International audience | Food additives are one major hallmark of ultra-processed food in the Western-diet, a food habit often associated with metabolic disorders. Among these additives, the whitener and opacifying agent titanium dioxide (TiO2) raises public health issues due to the ability of TiO2 nanoparticles (NPs) to cross biological barriers and accumulate in different systemic organs like spleen, liver and pancreas. However before their systemic passage, the biocidal properties of TiO2 NPs may alter the composition and activity of the gut microbiota, which play a crucial role for the development and maintenance of immune functions. Once absorbed, TiO2 NPs may further interact with immune intestinal cells involved in gut microbiota regulation. Since obesity-related metabolic diseases such as diabetes are associated with alterations in the microbiota-immune system axis, this raises questions about the possible involvement of long-term exposure to food-grade TiO2 in the development or worsening of these diseases. The current purpose is to review the dysregulations along the gut microbiota-immune system axis after oral TiO2 exposure compared to those reported in obese or diabetic patients, and to highlight potential mechanisms by which foodborne TiO2 NPs may increase the susceptibility to develop obesity-related metabolic disorders.

Quantification of four artificial sweeteners in Finnish surface waters with isotope-dilution mass spectrometry

The artificial sweeteners sucralose (SCL), acesulfame (ACS), saccharin (SAC), and cyclamate (CYC) have been detected in environmental waters in Europe and North America. Higher environmental levels are expected in view of the increasing consumption of these food additives. In this study, an isotope-dilution mass spectrometry (IDMS) LC–MS/MS method was developed and validated for quantifying the four artificial sweeteners in boreal lakes (n = 3) and rivers (n = 12). The highest concentrations of ACS, SAC, CYC and SCL were 9,600, 490, 210 and 1000 ng/L, respectively. ACS and SAC were detected in all studied samples, and CYC and SCL in 98% and 56% of the samples. Seasonal trends of ACS and SAC were observed in some rivers. ACS and SCL concentrations in rivers correlated linearly with population equivalents of the wastewater treatment plants in the catchment areas, whereas SAC and CYC concentrations depend more on the source.

In this study, the use of co-occurring discriminators of sewage and manure was assessed as a potential way to disentangle sewage and manure sources. A suite of human and veterinary derived chemical markers, which includes pharmaceuticals and compound such as food additives, has been identified for this purpose. The suite was selected in such a manner as to provide additional source characterisation, e.g. differentiating raw versus treated sewage inputs. An SPE–LC–MS/MS method was developed and validated for the determined suite of chemical markers with a detection limit of up to 50 pg L−1. This represents one of the lowest limits of detection for pharmaceuticals reported in literature. To illustrate the suitability of the proposed method to differentiate sewage and manure inputs to surface water bodies, results from surface water samples collected at monitoring sites corresponding to specific land use types within Ireland are discussed.

Propylene glycol (PG; 1,2-propanediol) has been commonly used as a food additive and vehicle in pharmaceutical preparations. PG can form rectus (R-) enantiomers and sinister (S-) enantiomers. Herein, Kunming mice were used as the animal model to evaluate the acute and subacute oral toxicity of R-PG, S-PG and RS-PG (1:1 racemic mixture of R-PG and S-PG). The median lethal doses of R-PG, S-PG and RS-PG administered by oral gavage to mice were 22.81 g/kg, 26.62 g/kg and 24.92 g/kg, respectively. In the 28-day oral subacute toxicity study, the body weight, organ weights, serum biochemical, and renal histology were examined. There was no difference in subacute toxicity among R-PG, S-PG and RS-PG. The administration of 1 and 5 g/kg/day PG for 28 days caused nephrotoxicity. The kidney somatic index and levels of blood urea nitrogen exhibited a significant increase. Moreover, the activities of superoxide dismutase, catalase, and glutathione peroxidase significantly decreased after the treatment with PG. The levels of malondialdehyde, tumor necrosis factor α, interleukin 1β, and interleukin 6 significantly increased in the kidney. The results show that the nephrotoxic effects of PG are induced by oxidative stress, and the activation of the inflammatory response is mediated by the NF-κB signaling pathway. Together, these findings provide information on R-PG, S-PG and RS-PG treatments for the risk assessment of toxicity and effects on human health.

