Analysis of heavy metal concentration in some vegetables using atomic absorption spectroscopy

This study assesses heavy metal levels in water, soil, and vegetables (swiss chard, lettuce, cabbage, collard green, tomato, green pepper and carrot) irrigated with waste water in Gamo, Ethiopia. The samples of soils, water, and vegetables were randomly collected, processed, and analyzed for heavy metals using atomic absorption spectrophotometry. The results obtained show that the irrigational water is profoundly contaminated with heavy metals Cd, Cr and Ni and Pb, Zn and Cu had the lowest concentration in irrigation water. The levels of Cd in Kulfo river area and Chamo Lake area and Ni in most of the farm soils were also found to be higher than the guideline values. The study also revealed that the mean levels of Cd in most vegetables and Cr and Pb in some vegetables were higher than the maximum recommended limits set by WHO/FAO. In general the results show that the highest concentration of the heavy metals was obtained from Kulfo river area compared to the Arbaminch textile share company area, Abaya Lake area, and Chamo Lake area. Cabbage was maximally contaminated with potential toxic elements followed by Swiss-chard, carrot, tomato, collard green, green pepper and lettuce. Hence, from kulfo river area frequent consumption of cabbage and Swiss chard may cause serious health risks to consumers. The levels of many elements were found to vary with location, suggesting localized inputs of the various contaminants related to industrial and other activities that generate wastewater. This study recommends regular monitoring of heavy metals in soils, waters, and foodstuffs to prevent excessive accrual in food chain.

Pesticide application has increased globally with increasing demand for food, and modernized Agriculture as a result of an explosion in the world’s population growth, especially in developing nations in Africa, Asia, and South America. However, pesticides have helped to improve productivity, protect the nutritive integrity of food crops, and ensure year-round food supplies worldwide. The production and consumption of pesticides persisted from one decade to another until the ecosystem started to suffer from its adverse effects on the environment and human health. Previous investigations revealed that pesticides found entry into the human food chain. In response to these problems, researchers all over the world have conducted several kinds of research on pesticide applications, and their residual contamination in food. This review crosses from the past to present researches on the usage of pesticides, their accumulation in food, and possible methods of their reduction as highlighted by researchers over many years. There is a need for continuous monitoring of pesticide residue profile in soil, crop produce, and animal products in developing countries so that it will not exceed maximum residue limits (MRLs).

Increasing consumption of chicken products in Iran makes it very important to analyze their residual heavy metal contents; therefore, the present study has been conducted to determine Pb, Cd, Cr, and Cu in commercial hen eggs, marketed in the city of Hamedan in 2016. In this descriptive study, a sum of 27 samples of hen eggs has been collected from the Hamedan City's market. After preparation and processing the samples in the laboratory, the concentration of metals has been determined in mixed albumen and yolk, using inductively coupled plasma-optical emission spectrometry. Also, all statistical analyses have been conducted, using the SPSS statistical package (version 20) with the results showing that the mean concentrations (mg/kg) of Pb, Cd, Cr, and Cu in the samples have been 0.29±0.16, 0.18±0.04, 0.31±0.03, and 2.81±1.56, respectively. Also, the mean contents of Cd and Cr have surpassed the maximum permissible levels (MPL), established by WHO/FAO. The computed health risk index values show that there is no potential risk for adults and children through egg consumption at the current rate in the study area.According to the results, considering the mean contents of Cd and Cr observed in egg samples have been higher than the MPL; therefore, it is recommended to pay serious attention to pollutants discharge in the environment and monitor chemical residue, especially heavy metals, in the foodstuff.

Show more [+] Less [-]A metal chaperone OsHIPP16 detoxifies cadmium by repressing its accumulation in rice crops

