Deposition, depletion, and potential bioaccumulation of bisphenol F in eggs of laying hens after consumption of contaminated feed

Increasing concerns over bisphenol A (BPA) as an endocrine disrupting chemical (EDC) and its adverse effects on both humans and animals have led to the substitution by structural analogs, such as bisphenol F (BPF), in many application areas. Information regarding to the carry-over of this emerging chemical in farm animals is essential for legislation and risk assessment purposes. In this study, a large-scale number of animal experiments were designed to investigate the transfer of BPF from feed to eggs. One control and three experimental groups of laying hens (72 hens per group) were fed with basal diets and BPF-contaminated feed at concentration levels of 0.1, 0.5 and 2.5 mg kg⁻¹, respectively, for two weeks. The hens were then fed with BPF-free diets for a further four weeks. Eggs were collected daily, and separated into egg yolk and white for BPF analysis. The effects of different levels of BPF exposure on laying performance followed a non-monotonic dose-response curve, since low level BPF (0.1 mg kg⁻¹) exposure did increase the laying rate, mean egg weight and daily feed intake, while high level BPF (2.5 mg kg⁻¹) exposure showed a decreasing trend. BPF residues were detected in both egg yolks and whole eggs after two days of administration, and plateau phase was achieved within 9–18 days. There are clear linear dose-response relationships between the plateau BPF concentrations in feed and eggs. The residue of BPF was found mainly in egg yolks with conjugated form and depleted slowly (still detected 21 days after feeding the BPF-free diet of the high level group). Mean carry-over rate of 0.59% BPF from feed to eggs was obtained. Compared with the carry-over rates of PCBs and dioxins, BPF showed a relatively minor trend of bioaccumulation in eggs. To the best of our knowledge, this is the first report on the deposition, depletion, and bioaccumulation study of bisphenols in farm animals. The quantity of data can therefore be helpful in the frame of risk assessment, especially for a comprehensive estimation of consumer exposure to the residues of bisphenols.

High levels of polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) were detected in free-range eggs, and these levels reached a concentration of 29.84 ± 7.45 pg of WHO-TEQ/g of fat. This value exceeded the EU maximum permitted level of 2.5 pg of WHO-TEQ/g of fat for PCDD/F congeners by twelve-fold. A chemical analysis (HRGC-HRMS) revealed elevated amounts of OCDD, OCDF, HxCDD, HpCDD and HpCDF. During the investigation, samples of feed, soil, wall scrapings, wooden ceiling of the henhouse and tissues from laying hens were examined for dioxin contents (30 samples altogether). The long and complicated investigation found that the source of dioxins in the poultry farm was pentachlorophenol-treated wood, which was used as structural components in the 40-year-old farm building adapted to a henhouse. The wooden building material contained PCDD/Fs at a concentration of 3922.60 ± 560.93 pg of WHO-TEQ/g and 11.0 ± 2.8 μg/kg of PCP. The potential risk associated with dioxin intake was characterized by comparing the theoretically calculated weekly and monthly intakes with the toxicological reference values (TRVs), namely the Tolerable Weekly Intake (TWI) and Provisional Tolerable Monthly Intake (PTMI) values of 14 pg of WHO-TEQ/kg of bw and 70 pg of WHO-TEQ/kg of bw, respectively. The intake of dioxins estimated for high egg consumers (approximately 5–6 eggs/week) exceeded the TWI and PTMI values, which may pose a risk of delayed adverse health effects. The estimated dose of PCDD/Fs and DL-PCBs for children consuming 5 eggs per week exceeded the TWI by as much as 450% because of their nearly 5-fold-lower body weight. Although the dioxin intake estimated for the average consumption of eggs in the general population did not exceed any of the TRVs applied (58.7% TWI and 51.1% PTMI), such a situation should be considered unacceptable from a public health perspective because eggs are not the only source of these contaminants.

