Mcr-1 and blaNDM₋₁ antibiotic resistance genes (ARGs) confer resistance to colistins and carbapenems, which are often antibiotics used as a last resort in tertiary care hospitals. Dissemination of these two ARGs in drinking water supply systems and their effect on healthy gut bacteria are poorly studied. In this study, the dissemination of mcr-1 and blaNDM₋₁ in a drinking water supply system, and their effect on the antibiotic resistance of mouse gut bacteria are explored.Metagenome analysis revealed that source water (Taipu river and Jinze reservoir) was polluted with ARGs. Mcr-1 and blaNDM₋₁ can be disseminated through the water distribution system. Even advanced water treatments (ozone and biological activated carbon (BAC)) could not effectively remove mcr-1 and blaNDM₋₁. Low concentrations of chloramine disinfectants in the water distribution system were not effective at limiting ARG abundance. Mobile genetic elements were also found to play a major role in the dissemination of ARGs via horizontal gene transfer (HGT) throughout the water supply system. Statistical analysis revealed that there was no effect of temperature on the abundance of mcr-1 and blaNDM₋₁ throughout the water supply system.A last resort ARG, mcr-1 can disseminate from drinking water to the healthy mouse gut. The presence of mcr-1 in a strain belonging to Enterococcus hirae, which is different from the strain belonging to the Bacillus cereus group isolated from drinking water, strongly supports the phenomena of HGT inside the gut.This research provides novel insights into the role of drinking water in disseminating ARGs to the gut and strongly suggests that drinking water may also play a major role apart from other factors known to be involved in the prevalence of last resort ARGs in the gut.

Although macrophytes are known to play vital roles in aquatic ecosystems, most quantitative aquatic toxicity data focus on fishes, water fleas, or algae, with limited ecotoxicity data published on macrophytes. Salvinia natans is a fast-growing plant commonly found in freshwater habitats. In this study, we verified a suitable disinfectant for preventing foreign contamination and formulated a culture medium for ensuring high productivity of S. natans. Finally, we established methodology for S. natans to be used in ecotoxicity testing of heavy metals and pesticides. As global regulations are being developed to harmonize guidelines and laboratory test species, S. natans is emerging as a potential candidate. The toxicity data publicly available for S. natans are very limited; hence, this study reports an advantageous culturing technique to optimize healthy growth of this species in the laboratory and presents optimal toxicity results, achieved by modifying the currently available test guidelines for Lemna. Our findings expand the currently limited range of test species for aquatic toxicity assays. We conclude that S. natans could serve as a valuable test species for aquatic toxicity assays.

Current swine industry practice is to house animals in confinement facilities which capture and store feces and urine as slurry in pits below the production area. Additives and disinfectants may be introduced into the manure pits. This study was conducted to measure the effects of additives and disinfectants on temporal changes in swine slurry characteristics. Slurry from a commercial swine production facility in southeast Nebraska, USA was collected and transferred to 57 L reactors located within a greenhouse. Selected additives and disinfectants were added to the reactors and physical properties, chemical characteristics, and antibiotic concentrations were monitored for 40 days. Concentrations of dry matter (DM), total nitrogen (TN), phosphorus pentoxide (P₂O₅), calcium (Ca), magnesium (Mg), zinc (Zn), iron (Fe), manganese (Mn), and copper (Cu) were significantly greater than the Control in each of the reactors containing additives. The reactors in which the additives MOC-7, More Than Manure®, Sludge Away, and Sulfi-Doxx were introduced had significantly greater values of chemical oxygen demand (COD), total volatile solids (TVS), total suspended solids (TSS), total solids (TS), dry matter (DM), TN, P₂O₅, Ca, Mg, Zn, Fe, Mn, Cu and chlortetracycline than the other additive treatments. Concentrations of TVS and TSS were significantly lower in the reactors containing Clorox® and Virkon™ than the other disinfectant treatments. The total dissolved solids (TDS) concentration of 26,500 mg L⁻¹ and pH value of 7.27 obtained for the reactors containing Tek-Trol were significantly greater than measurements obtained for the other treatments. Concentrations of chlortetracycline and tiamulin of 8840 and 28.8 ng g⁻¹, respectively, were significantly lower for the treatments containing Tek-Trol. The sodium (Na) concentration of 1070 mg L⁻¹ measured in the reactors containing Clorox® was significantly greater than values for the other disinfectant treatments. The introduction of selected additives and disinfectants may influence certain physical properties, chemical characteristics, and antibiotic concentrations of swine slurry.

The most important effect of natural organic materials in water is reacting with disinfectants and creating disinfectant by-products that are mostly carcinogenic. The aim of this study was to determine the optimum conditions for removal of humic acid (HA) by zeolite coated with nZVI nanoparticles (Zeolite/nZVI) from aqueous solutions. In this study, after synthesis of zeolite/nZVI, its structure and morphology were examined using FTIR, BET, XRF, and FESEM techniques. The effects of HA concentration, composite content, pH, and reaction time were evaluated. The experimental data were analyzed by Langmuir and Freundlich isotherm and pseudo-first-order and second-order kinetic models. Finally, the thermodynamic parameters of enthalpy (ΔΗ°), entropy (ΔS°), and Gibbs free energy (ΔG°) were calculated. The results of the analyses confirmed the accuracy of the composite structure. Its specific surface area by using BET method was 203.43 m²/g. The HA removal efficiency was obtained at 92.98% in optimum conditions of 50 mg/L concentration, 2 g/L composite dose, pH = 3, and reaction time of 60 min. The results of the isotherm and kinetic study showed that the HA adsorption process follows the Langmuir isotherm (R² = 0.9707) and pseudo-second-order kinetic. The maximum adsorption capacity of the composite was determined at 23.36 mg/g by Langmuir model. Thermodynamic parameters indicate that the adsorption of HA endothermic and the reaction cannot be done spontaneously. Zeolite/nZVI composite had good removal efficiency after five times of recycling. The present study showed that zeolite/nZVI can be used as an effective adsorbent for removal of HA from aqueous solutions.

