immunomodulatory effects of chemicals in fish.

The use of ultrasonication for cyanobacterial control in freshwater bodies has become increasingly popular during the last decades despite controversial efficiency on large scale application. Apart from that, little information is currently available regarding ultrasound toxicity potential towards non-target species. This work was designed to address this issue in the common carp using a low-power (7–9 W output) and dual-frequency (23 and 46 kHz) anti-cyanobacterial ultrasound device. Results showed that carps were unaffected by ultrasound exposure when exposed in floating cages in fish ponds over a 30-day period. The experiment duration was the main factor influencing all measured biological parameters in exposed and non-exposed organisms. Indeed, it was positively associated with an increase in fish condition factor. Cortisol level also tended to slightly increase over the number of days of experiment but its variation did not enable to sort out any ultrasound exposure-related stress. Moreover, an overall diminution along the experimental period of the expression level of a set of biomarkers could be reported, encompassing cellular antioxidant enzyme activities such as superoxide dismutase (SOD), glutathione peroxydase (GPx), catalase and glutathione S-transferase (GST), and lactate dehydrogenase activity. Subtle changes in these biomarkers were dependent of the type of enzyme activity and especially of the origin of fish (i.e., sampled pond) regardless of the presence of ultrasound equipment, reflecting thereby fish adaptation to local environmental conditions in each pond. In conclusion, this study does not provide indication that ultrasonication in the aforementioned conditions affects the welfare and physiological homeostasis of carps.

The Büyük Menderes River (BMR) is one of the largest rivers in Turkey. This river irrigates efficient farmlands and includes tributaries of other rivers and streams and many populated towns within its limits in the Ege region. Both the estuary and Işıklı Lake serve as a sanctuary for various waterbirds. Therefore, the BMR plays a critical role both for the inhabitants and for the ecosystem organisms in its environs. In the present study, we analyzed levels of metals including iron, barium, zinc, vanadium, cobalt, chromium, cadmium, copper, nickel, aluminum, arsenic, manganese, antimony, silver, selenium, boron, mercury, titanium, and lead in river water, sediment, fish (Cyprinus carpio; common carp), and in various waterbird (Fulica atra, Euroasian coot; Larus michahellis, yellow-legged gull; Ardea cinerea, grey heron; Larus melanocephalus, Mediterranean gull; and Pelecanus crispus, pelican) samples. Analyses were performed using an inductively coupled plasma–mass spectrometry (ICP-MS) instrument after sample preparation. Comparing metal concentrations among different sample types, it was found that barium, aluminum, and zinc are the major metals in river water, and zinc in common carp muscle, while iron, aluminum, and manganese are the major metals in sediments. Iron, zinc, copper, and aluminum were the highest in waterbird muscle tissue. Iron and barium were found to be the major metals in eggshell, while iron and zinc are the major metals in egg samples. A simple “worst-case scenario” model of risk assessment revealed that some of the analyzed metals may pose a risk for human health through consuming fish.

Concentrations of persistent organic pollutants (POPs) were quantified in river water and sediment, as well as in the liver and muscle tissues of Cyprinus carpio that were sampled four times in a year at three stations in the Büyük Menderes River (BMR). Potential biomarkers of possible cellular molecular damage, namely lipid peroxidation (LPO) degradation products, protein carbonyls (PCO) and DNA repair product 8-hydroxy-2′-deoxyguanosine (8-OHdG), were analysed. All the targeted pollutants were measurable both in biotic and abiotic samples. Interestingly, the results suggested that there was recent organochlorine pesticide (OCP) input into the river water in the first two sampling periods in all stations in contrast to prohibition, while input of polychlorinated biphenyls (PCBs) and polybrominated diphenylethers (PBDEs) was not detected. Liver POP concentrations were higher than in muscle, as expected, and were found to decrease from the first to the fourth sampling period in all stations, except PBDEs. Levels of LPO degradation products in the liver and in muscle tissues decreased from the first to the fourth sampling period. This suggests that these markers reflect the lipid damage in respective tissues due to the tissue burden of targeted POPs. Protein carbonyls were the highest in the first sampling period, followed by a dramatic decrease in the second, and then a gradual increase towards the fourth sampling period in all stations. 8-OHdG levels were lower in Sarayköy station in the first sampling period. Among the measured biomarkers, only several LPO degradation products were significantly correlated with OCPs and PCBs in liver tissue.

