In this study we aimed to assess the dioxin- and estrogen-like activities of contaminants extracted from twenty species of freshwater and seawater fishes, using luciferase reporter assays. Transfected MCF7 cells were treated with sample extracts and luciferase activities were then measured at 24-h of post-treatment. The mean values of the detected dioxin- and estrogen-like activities in the freshwater fishes were 25.3 pg TEQ/g ww and 102.3 pM EEQ/g ww whereas in the seawater fishes, the values were 46.2 pg TEQ/g ww and 118.8 pM EEQ/g ww. Using sample-relevant dosage of estrogen, inductions of cell proliferation markers (i.e. retinoblastoma, cyclin D) and stimulations of cell growth were revealed by Western blotting, colony formation and BrdU uptake assays. A cotreatment with TCDD significantly reduced these effects. Using the sample extracts with different dioxin- and estrogen-like activities, similar observation was revealed. The data highlighted the mixture effect of food contaminants on human health.

In an extensive environmental study, field samples, including soil, water, rice, vegetable, fish, human hair and urine, were collected at an abandoned tungsten mine in Shantou City, southern China. Results showed that arsenic (As) concentration in agricultural soils ranged from 3.5 to 935 mg kg−1 with the mean value of 129 mg kg−1. In addition, As concentration reached up to 325 μg L−1 in the groundwater, and the maximum As concentration in local food were 1.09, 2.38 and 0.60 mg kg−1 for brown rice, vegetable and fish samples, respectively, suggesting the local water resource and food have been severely contaminated with As. Health impact monitoring data revealed that As concentrations in hair and urine samples were up to 2.92 mg kg−1 and 164 μg L−1, respectively, indicating a potential health risk among the local residents. Effective measurements should be implemented to protect the local community from the As contamination in the environment. It is the first report on arsenic contamination and potential health risk implications at abandoned Lianhuashan tungsten mine.

Contaminated sediments in the St. Lawrence River remain a difficult problem despite decreases in emissions. Here, sediment and pore water phases were analyzed for total mercury (THg) and methyl mercury (MeHg) and diffusion from the sediment to the overlying water was 17.5 ± 10.6 SE ng cm−2 yr−1 for THg and 3.8 ± 1.7 SE ng cm−2 yr−1 for MeHg. These fluxes were very small when compared to the particle-bound mercury flux accumulating in the sediment (183 ± 30 SE ng cm−2 yr−1). Studies have reported that fish from the westernmost site have higher Hg concentrations than fish collected from the other two sites of the Cornwall Area of Concern, which could not be explained by differences in the Hg flux or THg concentrations in sediments, but the highest concentrations of sediment MeHg, and the greatest proportions of MeHg to THg in both sediment and pore water were observed where fish had highest MeHg concentrations.

We performed a trace analytical study covering nine hormonally active UV-filters by LC–MS/MS and GC–MS in river water and biota. Water was analysed at 10 sites above and below wastewater treatment plants in the river Glatt using polar organic chemical integrative samplers (POCIS). Four UV-filters occurred in the following order of decreasing concentrations; benzophenone-4 (BP-4) > benzophenone-3 (BP-3) > 3-(4-methyl)benzylidene-camphor (4-MBC) > 2-ethyl-hexyl-4-trimethoxycinnamate (EHMC). BP-4 ranged from 0.27 to 24.0 μg/POCIS, BP-3, 4-MBC and EHMC up to 0.1 μg/POCIS. Wastewater was the most important source. Levels decreased with higher river water flow. No significant in-stream removal occurred. BP-3, 4-MBC and EHMC were between 6 and 68 ng/L in river water. EHMC was accumulated in biota. In all 48 macroinvertebrate and fish samples from six rivers lipid-weighted EHMC occurred up to 337 ng/g, and up to 701 ng/g in 5 cormorants, suggesting food-chain accumulation. UV-filters are found to be ubiquitous in aquatic systems.

