A full life-cycle (240 days) bioassay using the terrestrial snail, Cantareus aspersus, allowing exposure during embryogenesis and/or the growth and reproduction phases, was used to assess the effects of Bypass®, a glyphosate-based herbicide (GlyBH), on a range of endpoints, including parameters under endocrine control. As a positive control, a mixture (R-A) made of diquat (Reglone®) and nonylphenols (NP, Agral®), known for its endocrine disrupting effects in other organisms, was tested. At environmental concentrations, both pesticides (R-A mixture and GlyBH) enhanced growth but reduced reproduction. The R-A mixture acted mainly on the fecundity through a delay in egg-laying of approximately 20 days and a strongly reduced number of clutches. This latter dysfunction may be caused by a permanent eversion of the penis, suggesting a disrupting effect at the neuro-endocrine level, which prevented normal mating. GlyBH acted on fertility, possibly due to a decrease in the fertilization of eggs laid by adults exposed during their embryonic development. These results, associated with the absence of observed effects on gonad histology of GlyBH exposed snails, suggested that the underlying mechanisms are neuro-endocrine.

Parabens are class of preservatives used in vast majority of commercial products, and a potential Endocrine Disrupting Chemical (EDC). The present study was undertaken to delineate the effects of n-butylparaben on F1 male progeny exposed maternally through gestation and lactation via subcutaneous route. The F0 dams were given subcutaneous injections of n-butylparaben from gestation day (GD) 6 to postnatal day (PND) 21 with doses of 10, 100, 1000 mg/kg Bw/day in corn oil. The F1 male rats were monitored for pubertal development and sexual maturation; these were sacrificed on PND 30, 45 and 75. On PND 75, these F1 male rats were subjected for fertility assessment with unexposed female rats.A delayed testicular descent at 100 and 1000 mg/kg Bw dose and delayed preputial separation at 10 mg/kg Bw dose was observed in exposed F1 male rats. Decreased sperm count, motility and Daily Sperm Production was observed at 100 mg/kg Bw dose at PND 75. Interestingly, the sperm transit time in the epididymis was accelerated at this dose. Significant perturbed testicular expression of steroid receptors (ERα and β, AR), INSL3 and StAR genes with increased T and LH levels indicates direct effect on spermatogenesis and steroidogenesis. These F1 generation adult rats were sub-fertile with increased (%) pre- and post-implantation loss at 100 and 1000 mg/kg Bw/day dose. This is the first report on n-butylparaben highlighting the involvement of testicular leydig cells with accelerated sperm transit time leading to reduced fertility in the maternally exposed F1 male rats through estrogenic/anti-androgenic action.

Pyrethroids are a class of widely used insecticides. Our recent epidemiological study of Chinese women reported that pyrethroid exposure was positively associated with the risk of primary ovarian insufficiency (POI). In this study, we utilized cypermethrin (CP), the most frequently detected pyrethroid in the environment, to recognize how lifelong and low-dose exposure to pyrethroids affects ovarian functions and the underlying mechanism(s). Female mice were exposed to CP at doses of human dietary intake of 6.7 μg/kg/day, an acceptable daily intake (ADI) of 20 μg/kg/day, or the chronic reference dose (RfD) of 60 μg/kg/day, starting from gestational day 0.5 until 44-week-old. We assessed effects on fertility, serum hormone levels, ovarian follicular development and ovarian transcriptomic profiles. Chronic exposure to CP at doses of ADI and RfD caused a significant reduction in the size of the primordial follicle pool on postnatal day (PND) 5 and the number of all types of follicles in 44-week-old mice, lower estrogen and higher gonadotropin levels, as well as decreased fertility. Significant increase in apoptosis and decrease in cell proliferation were observed in CP-exposed ovarian follicles from PND 5 and 44-week-old mice. Ovarian transcriptomic data showed that the pro-apoptotic protein BMF and the cell cycle inhibitor p27 were significantly up-regulated in CP-exposed ovaries. Cyp17a1, Cyp19a1 and Hsd17b1 genes involved in the key steps of steroidogenesis were down-regulated in the ovaries of female mice exposed to CP. This study first reported that lifelong exposure to CP at doses of ADI or RfD caused an ovarian phenotype similar to human POI in female mice and provided a mechanistic explanation. Our findings suggest that lifelong exposure to pyrethroids of low doses, which are recommended as ‘safe’ dosages, may have a significant impact on the ovarian health of female mammals and humans.

Testicular junctions are pivotal to male fertility and regulated by constituent proteins. Increasing evidence suggests that environmental chemicals, including bisphenol A (BPA), may impact these proteins, but whether the impacts persist for generations is not yet known. Here, we investigate the effect of BPA (a ubiquitous endocrine-disrupting chemical) on testis and sperm functions and whether the effects are transferred to subsequent generations. Male mice (F0) were exposed to corn oil (Control) or 5 or 50 mg BPA/kg body weight/day from 6 to 12 weeks of age. The F0 were mated with wild-type females to produce the first filial (F1) generation. F2 and F3 were produced using similar procedures. Our results showed that BPA doses decreased the levels of some junctional proteins partly via binding with estrogen receptors (ERα and Erβ), upregulation of p-ERK1/2, P85, p-JNK and activation of p38 mitogen-activated protein kinase signaling. Consequently, testicular histological abnormalities, disrupted spermatogenesis, decreased sperm count, and inability to fertilize eggs were observed in mice exposed to BPA. These effects were transferred to successive generations (F2), partly through DNA methylation, but mostly alleviated in F3 males. Our findings suggest that paternal exposure to chemicals promoting alteration of testicular junctional proteins and its transgenerational inheritance is a key component of the origin of male reproductive health problems.