Cadmium (Cd) is an environmentally polluted toxic heavy metal and seriously risks food safety and human health through food chain. Mining genetic potentials of plants is a crucial step for limiting Cd accumulation in rice crops and improving environmental quality. This study characterized a novel locus in rice genome encoding a Cd-binding protein named OsHIPP16, which resides in the nucleus and near plasma membrane. OsHIPP16 was strongly induced by Cd stress. Histochemical analysis with pHIPP16::GUS reveals that OsHIPP16 is primarily expressed in root and leaf vascular tissues. Expression of OsHIPP16 in the yeast mutant strain ycf1 sensitive to Cd conferred cellular tolerance. Transgenic rice overexpressing OsHIPP16 (OE) improved rice growth with increased plant height, biomass, and chlorophyll content but with a lower degree of oxidative injury and Cd accumulation, whereas knocking out OsHIPP16 by CRISPR-Cas9 compromised the growth and physiological response. A lifelong trial with Cd-polluted soil shows that the OE plants accumulated much less Cd, particularly in brown rice where the Cd concentrations declined by 11.76–34.64%. Conversely, the knockout oshipp16 mutants had higher levels of Cd with the concentration in leaves being increased by 26.36–35.23% over the wild-type. These results suggest that adequate expression of OsHIPP16 would profoundly contribute to Cd detoxification by regulating Cd accumulation in rice, suggesting that both OE and oshipp16 mutant plants have great potentials for restricting Cd acquisition in the rice crop and phytoremediation of Cd-contaminated wetland soils.

Heavy metal toxicity has become an impediment to agricultural productivity, which presents major human health concerns in terms of food safety. Among them, arsenic (As) a non-essential heavy metal has gained worldwide attention because of its noxious effects on agriculture and public health. The increasing rate of global warming and anthropogenic activities have promptly exacerbated As levels in the agricultural soil, thereby causing adverse effects to crop genetic and phenotypic traits and rendering them vulnerable to other stresses. Conventional breeding and transgenic approaches have been widely adapted for producing heavy metal resilient crops; however, they are time-consuming and labor-intensive. Hence, finding new mitigation strategies for As toxicity would be a game-changer for sustainable agriculture. One such promising approach is harnessing plant microbiome in the era of ‘omics’ which is gaining prominence in recent years. The use of plant microbiome and their cocktails to combat As metal toxicity has gained widespread attention, because of their ability to metabolize toxic elements and offer an array of perquisites to host plants such as increased nutrient availability, stress resilience, soil fertility, and yield. A comprehensive understanding of below-ground plant-microbiome interactions and their underlying molecular mechanisms in exhibiting resilience towards As toxicity will help in identifying elite microbial communities for As mitigation. In this review, we have discussed the effect of As, their accumulation, transportation, signaling, and detoxification in plants. We have also discussed the role of the plant microbiome in mitigating As toxicity which has become an intriguing research frontier in phytoremediation. This review also provides insights on the advancements in constructing the beneficial synthetic microbial communities (SynComs) using microbiome engineering that will facilitate the development of the most advanced As remedial tool kit in sustainable agriculture.

Plants detoxify toxic metal(loid)s by accumulating diverse metabolites. Beside scavenging excess reactive oxygen species (ROS) induced by metal(loid)s, some metabolites chelate metal(loid) ions. Classically, thiol-containing compounds, especially glutathione (GSH) and phytochelatins (PCs) are thought to be the major chelators that conjugate with metal(loid)s in the cytoplasm followed by transport and sequestration in the vacuole. In addition to this classical detoxification pathway, a role for secondary metabolites in metal(loid) detoxification has recently emerged. In particular, anthocyanins, a kind of flavonoids with ROS scavenging potential, contribute to enhanced arsenic tolerance in several plant species. Evidence is accumulating that, in analogy to GSH and PCs, anthocyanins may conjugate with arsenic followed by vacuolar sequestration in the detoxification event. Exogenous application or endogenous accumulation of anthocyanins enhances arsenic tolerance, leading to improved plant growth and productivity. The application of some plant hormones and signaling molecules stimulates endogenous anthocyanin synthesis which confers tolerance to arsenic stress. Anthocyanin biosynthesis is transcriptionally regulated by several transcription factors, including myeloblastosis (MYBs). The light-regulated transcription factor elongated hypocotyl 5 (HY5) also affects anthocyanin biosynthesis, but its role in arsenic tolerance remains elusive. Here, we review the mechanism of arsenic detoxification in plants and the potential role of anthocyanins in arsenic tolerance beyond the classical points of view. Our analysis proposes that anthocyanin manipulation in crop plants may ensure sustainable crop yield and food safety in the marginal lands prone to arsenic pollution.