Current European Union regulation regarding polychlorinated dibenzo-p-dioxins, polychlorinated dibenzofurans (PCDD/Fs) and dioxin-like polychlorinated biphenyls (dl-PCBs) in food and feed is based on Toxic Equivalent Quotient (TEQ) concept. For confirmatory purpose, the isotope-dilution method associated to a measurement by gas chromatography coupled with high resolution mass spectrometry is usually the method of choice for precisely measuring the 29 target congeners in three separated fractions. Time and cost related to these analyses are very significant. Various kinds of screening concepts can be considered. In the present study, we elaborated and validated a prediction model for the 2005 World Health Organization TEQ in fish, based on the measurement of 4 PCDD/F and 2 non-ortho dl-PCB congeners, potentially analyzable in a single extracted fraction by gas chromatography coupled with mass spectrometry. Large independent datasets have been used for model elaboration (n = 108) and validation (n = 363, n = 357 and n = 6). This study describes a statistical regression model approach for screening PCDD/Fs and dl-PCBs in fish.

In this work the risk posed to seawater organisms, predators and humans is assessed, as a consequence of exposure to 12 organic micro-pollutants, namely metronidazole, trimethoprim, erythromycin, simazine, flumequine, carbaryl, atrazine, diuron, terbutryn, irgarol, diphenyl sulphone (DPS) and 2-thiocyanomethylthiobenzothiazole (TCMTB). The risk assessment study is based on a 1-year monitoring study at a Spanish marine fish farm, involving passive sampling techniques. The results showed that the risk threshold for irgarol concerning seawater organisms is exceeded. On the other hand, the risk to predators and especially humans through consumption of fish is very low, due to the low bioconcentration potential of the substances assessed.

To determine the potential input sources of polybrominated diphenyl ethers (PBDEs) to fish farming environments in South China, samples of seven various environmental matrices were collected from October 2006-September 2007. Tri- to deca-BDEs were detected in all samples analyzed, with mean concentrations (±standard deviations) at 5.7 ± 3.6 ng/L in pond water, 15 ± 11 ng/g dry wt. in pond sediment, 12 ± 3.8 ng/g dry wt. in bank soil, 21 ± 20 ng/g lipid wt. in fish, and 93 ± 62 ng/g lipid wt. in fish feeds. In addition, BDE-209 was the major constituent in all samples except fish and BDE-47 was predominant in fish samples. Relatively high abundances of BDE-49 were detected in all the samples compared to those in the penta-BDE technical products. Several bioaccumulation factors were evaluated. Finally, statistical analyses suggested that fish feed, as well as pond water at a lesser degree, may have been the major source of PBDEs in freshwater farmed fish.

The responses of Dicentrarchus labrax and Liza aurata to aquatic pollution were assessed in a contaminated coastal lagoon, using both traditional and novel biomarkers combined. DNA damage, assessed by comet assay, was higher in both fish species from the contaminated sites, whereas levels of cytochrome P450 1A1 gene expression were not significantly altered. The liver histopathological analysis also revealed significant lesions in fish from contaminated sites. Alterations in ras and xpf genes were analysed and additional pollutant-responsive genes were identified. While no alterations were found in ras gene, a downregulation of xpf gene was observed in D. labrax from a contaminated site. Suppression subtractive hybridization applied to D. labrax collected at a contaminated site, revealed altered expression in genes involved in energy metabolism, immune system activity and antioxidant response. The approach and results reported herein demonstrate the utility of anchoring traditional biomarker responses alongside novel biomarker responses.

High mountains may play significant roles in the global transport of persistent organic pollutants (POPs). This work aims to investigate the levels, patterns and distribution of semi-volatile organoclorine pollutants and to improve the understanding of the long-range atmospheric transport and fate of contaminants on the Tibetan Plateau. A total of 60 fish samples were collected from eight lakes located between 2813 and 4718 m above sea level across the Plateau. Concentrations of polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs) including dichlorodiphenyltrichloroethane and its metabolites (DDTs), hexachlorocyclohexanes (HCHs) and hexachlorobenzene (HCB) were measured in fish muscle. The results showed that concentrations of DDT, HCH and HCB were comparable to or lower than those found in remote mountains of Europe, Canada and US, while PCB concentrations in fish were, on average, about 4-150 times lower on Tibet than at other mountain areas. The transport and fate of contaminants in the Plateau are significantly influenced by the unique climatological and meteorological conditions, particularly by the summer Indian monsoon and winter westerly jet stream.