Microcystins (MCs) produced by cyanobacteria can accumulate in freshwater fish tissues and pose potential risk to human health. In this study, variations in MC content were examined in water samples and fish tissues during a Microcystis aeruginosa-dominated bloom in a fish pond in China, from June 2009 to August 2009. A total of ten microcystin variants were detected in water samples in the fish pond, including MCLR, MCRR, MCYR, MCLF, MCLY, MCLA, and MCLW, and other three undefined MC variants. MCLR was the dominant variant in the water samples, followed by MCLF and MCLY. The highest MC concentration in the common carp (Aristichthys nobilis) was found in the order intestine > spleen > muscle; however, the order with the most MC variants detected was muscle > spleen = kidney = gallbladder > liver = intestine. The highest overall MC concentration detected in the water samples was 12.24 μg L⁻¹, and the estimated daily intake for humans from the consumption of fish was about 0.48 μg MCLR eq kg⁻¹ body weight (BW), 12-fold the tolerable daily intake (TDI) proposed by the World Health Organization. Therefore, the combined risks from skin exposure and food intake in this fish pond should not be overlooked.

The objective of this study is to evaluate the current status of heavy metal concentrations in constructed wetland, Shaoguan (Guangdong, China). Sediments, three wetland plants (Typha latifolia, Phragmites australis, and Cyperus malaccensis), and six freshwater fish species [Carassius auratus (Goldfish), Cirrhinus molitorella (Mud carp), Ctenopharyngodon idellus (Grass carp), Cyprinus carpio (Wild common carp), Nicholsicypris normalis (Mandarin fish), Sarcocheilichthys kiangsiensis (Minnows)] in a constructed wetland in Shaoguan were collected and analyzed for their heavy metal compositions. Levels of Pb, Zn, Cu, and Cd in sediments exceeded approximately 532, 285, 11, and 66 times of the Dutch Intervention value. From the current study, the concentrations of Pb and Zn in three plants were generally high, especially in root tissues. For fish, concentrations of all studied metals in whole body of N. mormalis were the highest among all the fishes investigated (Pb 113.4 mg/kg, dw; Zn 183.1 mg/kg, dw; Cu 19.41 mg/kg, dw; 0.846 mg/kg, dw). Heavy metal accumulation in different ecological compartments was analyzed by principle component analysis (PCA), and there is one majority of grouped heavy metals concentration as similar in composition of ecological compartment, with the Cd concentration quite dissimilar. In relation to future prospect, phytoremediation technology for enhanced heavy metal accumulation by constructed wetland is still in early stage and needs more attention in gene manipulation area.

The aim of this study was to investigate heavy metals and selected minerals contents in filets of pikeperch (Sander lucioperca), carp (Cyprinus carpio) and flathead grey mullet (Mugil cephalus), the major fish species produced in Sidi-Salem reservoir; the largest Tunisian freshwater ecosystem. Concentrations of Ca, Mg, Fe, Zn, Cd, Pb and Hg were determined by inductively coupled plasma-optical emission spectrometry, while concentrations of Na and P were determined by flame photometry and spectrophotometry, respectively. Results concerning heavy metals are considered quantitatively reliable for Hg and Cd but must be taken with precautions concerning Pb levels as the LOD (limit of detection) of the analytical process was higher than acceptable limit. They showed greater accumulations of both Hg and Cd in filets of pikeperch than in filets of carp and mullet, but none of the values exceeded the normative maximum levels (0.500 and 0.050 mg kg⁻¹ w.w., respectively). Considering provisional intakes, target hazard quotient and hazard index, estimated for a fish portion per week, consumption of any of the species did not appear to be potentially hazardous for the health as they were far below threshold values. A fish portion would provide substantial amounts (>15% of daily requirements) in essential mineral nutrients (P, Mg and Fe) and thus should be considered an interesting contribution to a healthy diet. It is strongly advocated to authorities including freshwater fish in the official national annual control survey of contaminants in fishery products, which in addition to guaranteeing security of consumers, would also allow diversifying valorisation opportunities and thus increase the economic value of this fish production.