Bacillus coagulans is regarded as a clean, safe and helpful probiotic additive in the production of livestock and poultry breeds. Some studies have also shown that Bacillus coagulans can adsorb heavy metals in water, even in the gut of animals. However, whether Bacillus coagulans feeding influences antibiotic resistance gene (ARG) abundance in the gut of lead-exposed laying hens is unknown. To better apply such probiotics in the breeding industry, the present study employed Bacillus coagulans R11 and laying hens in model experiments to test ARG changes in the cecum of laying hens under lead exposure and B. coagulans R11 feeding. The results showed that there was the trend for ARG abundance decreasing in feeding B. coagulans R11 without lead exposure to laying hens in the cecum; however, feeding B. coagulans R11 to laying hens exposed to lead obviously increased the abundances of aminoglycoside and chloramphenicol ARGs. Further experiment found that hydroquinone, dodecanedioic acid, gibberellin A14, alpha-solanine, jasmonic acid and chitin were involved in the abundances of ARGs in the cecum, in addition the abundances of these compounds were also significantly enhanced by lead exposure or combination effects of lead and B. coagulans R11. As a result, the ARG hazards increased with feeding B. coagulans R11 to laying hens exposed to lead, and the key compounds which influenced by the combination effects of lead and B. coagulans R11 might influence the ARGs abundance.

Poultry manure is a reservoir for antibiotics and antibiotic resistance genes and composting is an effective biological treatment for manure. This study explored the effect of using two methods of adding a complex microbial agent to the composting of laying-hen manure on doxycycline degradation and tetracycline resistance genes elimination. The results showed that incorporating a complex microbial agent at 0.8% (w/w) on the 0ᵗʰ and 11th day (group MT2) effectively degraded doxycycline with a final degradation rate of 46.83 ± 0.55%. The half-life of doxycycline in this group was 21.90 ± 0.00 days and was significantly lower than that of group MT1 (1.6% (w/w) complex microbial agent added on the 0ᵗʰ day) and group DT (compost without complex microbial agent). But there was no significant difference in the final degradation rate of doxycycline between group DT and group MT1. The addictive with the complex microbial agent changed the microbial community structure. Bacteroidetes, Firmicutes and Proteobacteria were the dominant phyla during composting. Aerococcus, Desemzia, Facklamia, Lactobacillus, Streptococcus, and Trichococcus were the bacteria related to the degradation of doxycycline. Moreover, the incorporation of a complex microbial agent could decrease the risk on spreading tetracycline resistance genes. The single addition promoted the elimination of tetM, whose possible hosts were Enterococcus, Lactobacillus, Staphylococcus, and Trichococcus. Adding the complex microbial agent twice promoted the elimination of tetX, which was related to the low abundance of Chryseobacterium, Flavobacterium and Neptunomonas in group MT2. Redundancy analysis showed that the bacterial community, residual doxycycline and physiochemical properties have a potential effect on the variation in tetracycline resistance genes levels. Overall, adding the complex microbial agent twice is an effective measure to degrade doxycycline.

The effluent stream of the anaerobic digestion processes, the digestate, accommodates high residual organic content that needs to be further treated before discharge. Anaerobic treatment of digestate would not only reduce the residual organic compounds in digestate but also has a potential to capture the associated biogas. High-rate anaerobic reactor configurations can treat the waste streams using lower hydraulic retention times which requires less footprint opposed to the conventional completely stirred tank reactors. This study investigated the high-rate anaerobic treatment performance and the associated biogas capture from the digestate of a manure mixture composed of 90% laying hen and 10% cattle manures in fixed-film reactors. The results indicated that it was possible to reduce total chemical oxygen demand content of the digestate by 57–62% in 1.3–1.4 days of hydraulic retention time. The corresponding biogas yields obtained were in the range of 0.395–0.430 Lbiogas/g VSadded which were found to be comparable to many raw feedstocks. Moreover, significant total phosphorus reduction (36–47%) and greenhouse gas capture (over 14.5–18.1 tCO2e/d per m3 digestate) were also recorded in the anaerobic fixed-film reactors.

