Arsenic (As) toxicity is a problem that needs to be solved in terms of both human health and agricultural production in the vast majority of the world. The presence of As causes biomass loss by disrupting the balance of biochemical processes in plants and preventing growth/water absorption in the roots and accumulating in the edible parts of the plant and entering the food chain. A critical method of combating As toxicity is the use of biosafe, natural, bioactive compounds such as hesperidin (HP) or chlorogenic acid (CA). To this end, in this study, the physiological and biochemical effects of HP (100 μM) and CA (50 μM) were investigated in Zea mays under arsenate stress (100 μM). Relative water content, osmotic potential, photosynthesis-related parameters were suppressed under stress. It was determined that stress decreased the activities of the antioxidant system and increased the level of saturated fatty acids and, gene expression of PHT transporters involved in the uptake and translocation of arsenate. After being exposed to stress, HP and CA improved the capacity of superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), glutathione S-transferase (GST) and glutathione peroxidase (GPX) and then ROS accumulation (H₂O₂) and lipid peroxidation (TBARS) were effectively removed. These phenolic compounds contributed to maintaining the cellular redox status by regulating enzyme/non-enzyme activity/contents involved in the AsA-GSH cycle. HP and CA reversed the adverse effects of excessive metal ion accumulation by re-regulated expression of the PHT1.1 and PHT1.3 genes in response to stress. Exogenously applied HP and CA effectively maintained membrane integrity by regulating saturated/unsaturated fatty acid content. However, the combined application of HP and CA did not show a synergistic protective activity against As stress and had a negative effect on the antioxidant capacity of maize leaves. As a result, HP and CA have great potentials to provide tolerance to maize under As stress by reducing oxidative injury and preserving the biochemical reactions of photosynthesis.

The toxicity of nanoplastics to aquatic organisms has been widely studied in terms of biochemical indicators. However, there is little discussion about the underlying toxic mechanism of nanoplastics on microalgae. Therefore, the chronic effect of polystyrene (PS) nanoplastics (80 nm) on Chlorella pyrenoidosa was investigated, in terms of responses at the biochemical and molecular/omic level. It was surprising that both inhibitory and promoting effects of nanoplastcis on C. pyrenoidosa were found during chronic exposure. Before 13 days, the maximum growth inhibition rate was 7.55% during 10 mg/L PS nanoplastics treatment at 9 d. However, the inhibitory effect gradually weakened with the prolongation of exposure time. Interestingly, algal growth was promoted for 1–5 mg/L nanoplastics during 15–21 d exposure. Transcriptomic analysis explained that the inhibitory effect of nanoplastics could be attributed to suppressed gene expression of aminoacyl-tRNA synthetase that resulted in the reduced synthesis of related enzymes. The promotion phenomenon may be due to that C. pyrenoidosa defended against nanoplastics stress by promoting cell proliferation, regulating intracellular osmotic pressure, and accelerating the degradation of damaged proteins and organs. This study is conducive to provide theoretical basis for evaluating the actual hazard of nanoplastics to aquatic organisms.

The present study was aimed at investigating the effects of different acids and pH neutralizers applied to dredged marine sediment for the treatment of heavy metals, and the resulting influence on the sediment quality as a plant growth medium. The inspection of barley germination in the dredged marine sediment revealed that residual salts are critical plant stressors whose adverse effects exceed those exhibited by high-level heavy metals and petroleum hydrocarbons present in the sediment. Acid washing and pH neutralization reduced not only the heavy metal contents but also the sediment salinity (by factors of 6.1–9.5), resulting in 100% germination of barley. For acid-washed and calcium-oxide-neutralized sediment, the barley growth was comparable to that observed in untreated and water washed sediment despite factors of 5.2–8.0 greater sediment salinity in the former. This result represents the protective effect of residual calcium against sodium and chloride toxicity. Water washing of acid-washed and pH-neutralized sediments further enhanced barley growth owing to the reduction in osmotic pressure. This study showed the effect of different sediment-washing reagents on the product quality. It also indicated the significance of balancing the enhancement of product quality and economic cost of further treatment requirements.

