Physalis pubescens L. alleviates testicular disruptions associated with streptozotocin-induced diabetes in male Wistar rats, Rattus norvegicus

Testicular impairment is a serious complication of diabetes that is mediated by oxidative stress and inflammation. Physalis has antioxidative and anti-inflammatory actions. Thus, the present study investigated the ameliorative role of Physalis juice (PJ) prepared from the fruits against testicular damages in streptozotocin (STZ)-induced diabetic rats. Adult male Wistar rats were divided randomly into five groups (n=6): control, orally administered 5 mL PJ/kg daily (PJ), injected intraperitoneally with a single dose of 55 mg STZ/kg without treatment (STZ), or treated daily with PJ (STZ+PJ) or with 500 mg metformin/kg (STZ+Met), for 28 days. The STZ group showed a marked elevation in the blood glucose level by 230%, whereas remarkable declines in the serum levels of testosterone (44%), follicle-stimulating hormone (FSH) (48%), and luteinizing hormone (LH) (36%), as compared to controls. In comparison to controls, the testis of the STZ group showed remarkable declines in the testis weight (15%), the glutathione (GSH) content (45%), mRNA and protein levels of B-cell lymphoma-2 (Bcl-2) (48 and 35%), mRNA and activities of superoxide dismutase (SOD) (63 and 40%), catalase (CAT) (56 and 31%), glutathione peroxidase (GPx) (51 and 44%), and glutathione reductase (GR) (62 and 43%), whereas marked elevations in the levels of interleukin-1 beta (IL-1β (169%), tumor necrosis factor-alfa (TNFα) (85%), nitric oxide (NO) (96%), malondialdehyde (MDA) (83%), mRNA and protein levels of Bcl-2-associated X protein (Bax) (400 and 61%), and mRNA level of caspase-3 (Cas-3) (370%). Some histopathological alterations were observed in the testicular tissue of the STZ group. In contrast, PJ markedly alleviated all the abovementioned disturbances. In conclusion, PJ at a dose of 5 mL/kg attenuated the diabetes-associated testicular impairments, which may be due to its antioxidative, anti-inflammatory, and antiapoptotic actions.

Bisphenol A (BPA) received heightened attention in the recent years due to humans continuously being exposed to it. This study explores the effect of taurine or curcumin on subacute BPA treatment-induced nephrotoxicity in rats (Rattus norvegicus). Forty-two adult albino male rats were exposed to BPA (130 mg/kg daily) for 28 days by gastric gavage. BPA led to lipid peroxidation, inhibiting antioxidant enzyme activities like catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and glutathione S-transferase (GST). BPA exposure also induced histopathological changes like tubular and glomerular degeneration, vascular congestion, and interstitial cell infiltration in kidney tissue. Cotreatment with taurine (100 mg/kg daily) or curcumin (100 mg/kg daily) alleviated the lipid peroxidation level and antioxidant enzyme activities and histological alterations brought about by BPA. In this study, curcumin and taurine application provided protection against renal toxicity caused by BPA but did not prevent toxic effect completely.

Metal-oxide nanoparticles (NPs), as a new emerging technological compound, promise a wide range of usage areas and consequently have the potential to cause environmental toxicology. In the present work, aluminum (Al₂O₃), copper (CuO), and titanium (TiO₂) nanoparticles (NPs) were administered via oral gavage to mature female rats (Rattus norvegicus var. albinos) for 14 days with a dose series of 0 (control), 0.5, 5, and 50 (mg/kg b.w./day). Enzyme activities of the antioxidant system such as catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione S-transferase (GST), and glutathione reductase (GR) in the liver were measured. Transmission electron microscope (TEM) images of the liver were taken to demonstrate NP accumulation and distribution in liver tissue. Data showed that all NPs caused some significant (P > 0.05) alterations in the activities of antioxidant enzymes. CAT activity increased after CuO and TiO₂ administrations, while SOD activity decreased after Al₂O₃ administration. The activities of enzymes associated with glutathione (GR, GPx, GST) metabolisms were also significantly altered by NPs. GPx activity increased in rats received Al₂O₃, CuO NPs, while GR activity increased only by Al₂O₃. However, there were increases (TiO₂) and decreases (CuO) in GST activity in the liver of rats. TEM images of the liver demonstrated that all NPs accumulated in the liver, even at the lowest dose. This study indicated that the antioxidant enzymes in the liver of rats were affected by all NPs, suggesting the antioxidant system of rats suffered after NP administration.