Due to greater consumption of poultry products and an increase in exports, more poultry houses will be needed. Therefore, it is important to investigate ways that poultry facilities can coexist in close proximity to residential areas without odors and environmental challenges. Ammonia (NH₃) is the greatest concern for environmental pollution from poultry production. When birds consume protein, they produce uric acid, ultimately converted to NH₃ under favorable conditions. Factors that increase production include pH, temperature, moisture content, litter type, bird age, manure age, relative humidity, and ventilation rate (VR). NH₃ concentration and emissions in poultry houses depend on VR; seasons also have effects on NH₃ production. Modern ventilation systems can minimize NH₃ in enclosed production spaces quickly but increase its emissions to the environment. NH₃ adversely affects the ecosystem, environment, and health of birds and people. Less than 10 ppm is the ideal limit for exposure, but up to 25 ppm is also not harmful. NH₃ can be minimized by housing type, aerobic and anaerobic conditions, manure handling practices, litter amendment, and diet manipulation without affecting performance and production. Antibiotics can minimize NH₃, but consumers have concerns about health effects. Administration of probiotics seems to be a useful replacement for antibiotics. More studies have been conducted on broilers, necessitating the need to evaluate the effect of probiotics on NH₃ production in conjunction with laying hen performance and egg quality. This comprehensive review focuses on research from 1950 to 2018.

This study investigated the effects and the possible mechanisms of intestinal content (IC) from laying hens on in vitro lead (Pb²⁺) biosorption of four probiotic bacterial strains (Bifidobacterium longum BB79, Lactobacillus paracasei Kgl6, Lactobacillus pentosus ITA23, and Lactobacillus acidipiscis ITA44). The total Pb²⁺ removal capacity of the four probiotic strains, with and without capsule polysaccharides (CPSs), increased in the presence of IC compared to the control (without IC). SEM imaging revealed certain unidentified particles from the IC adhered on the surface of bacterial cells sorted out using flow cytometry. Follow-up experiment showed an overall trend of increase in the Pb²⁺ removal capacity of the sorted bacteria, but statistically significant for L. pentosus ITA23 and B. longum BB79 after incubation with IC, particularly with the suspended solid portion of the IC. In addition, the Fourier transform infrared spectrophotometer data showed that functional groups such as C-H, O-H, C=O, and C-O-C which possibly associated with Pb²⁺ binding were mainly presented in the suspended solid portion of IC. Putting the above together, we postulated that the enhanced Pb²⁺ binding capacity the probiotic bacteria incubated in IC is due to the adherence of the yet to be identified particles which could much exist in suspended solid portion of IC containing negatively charged functional groups which bind with the positive Pb²⁺ ions.

The present study was designed to alleviate the negative biohazards of high ambient temperature on the productive performance and physiological status of laying hens. A total of 135 Bovans laying hens were distributed into nine groups in a 3 × 3 factorial design experiment. Basal diet was supplemented with vitamin E at levels of 0, 250, and 500 mg /kg diet. Within each dietary vitamin E level, each diet was supplemented with sodium selenite as a source of selenium (Se) to supply 0, 0.25, and 0.50 mg Se/kg diet. Results showed that supplementing layer’s diet with 500 mg vitamin E/kg was accompanied with the lowest feed consumption (FC) and feed conversion ratio (FCR). The interaction among vitamin E and Se levels exerted significant effects only on FC and FCR. Insignificant differences were observed in egg quality criteria due to the treatments studied. Increasing vitamin E level was associated with a gradual decrease in basophil count and an increase in monocytes. A gradual decrease in the count of each of heterophils, monocytes, and eosinophils was observed with the elevation in the dietary Se level. The combination among vitamin E and Se levels produced a significant effect on all hematological parameters studied. As vitamin E increased, a marked decrease in serum AST and a gradual increase in total lipids, total cholesterol, and calcium were observed. As the level of dietary Se increased, serum total protein, albumin, T₄, total cholesterol, and total lipids increased. No significant impacts were detected for the interaction among vitamin E and Se levels on any of blood constituents determined except serum globulin, ALT, and calcium. In conclusion, the combination between vitamin E and Se showed a good ability to alleviate the harmful impacts of heat stress and produced the highest productive performance when compared with the other groups, which exhibit the synergistic effect between the two antioxidants.