In this study, the tips of Myriophyllum aquaticum (M. aquaticum) plants were planted in open-top plastic bins and treated by simulated wastewater with various ammonium-N concentrations for three weeks. The contents of related carbohydrates and key enzyme activities of carbon metabolism were measured, and the mechanisms of carbon metabolism regulation of the ammonia tolerant plant M. aquaticum under different ammonium-N levels were investigated. The decrease in total nonstructural carbohydrates, soluble sugars, sucrose, fructose, reducing sugar and starch content of M. aquaticum were induced after treatment with ammonium-N during the entire stress process. This finding showed that M. aquaticum consumed a lot of carbohydrates to provide energy during the detoxification process of ammonia nitrogen. Moreover, ammonia-N treatment led to the increase in the activitives of invertase (INV) and sucrose synthase (SS), which contributed to breaking down more sucrose to provide substance and energy for plant cells. Meanwhile, the sucrose phosphate synthase (SPS) activity was also enhanced under stress of high concentrations of ammonium-N, especially on day 21. The result indicated that under high-concentration ammonium-N stress, SPS activity can be significantly stimulated by regulating carbon metabolism of M. aquaticum, thereby accumulating sucrose in the plant body. Taken together, M. aquaticum can regulate the transformation of related carbohydrates in vivo by highly efficient expression of INV, SPS and SS, and effectively regulate the osmotic potential, thereby delaying the toxicity of ammonia nitrogen and improving the resistance to stress. It is very important to study carbon metabolism under ammonia stress to understand the ammonia nitrogen tolerance mechanism of M. aquaticum.

Earthworms improve the soil fertility and they are also sensitive to soil contaminants. Earthworms (Eisenia fetida), standard reference species, were usually chosen to culture and handle for toxicity tests. Metabolic responses in earthworms exposed to 2, 2′, 4, 4′-tetrabromodiphenyl ether (BDE-47) and decabromodiphenyl ether (BDE-209) were inhibitory and interfered with basal metabolism. In this study, 1H-NMR based metabolomics was used to identify sensitive biomarkers and explore metabolic responses of earthworms under sub-lethal BDE-47 and BDE-209 concentrations for 14 days. The results revealed that lactate was accumulated in earthworms exposed to BDE-47 and BDE-209. Glutamate increased significantly when the concentration of BDE-47 and BDE-209 reached 10 mg/kg. The BDE-47 exposure above 50 mg/kg concentration decreased the content of fumarate significantly, which was noticed different from that of BDE-209. Whereas, the BDE-207 or BDE-209 exposure increased the protein degradation into amino acids in vivo. The increased betaine content indicated that earthworms may maintain the cell osmotic pressure and protected enzyme activity by metabolic regulation. Moreover, the BDE-47 and BDE-209 exposure at 10 mg/kg changed most of the metabolites significantly, indicating that the metabolic responses were more sensitive than growth inhibition and gene expression. The metabolomics results revealed the toxic modes of BDE-47 and BDE-209 act on the osmoregulation, energy metabolism, nerve activities, tricarboxylic acid cycle and amino acids metabolism. Furthermore, our results highlighted that the 1H-NMR based metabolomics is a strong tool for identifying sensitive biomarkers and eco-toxicological assessment.

Hexabromocyclododecane (HBCD), a ubiquitous suspected contaminant, is one of the world's most prominent brominated flame retardants (BFRs). In the present study, earthworms (Eisenia fetida) were exposed to HBCD. The expression of selected antioxidant enzyme genes was measured, and the metabolic responses were assessed using nuclear magnetic resonance (NMR) to identify the molecular mechanism of the antioxidant stress reaction and the metabolic reactions of earthworms to HBCD. A significant up-regulation (p < 0.05) of superoxide dismutase (SOD) gene expression was detected, with the highest gene expression level of SOD appearing at a dose of 400 mg kg⁻¹ dw (2.06-fold, p < 0.01). However, the glutathione transferase (GST) gene expression levels did not differ significantly (p > 0.05). Principal component analysis (PCA) of the metabolic responses showed that all groups could be clearly differentiated, and the highest concentration dose group was the most distant from the control group. Except for fumarate, the measured metabolites, which included adenosine triphosphate (ATP), valine, lysine, glycine, betaine and lactate, revealed significant (p < 0.05) increases after 14 days of exposure to HBCD. HBCD likely induces high levels of anaerobic respiration, which would result in high levels of ATP and lead to the disintegration of proteins into amino acids, including valine and lysine, to produce energy. The observed changes in osmotic pressure were indicative of damage to the membrane structure. Furthermore, this study showed that NMR-based metabolomics was a more sensitive tool than measuring the gene expression levels for elucidating the mode of toxicity of HBCD in earthworm exposure studies.