Surface water is one of the primary sources of irrigation water for produce production; therefore, its contamination by foodborne pathogens, such as Salmonella, may substantially impact public health. In this study, we determined the presence of Salmonella in surface water and characterized the relationship between Salmonella detection and environmental and anthropogenic factors. From April 2019 to February 2020, 120 samples from 30 sites were collected monthly in four watersheds located in two different central Chile agricultural regions (N = 1080). Water samples from rivers, canals, streams, and ponds linked to each watershed were obtained. Surface water (10 L) was filtrated in situ, and samples were analyzed for the presence of Salmonella. Salmonella was detected every month in all watersheds, with a mean detection percentage of 28% (0%–90%) across sampling sites, regardless of the season. Overall, similar detection percentages were observed for both regions: 29.1% for Metropolitan and 27.0% for Maule. Salmonella was most often detected in summer (39.8% of all summer samples tested positive) and least often in winter (14.4% of winter samples). Random forest analysis showed that season, water source, and month, followed by latitude and river, were the most influential factors associated with Salmonella detection. The influences of water pH and temperature (categorized as environmental factors) and factors associated with human activity (categorized as anthropogenic factors) registered at the sampling site were weakly or not associated with Salmonella detection. In conclusion, Salmonella was detected in surface water potentially used for irrigation, and its presence was linked to season and water source factors. Interventions are necessary to prevent contamination of produce, such as water treatment before irrigation.

The present study evaluated the interactive effects of global change and heavy metals on the growth and development of three soybean [Glycine max (L.) Merrill] cultivars and the consequences on yield and food safety. Soybean cultivars (Alim 3.14 from Argentina, and ES Mentor and Sigalia, from Germany) were grown until maturity in heavy metals polluted soils from the Rhine Valley, Germany, at two CO₂ concentrations (400 and 550 ppm) and heat stress (HS) episodes (9 days with 10 °C higher than maximum regular temperature) during the critical growth period in controlled environmental chambers. Different morpho-physiological parameters, heavy metal concentration in aerial organs, seed quality parameters, and toxicological index were recorded. The results showed that no morphological differences were observed related to CO₂. Moreover, Alim 3.14 showed the highest yield under control conditions, but it was more sensitive to climatic conditions than the German cultivars, especially to heat stress which strongly reduces the biomass of the fruits. Heavy metals concentration in soil exceeds the legislation limits for agricultural soils for Cd and Pb, with 1.6 and 487 mg kg⁻¹ respectively. In all cultivars, soybeans accumulated Cd in its aerial organs, and it could be translocated to fruits. Cd concentration in seeds ranged between 0.6 and 2.4 mg kg⁻¹, which exceed legislation limits and with toxicological risk to potential Chinese consumers. Pb levels were lower than Cd in seeds (0.03–0.17 mg kg⁻¹), and the accumulation were concentrated in the vegetative organs, with 93% of the Pb incorporated. Moreover, pods accumulated 11 times more Pb than seeds, which suggests that they act as a barrier to the passage of Pb to their offspring. These results evidence that soybean can easily translocate Cd, but not Pb, to reproductive organs. No regular patterns were observed in relation to climatic influence on heavy metal uptake.

Dioxins and dioxin-like polychlorinated biphenyls (dl-PCBs) in fish fillet counteract the health benefits of fish products. In this study, food waste was used as a protein alternative to replacing fishmeal commonly used in the commercial fish feed, aiming to cultivate Sabah grouper with acceptable levels of dioxins and dl-PCBs. The concentrations of dioxins and dl-PCBs, as well as the fish growth performance, were compared between the fish groups fed with food waste-based feed (FWBF) and commercial feed (Nanyu®, control). The results showed that the concentrations of polychlorinated dibenzo-p-dioxins (PCDDs) (1.22 pg/g dry weight (d.w.)) and non-ortho-dl-PCBs (13.0 pg/g d.w.) were significantly lower (p < 0.05) in the FWBF than in the control feed (commercial feed) (PCDDs: 2.35 pg/g d.w.; non-ortho-dl-PCBs: 27.2 pg/g d.w.). The growth performance of the fish group fed with the FWBF was comparable to that fed with the control feed. There were no significant differences between the WHO₂₀₀₅-TEQ values of different fish fillets (1.00, 1.11, and 1.10 pg WHO₂₀₀₅-TEQ/g d.w. for FWBF group, control feed group, and local market fish, respectively). Based on the guidelines provided by European Food Safety Authority (ESFA) and U.S. Environmental Protection Agency (USEPA), the fish fed with the FWBF were safe for human consumption (hazard index values: 0.284–0.522; cancer risk range: 2.59–2.97 × 10⁻⁵). The findings of this study suggest that food waste could serve as an alternative protein source for cultivating Sabah grouper with acceptable levels of dioxins and dl-PCBs.