Exposure of females to fine particulate matter ≤2.5 μm in diameter (PM2.5) prior to pregnancy could produce adverse impact on fertility and enhances susceptibility of the offspring to a variety of diseases. In the current study, female C57BL/6 mice (6 weeks of age) were exposed to either concentrated PM2.5 or filtered air (average PM2.5 concentration: 115.60 ± 7.77 vs. 14.07 ± 0.38 μg/m⁻³) using a whole-body exposure device for 12 weeks. Briefly, PM2.5 exposure decreased anti-Müllerian hormone level (613.40 ± 17.36 vs 759.30 ± 21.90 pg mL⁻¹, P＜0.01) and increased reactive oxygen species (ROS) level (45.39 ± 0.82 vs 24.20 ± 0.85 arbitrary unit in fluorescence assay, P＜0.01) in oocytes. The exposure increased oocyte degeneration rate (21.5% vs 5.1%, respectively (P＜0.01) and decreased the 2-cell formation rate (71.9% vs 86.0%, P < 0.01). Transcriptome profiling using RNA sequencing showed wide spectrum of abnormal expression of genes, particularly those involved in regulating the mitochondrial respiratory complex in oocytes and metabolic processes in blastocysts. The exposure decreased litter size (6 ± 0.37 vs 7 ± 0.26, P＜0.05) and weight (1.18 ± 0.02 vs 1.27 ± 0.02 g, P＜0.01). In summary, PM2.5 exposure decreased female fertility, possibly through increased ROS production in oocytes and metabolic disturbances in developing embryos. The cause-effect relationship, however, requires further investigation.

This research was conducted to quantitatively evaluate the application effect of the frost-resistant ecological substrate in the rock slope of the hydropower station. Field sampling and laboratory tests were conducted to determine the erosion resistance and fertility of frost-resistant ecological substrate, and the test results were compared with those of natural soils with similar site conditions. The research conclusions were as follows. Compared with the natural soil, the content of > 0.25 mm mechanical-stable aggregates, > 0.25 mm water-stable aggregates, average weight diameter, geometric average diameter, organic matter, available nitrogen, available phosphorus, and available potassium of frost-resistant ecological substrate, significantly increased. On the contrary, erodibility factor, percentage aggregate disruption, aggregate degree, and dispersion rate decreased evidently. These results showed that erosion resistance and fertility of the frost-resistant ecological substrate have a better prospect in the engineering application of alpine regions. In addition, the principal component analysis showed that the principal component value of frost-resistant ecological substrate increased by 1.9 times that of natural soil. According to the correlation study, the increase in the amount of > 0.25 mm macro-aggregates and organic matter is the primary reason that ecological substrate has greater stability and fertility than natural soil. In conclusion, the frost-resistant ecological substrate was a suitable soil to create a suitable vegetation growth environment on the surface of rock slope in the alpine region.

The degradation of cyanobacterial blooms often causes hypoxia and elevated concentrations of ammonia, which can aggravate the adverse effects of blooms on aquatic organisms. However, it is not clear how one stressor would work in the presence of other coexistent stressors. We studied the toxic effects of elevated ammonia under hypoxia using a common yet important cladoceran species Daphnia similis isolated from heavily eutrophicated Lake Taihu. A 3 × 2 factorial experimental design was conducted with animals exposed to three un-ionized ammonia levels under two dissolved oxygen levels. Experiments lasted for 14 days and we recorded the life-history traits such as survival, molt, maturation, and fecundity. Results showed that hypoxia significantly decreased survival time and the number of molts of D. similis, whereas ammonia had no effect on them. Elevated ammonia significantly delayed development to maturity in tested animals and decreased their body sizes at maturity. Both ammonia and hypoxia were significantly detrimental to the number of broods, the number of offspring per female, and the number of total offspring per female, and significantly synergistic interactions were detected. Our data clearly demonstrate that elevated ammonia and hypoxia derived from cyanobacterial blooms synergistically affect the cladoceran D. similis.

Human data on the relationship of semen quality with pesticide and metals are mostly inconsistent. The purpose of the study is to confirm the toxicity of organochlorine pesticide β- and γ-hexachlorocyclohexane (HCH), DDE and DDD, and metals lead or cadmium on sperm motility in epidemiological study among fertile and infertile men and to determine whether in vivo and in vitro results are in the same direction. Semen analysis and estimation of the toxicants were done in 60 fertile and 150 infertile men. In the in vitro studies, sperm were exposed to the highest levels of these toxicants found in vivo, as well as five and ten times higher, and to the mixture of all compounds. The study assesses sperm viability and motility for a period ranging between 30 min and 96 h. Epidemiological data showed an inverse correlation of toxicant with sperm motility. In vitro study showed that γ-HCH and lead after 12 h, cadmium after 8 h, and coexposure to toxicants after 6 h of exposure caused significant concentration- and duration-dependent decline in sperm motility. Data of in vitro study were concurrent with epidemiological finding that might be useful in establishing the possible association between exposure and effect of these selected pollutants on sperm motility.