A Terrestrial Biotic Ligand Model (TBLM) for Ni toxicity to barley root elongation (RE) developed from experiments conducted in sand culture was used to predict toxicity in non-calcareous soils. Ca2+ and Mg2+ concentrations and pH in sand solution were varied individually and TBLM parameters were computed. EC50 increased as Mg2+ increased, whereas the effect of Ca2+ was insignificant. TBLM parameters developed from sand culture were validated by toxicity tests in eight Ni-amended, non-calcareous soils. Additional to Ni2+ toxicity, toxicity from all solution ions was modelled independently as an osmotic effect and needed to be included for soil culture results. The EC50s and EC10s in soil culture were predicted within twofold of measured results. These are close to the results obtained using parameters estimated from the soil culture data itself.

Serious environmental pollution of heavy metals has attracted people's attention in recent years and halophiles seem to be potential bioremediation in the controlling of heavy metals contamination. In this study, the adaptive mechanism of halophilic Brachybacterium muris (B. muris) in response to salt stress and its mitigation of copper (Cu) toxicity in hydroponic plants were investigated. The cell morphology was observed using transmission electron microscopy. The cell membrane composition and fluidity were examined by the combination of gas chromatography, gas chromatography-mass spectrometry, ultra-high performance liquid chromatography-mass spectrometry, and fluorescence spectrophotometry. Moreover, the metabolic pathways of B. muris in response to salt stress were analyzed using the prokaryotic transcriptomics approach. A hydroponic co-culture model was further conducted to explore the effects of B. muris on wheat seedlings subjected to Cu toxicity. It was found that B. muris can respond to high osmotic pressure by improving the cell membrane fluidity, altering the cell morphology and cell membrane compositions. The proportion of unsaturated fatty acids, phosphatidylethanolamine, and phosphatidylinositol in B. muris cell membranes increased significantly, while zymosterol, fecosterol, and ergosterol contents decreased under a high salinity situation. Further transcriptomic analysis showed that genes encoding L-glutamate synthase, glutamate ABC transporter ATP-binding protein, and sodium cotransporter were up-regulated, indicating that both the synthesis and transport of glutamate were significantly enhanced under high osmotic pressure. Additionally, B. muris alleviated the inhibitory effect of Cu²⁺ on wheat seedlings' growth, causing a 30.14% decrease in H₂O₂ content and a significant increase of 83.86% and 45.96% in POD activity and GSH content in wheat roots, respectively. The findings of this study suggested that the salt-tolerant B. muris may serve as a promising strategy for improving the bioremediation of metal-contaminated saline water and soils.

The negative effects of pollution on amphibians are especially high when animals are additionally stressed by other environmental factors such as water salinity. However, the stress provoked by salinity may vary among populations because of adaptation processes. We tested the combined effect of a common fertilizer, ammonium nitrate (0–90.3 mg N–NO3NH4/L), and water salinity (0–2‰) on embryos of two Pelophylax perezi populations from ponds with different salinity concentrations. Embryos exposed to the fertilizer were up to 17% smaller than controls. Survival rates of embryos exposed to a single stressor were always below 10%. The exposure to both stressors concurrently increased mortality rate (>95%) of embryos from freshwater. Since the fertilizer was lethal only when individuals were stressed by the salinity, it did not cause lethal effects on embryos naturally adapted to saline environments. Our results underscore the importance of testing multiple stressors when analyzing amphibian sensitivity to environmental pollution. Natural resistance to salinity minimizes the impact of chemical fertilizers on amphibian embryos.

Anthropogenic stressors can have an impact in a broad range of physiological processes and can be a major selective force leading to rapid evolution and local population adaptation. In this study, three populations of the invasive crayfish Procambarus clarkii were investigated. They are geographically separated for at least 20 years, and live in different abiotic environments: a freshwater inland lake (Salagou lake) with no major anthropogenic influence and two other coastal wetlands regularly polluted by pesticides along the Mediterranean coast (Camargue region and Bages-Sigean lagoon). Collected adults were genetically characterized using the mitochondrial COI gene and haplotype frequencies were analyzed for genetic variability within and between populations. Results revealed a higher genetic diversity for these invasive populations than any previous report in France, with more than seven different haplotypes in a single population. The contrasting genetic diversity between the Camargue and the other two populations suggest different times and sources of introduction. To identify differences in key physiological responses between these populations, individuals from each population were maintained in controlled conditions. Data on oxygen consumption rates indicate that the Salagou and Bages-Sigean populations possess a high inter-individual variability compared to the Camargue population. The low individual variability of oxygen consumption and low genetic diversity suggest a specific local adaptation for the Camargue population. Population-specific responses were identified when individuals were exposed to a pesticide cocktail containing azoxystrobin and oxadiazon at sublethal concentrations. The Salagou population was the only one with altered hydro-osmotic balance due to pollutant exposure and a change in protease activity in the hepatopancreas. These results revealed different phenotypic responses suggesting local adaptations at the population level.