Pesticides can cause oxidative stress resulting to deleterious effects in animal metabolisms. Cyfluthrin is a synthetic pyrethroid used worldwide to protect crops and to eliminate pests. Thus, the aim of this study was to investigate the effects of the cyfluthrin on the level of malondialdehyde (MDA) and activities of catalase (CAT), glutathione peroxidase (GPx), and acetylcholinesterase (AChE) in the liver and kidney of Wistar Albino Sprague Dawley rats (Rattus norvegicus var. albinos) following intraperitoneal treatment of cyfluthrin (1.2, 12, and 120 mg/kg b.w./day) for 21 days. Comparisons were made with two control solutions named as serum physiologic and solvent in which cyfluthrin was dissolved. CAT activity in the liver and kidney of rats did not change after the lowest cyfluthrin treatment, while its activity significantly decreased at the higher doses. In general, cyfluthrin significantly decreased the activity of GPx in the liver and kidney at all doses, while MDA levels in the liver increased at all doses. Cyfluthrin significantly decreased AChE activity in the liver of rats at all doses, while this was true at the highest dose for the kidney. This study showed that the studied biomarkers were effective in determining the toxic effects of cyfluthrin. Thus, they should be used to monitor pesticide-affected areas before untargeted animals, including humans who suffer from the use of pesticides.

Rodents infest urban environments, causing damage and acting as vectors for disease transmission. Currently, anticoagulants are the most widely used chemical rodenticides, and their extensive and widespread use can contaminate the environment. To ensure effectiveness and avoid accumulation of rodent baits in the environment, it is important to evaluate how long rodent baits maintain their palatability and efficacy. In rodent control programs, rodent baits are placed in locations such as sewers, but after a few days, baits appear altered, causing doubts about the control efficacy. For this reason, baits are replaced periodically, which increases costs and generation of chemical waste. The objective of this study was to evaluate the palatability and efficacy of commercial paraffin-type rodent bait blocks placed in sewers in São Paulo City over a period of 90 days. Bait blocks were placed in sewers and collected after 30, 60, and 90 days. Additionally, in a laboratory two-choice test, wild-caught urban Norway rats were offered 40–60 g of bait and an equal volume of standard rat pellets. The amount of bait and rat pellet consumed was registered, the palatability was calculated, and the efficacy was measured as the percentage mortality over 14 days. The results showed that, even when they had an altered appearance, bait blocks remained palatable to the rats and were effective after at least 90 days. Leaving bait blocks for longer periods could be an effective strategy for reducing costs and could help to ensure the control of urban rodents in an environmentally sustainable way.

The main objective of this study was to determine how rat tapeworms affect the excretion of zinc and cadmium through rat feces. Male rats (Rattus norvegicus var. alba) were divided into four groups, and the experiment was conducted over a 6-week period. The control groups (00; 0T) were provided with a standard ST-1 rodent mixture and received 10.5 mg of Zn/week. Groups P0 and PT were fed a mixture supplemented with the hyperaccumulating plant Arabidopsis halleri at a dosage of 123 mg Zn/week and 2.46 mg Cd/week. Groups 0T and PT were infected with the rat tapeworm (Hymenolepis diminuta). Fecal samples were collected 24 h post exposure. Zinc and cadmium concentrations in rat feces were analyzed using inductively coupled plasma optical emission spectrometry. Tapeworm presence decreased the amount of metals excreted through the feces of the host throughout the entire experiment, with the exception of 1 week (control group). No statistically significant differences between zinc excretion rates in the control groups (00 and 0T) were detected at any time throughout the experiment. A statistically significant difference between zinc excretion rates (p < 0.05) in the exposed groups (P0 and PT) was detected in 2 of the 6 monitored weeks. Group PT excreted significantly less cadmium (p < 0.01) than group P0 did in three of the 6 weeks. Overall, our results indicate that tapeworms are able to influence the excretion of metals by their host. Tapeworms accumulate metals from intestinal contents. It is not clear whether tapeworms carry out this process before the host tissues absorb the metals from the intestines or the tapeworms accumulate metals excreted from the body of the host back to the intestines. Most likely, it is a combination of both phenomena.

Pesticides are used to protect crops and to eliminate pests, though non-target organisms such as mammals are also affected from their usage. Etoxazole (organoflourine pesticide) is an acaricide used to combat spider mites which are the parasites of various crops. The present study aims to investigate the effects of etoxazole on the level of MDA (malondialdehyde) and activities of CAT (catalase), GPx (glutathione peroxidase), and AChE (acetylcholinesterase) in liver and kidney tissues of Wistar rats (Rattus norvegicus var. albinos). Rats received etoxazole intraperitoneally with doses of 2.2, 11, and 22 mg/kg b.w./day for 21 days. Control rats received the same volume of the serum physiologic. Following etoxazole exposures, activities of CAT, GPx, and AChE in the liver and kidney of rats significantly decreased at all doses compared to control group. Oppositely, MDA levels in these tissues increased significantly at all doses following etoxazole exposures. The present study demonstrated that etoxazole, at all doses, had toxic effects in the liver and kidney parameters, suggesting their possible use as effective biomarkers in determining the toxic effects of etoxazole. This may suggest that these biomarkers could also be used as a tool to monitor pesticide-affected areas before severe toxic effects begin in non-target animals and humans.