Background, aim and scope Toxicity testing has become a suitable tool for wastewater evaluation included in several reference documents on best available techniques of the Integrated Pollution Prevention and Control (IPPC) Directive. The IPPC Directive requires that for direct dischargers as well as for indirect dischargers, the same best available techniques should be applied. Within the study, the whole effluent assessment approach of OSPAR has been applied for determining persistent toxicity of indirectly discharged wastewater from the metal surface treatment industry. Materials and methods Twenty wastewater samples from the printed circuit board and electroplating industries which indirectly discharged their wastewater to municipal wastewater treatment plants (WWTP) have been considered in the study. In all factories, the wastewater partial flows were separated in collecting tanks and physicochemically treated in-house. For assessing the behaviour of the wastewater samples in WWTPs, all samples were biologically pretreated for 7 days in the Zahn-Wellens test before ecotoxicity testing. Thus, persistent toxicity could be discriminated from non-persistent toxicity caused, e.g. by ammonium or readily biodegradable compounds. The fish egg test with Danio rerio, the Daphnia magna acute toxicity test, the algae test with Desmodesmus subspicatus, the Vibrio fischeri assay and the plant growth test with Lemna minor have been applied. All tests have been carried out according to well-established DIN or ISO standards and the lowest ineffective dilution (LID) concept. Additionally, genotoxicity was tested in the umu assay. The potential bioaccumulating substances (PBS) were determined by solid-phase micro-extraction and referred to the reference compound 2,3-dimethylnaphthalene. Results The chemical oxygen demand (COD) and total organic carbon (TOC) values of the effluents were in the range of 30-2,850 mg L⁻¹ (COD) and 2-614 mg L⁻¹ (TOC). With respect to the metal concentrations, all samples were not heavily polluted. The maximum conductivity of the samples was 43,700 µS cm⁻¹ and indicates that salts might contribute to the overall toxicity. Half of the wastewater samples proved to be biologically well treatable in the Zahn-Wellens test with COD elimination above 80%, whilst the others were insufficiently biodegraded (COD elimination 28-74%). After the pretreatment in the Zahn-Wellens test, wastewater samples from four (out of ten) companies were extremely ecotoxic especially to algae (maximum LIDA = 16,384). Three wastewater samples were genotoxic in the umu test. Applying the rules for salt correction of test results as allowed in the German Wastewater Ordinance, only a small part of toxicity could be attributed to salts. Considering the PBS, wastewater from the metal surface treatment industry exhibited very low levels of PBS. In one factory, the origin of ecotoxicity has been attributed to the organosulphide dimethyldithiocarbamate (DMDTC) used as a water treatment chemical for metal precipitation. The assumption based on rough calculation of input of the organosulphide into the wastewater was confirmed in practice by testing its ecotoxicity at the corresponding dilution ratio after pretreatment in the Zahn-Wellens test. Whilst the COD elimination of DMDTC was only 32% in 7 days, the pretreated sample exhibited a high ecotoxicity to algae (LIDA = 1,536) and luminescent bacteria (LIDlb = 256). Discussion Comparative data from wastewater surveillance by authorities (data from 1993 to 2007) confirmed the range of ecotoxicity observed in the study. Whilst wastewater from the metal surface treatment industry usually did not exhibit ecotoxicity (median LID 1-2), the maximum LID values reported for the algae, daphnia and luminescent bacteria tests were very high (LIDA up to 3,072, LIDD up to 512 and LIDlb up to 2,048). DMDTC was found to be one important source of ecotoxicity in galvanic wastewater. DMDTC is added in surplus, and according to the supplier, the amount in excess should be detoxified with ferric chloride or iron sulphate. The operator of one electroplating company had not envisaged a separate treatment of the organosulphide wastewater but was assuming that excess organosulphide would be bound by other heavy metals in the sewer. DMDTC degrades via hydrolysis to carbon disulfide (which is also toxic to animals and aquatic organisms), carbonyl sulphide, hydrogen sulphide and dimethylamine, but forms complexes with metals which stabilise the compound with respect to transformation. Although no impact on the WWTP is expected, the question arises whether the organosulphide is completely degraded during the passage of the WWTP. Conclusions and recommendations The results show that the organic load of wastewater from the electroplating industry has been underestimated by focussing on inorganic parameters such heavy metals, sulphide, cyanide, etc. Bioassays are a suitable tool for assessing the ecotoxicological relevance of these complex organic mixtures. The proof of biodegradability of the organic load (and its toxicity) can be provided by the Zahn-Wellens test. The environmental safety of water treatment chemicals should be better considered. The combination of the Zahn-Wellens test followed by the performance of ecotoxicity tests turned out to be a cost-efficient suitable instrument for the evaluation of indirect dischargers and considers the requirements of the IPPC Directive.