The objective of the current study was to examine the effect of the inclusion levels of distiller’s dried grains with solubles (DDGS) which partially and fully replaced soybean meal (SBM) in laying hen diets with or without enzyme or vitamin E supplementation on nutrients digestibility and excreted nitrogen and phosphorous. For these purposes, a total of 180 Hisex Brown laying hens at 22 weeks of age were randomly distributed into 15 treatment groups in a 5 × 3 factorial design experiment including five levels of DDGS (0, 5.5, 11, 16.5 and 22% replaced for the same percent of SBM which represents 0, 25, 50, 75 and 100% respectively in the diet) and three levels of feed additives (without, 250 mg enzyme/kg diet or 200 mg vitamin E/kg diet) through 22–42 weeks of age. Results revealed that digestion coefficient values of nutrients including DM, OM, CP, EE, NFE and CF were more preferable in hens fed diets involved 25% DDGS instead of SBM than that of the control diet and other treatment groups. In contrast, hens fed 100% substitution of DDGS caused significant (P ≤ 0.01) decrease in digestibility coefficients of all nutrients. The amount of daily excreted N was decreased in hens fed 25 or 50% DDGS substitution, whilst feeding 75 or 100% DDGS led to an elevation in N excretion. The proportion of excreted phosphorous was statistically (P ≤ 0.01) decreased as the level of DDGS increased. Generally, neither enzyme nor vitamin E supplementation had any significant effect on the studied parameters. It could be concluded that DDGS could be used up to 50% as a replacement of SBM (11% of the diet) without any adverse effects on nutrients digestibility. Laying hen farms using DDGS up to 25 or 50% substituted for SBM in its diets (5.5 or 11% of the diet) will have clearer environment of nitrogen and phosphorous pollution than others whose diets free of DDGS.

The present study aimed to investigate the effect of the dietary levels of protein, methionine (Met), as well as probiotic on productive performance, feed utilization, and environmental pollution by N in Lohmann Brown laying hens. A total number of 160 Lohmann Brown laying hens at 20 weeks of age were randomly divided into 8 treatment groups using a 2 × 2 × 2 factorial design experiment. The experiment involved two levels of crude protein (16 and 18 %), two levels of Met (0.45 and 0.50 %), and two concentrations of probiotic (0 or 1 g/kg diet, with a concentration of 10¹⁰ CFU/g of Lactobacillus acidophilus) within 20–42 weeks of age. Results revealed that egg production parameters were significantly (P < 0.01) improved for hens fed diets of 18 % CP comparing with that of 16 % protein within the period from 26 to 30 weeks of age. Protein utilization and feed efficiency values were enhanced with 0.67 and 0.72 % Met during the period of 26–30 weeks of the age. For the N pollution, results showed that increasing crude protein in the diet from 16 to 18 % caused significant (P < 0.01) increase in the excreted N from 0.349 to 0.492 g/d. The methionine level of 0.72 % recorded the highest values of total consumed N being 3.98 g/d and excreted N being 0.527 g/d comparing with the other levels. It could be concluded that the best productive performance could be given by using 0.72 % total sulfur amino acids (TSAA). Furthermore, the dietary level of 18 % CP with 0.72 % Met is preferred in feeding laying hens through the whole experimental period. Ecologically, reducing the level of crude protein in layer diets to be 16 % along with the supplementation of Met can play an important role in minimizing the pollution with N from poultry excretion.