Pollution by chemical substances is of concern for the maintenance of healthy and sustainable aquatic environments. While the occurrence and fate of numerous emerging contaminants, especially pharmaceuticals, is well documented in freshwater, their occurrence and behavior in coastal and marine waters is much less studied and understood. This study investigates the occurrence of 58 chemicals in the open surface water of the Western Mediterranean Sea for the first time. 70 samples in total were collected in 10 different sampling areas. 3 pesticides, 11 pharmaceuticals and personal care products and 2 artificial sweeteners were detected at sub-ng to ng/L levels. Among them, the herbicide terbuthylazine, the pharmaceuticals caffeine, carbamazepine, naproxen and paracetamol, the antibiotic sulfamethoxazole, the antibacterial triclocarban and the two artificial sweeteners acesulfame and saccharin were detected in all samples. The compound detected at the highest concentration was saccharin (up to 5.23 ng/L). Generally small spatial differences among individual sampling areas point to a diffuse character of sources which are likely dominated by WWTP effluents and runoffs from agricultural areas or even, at least for pharmaceuticals and artificial food additives, from offshore sources such as ferries and cruising ships. The implications of the ubiquitous presence in the open sea of chemicals that are bio-active or toxic at low doses on photosynthetic organisms and/or bacteria (i.e., terbuthylazine, sulfamethoxazole or triclocarban) deserve scientific attention, especially concerning possible subtle impacts from chronic exposure of pelagic microorganisms.

The detection rates of pharmaceuticals (Ps), personal care products (PCPs), current-use pesticides (CUPs) and a food additive (FA) in Brisbane River estuary (Queensland), Sydney estuary (New South Wales) and the Yarra River estuary (Melbourne, Victoria) were: Ps: 16/25, 7/25 and 12/25, respectively, CUPs; 28/53, 5/53 and 23/53, respectively, PCPs: 1/3, 0/3 and, 1/3, respectively and FA; 1/1, 1/1 and 1/1, respectively. Diuron was measured in all estuarine samples, simazine, MCPA and 2,4 D were also commonly measured. Pharmaceuticals: carbamazepine, iopromide paracetamol tramadol and venlafaxine were also commonly measured across the estuaries. Generally, analytes were prominent in Brisbane River estuary, followed by Yarra River/Sydney estuary. Inputs of Ps are likely from leakages or effluents of WWTPs; CUPs are potentially from agricultural and parklands via surface run-off in Brisbane River estuary, while for Sydney and Yarra estuaries, which have separate stormwater and sewer systems, sources are likely to be ingression and leakage.

The concentration of nine trace elements were analyzed in the different tissue organs of commonly available crabs (Portunus sanguinolentus, Portunus pelagicus and Scylla serrate) and bivalve (Polymesoda erosa) species collected from the Miri coast, Borneo in order to evaluate the potential health risk by consumption of these aquatic organisms. Among the analyzed organs, metal accumulation was higher in the gill tissues. The essential (Cu and Zn) and non-essential (Pb and Cd) elements showed the highest (i.e. Zn) and lowest concentrations (i.e. Cd) in their tissue organs, respectively. The estimated daily intake and hazard indices of all metals in the muscle indicate that the measured values were below the provisional tolerable daily intake suggested by the joint FAO/WHO Expert Committee on Food Additives. Compared to Malaysian and international seafood guideline values the results obtained from the present study are lower than the permissible limits and safe for consumption.

The main objectives of the present study were to measure the concentration of mercury in Alosa spp. muscle tissue and to assess the health risk to consumers. For this purpose, fish samples were collected from four regions of the southern coast of the Caspian Sea. Mean concentrations of mercury in the regions of Bandar Torkaman, Feridonkenar, Chalos and Bandar Anzali were 264.10, 73.74, 161.90, and 183.80 μg g−1 dw, respectively. The mercury averages obtained in all four regions were below the standards recommended by the World Health Organization (WHO), the Food and Agriculture Organization (FAO), the Standardization Administration of China (SAC) and the Joint FAO/WHO Expert Committee on Food Additives (JECFA). The values of hazard quotient also showed no potential health risk for consumer. However, the assessment of other contaminants is recommended for more accurate conclusions about the health risks of consumers through the consumption of the fish.