Pre-oxidation in water treatment is considered an effective method to enhance the removal of algal cells and their exuded organic matters. However, pre-oxidation also alters the characteristics of algae and consequently influences disinfection processes. The existing studies mainly focused on the stationary growth phase, but little is known for the exponential and declined phases. The objectives of this study were to examine the effects of pre-ozonation on the integrity of algal cells, the release of algal organic matters, and the formation of disinfection by-products like N-nitrosodimethylamine (NDMA) from Microcystis aeruginosa (M. aeruginosa) at three growth phases. The results demonstrated that pre-ozonation was efficient to inactivate M. aeruginosa cells. The severity of M. aeruginosa cell damage increased as the ozone dosage increased from 0.5 to 2.0 mg/L. The damage of cell membranes resulted in the release of intracellular organic matters. Excitation-emission matrix spectra (EEMS) analysis indicated that ozone mainly reacted with soluble microbial products (SMP). With the increase of ozone concentration, although the trend of NDMA formation was similar for all three growth phases, more production of NDMA by algal cells was observed at the declined phase. In the post-disinfection process, chloramine showed the potential as a more suitable disinfectant than chlorination after pre-ozonation to minimize the NDMA formation. Therefore, appropriate pre-ozonation is beneficial to reduce the NDMA formation from exponential algae, while has no significant change during both stationary and declined phases.

Recent developments in the nanotechnology field have created opportunities to design new biomaterials for Staphylococcus aureus biofilm eradication. These biomaterials including disinfectant-loaded nanoparticles could overcome the limitations of conventional disinfectants. The objective of this study was to assess the biocidal activity of five commercial disinfectants (DC&R®, VirkonS®, TH4++, Tek-Trol, and peracetic acid) alone and as with silver and copper nanocomposites on S. aureus biofilm at different concentrations and exposure times. Consequently, 227 samples were collected from two broiler farms, two-layer farms, and three abattoirs at El-Dakahlia Province, Egypt, during summer 2018. The samples were collected from birds as well as the surrounding environment. S. aureus strains were isolated and biofilm producers were phenotypically evaluated by Congo red agar (CRA) test. Besides, 4 biofilm-associated genes including bap, fnbA, cna, and ebps were genotypically detected by PCR technology. Out of 227 collected samples, 141 (62.1%) strains were identified as S. aureus, while 127 strains (90.1%) were S. aureus biofilm producers for all examined samples except for hand swabs of abattoir workers. The prevalence of fnbA and bap genes was 79.5% (101/127) and 20.5% (26/127), respectively but, no strains harbored cna or ebps genes. Tested nanocomposites were prepared using an aqueous solution of metal salts such as copper sulfate and silver nitrate and added to the same amount of disinfectant solution. The obtained nanocomposites were characterized by transmission electron microscopy (TEM) and zeta potential which showed spherical and elongated particles and with a surface charge of disinfectants—silver and copper nanocomposites—of 2.92 and 3.43 mV, respectively. Complete eradication of S. aureus biofilm was observed after treatment with disinfectants loaded onto silver (AgNPs) and copper (CuNPs) nanoparticles in varying concentrations as well as at different exposure times in comparing to disinfectants alone. Our results exhibited the potential applications of disinfectant nanocomposites in complete eradication of S. aureus biofilm in farms and abattoirs without developing of disinfectant resistant bacteria.

Chlorine-based compounds have been used as a disinfectant in drinking water treatment plants for decades because of their excellent sterilisation efficiency and low cost. However, the formation of disinfection by-products during chlorination is a cause for concern. Peracetic acid (PAA) is a strong oxidant with a redox potential higher than that of chlorine and does not form harmful disinfection by-products. It is thus a potential alternative for chlorine-based disinfectants. However, PAA decomposes rapidly in water at a rate that is highly affected by many factors, such as organic compounds and pH. The aim of this study is to investigate the stability of PAA during drinking water disinfection. To accomplish this, we studied methods for rapid detection of residual PAA and PAA decay in drinking water. Residual PAA was detected in water by the spectrophotometry-total chlorine reagent (SPTCR) method with a PAA concentration range of 0.090–10 mg/L (R² = 0.9943). Decay tests of PAA in drinking water and other sources of water showed that the decay process conformed to the first-order kinetic model with fast and slow reactions. Among four factors, pH was the key factor in the decay process because an alkaline environment significantly promotes the decomposition of PAA. In addition, total organic carbon (TOC), conductivity, and initial PAA concentration also affected PAA decay. Experimental and statistical analyses suggested that these factors affected PAA decay in the following descending order of influence: TOC, initial PAA concentration, and conductivity. In real water matrices, the PAA decay rate increased with increasing initial PAA concentration.
