Inoculation of soil or seeds with plant growth promoting bacteria ameliorates metal toxicity to plants by changing metal speciation in plant tissues but the exact location of these changes remains unknown. Knowing where the changes occur is a critical first step to establish whether metal speciation changes are driven by microbial metabolism or by plant responses. Since bacteria concentrate in the rhizosphere, we hypothesised steep changes in metal speciation across the rhizosphere. We tested this by comparing speciation of zinc (Zn) in roots of Brassica juncea plants grown in soil contaminated with 600 mg kg⁻¹ of Zn with that of bulk and rhizospheric soil using synchrotron X-ray absorption spectroscopy (XAS). Seeds were either uninoculated or inoculated with Rhizobium leguminosarum bv. trifolii and Zn was supplied in the form of sulfide (ZnS nanoparticles) and sulfate (ZnSO₄). Consistent with previous studies, Zn toxicity, as assessed by plant growth parameters, was alleviated in B. juncea inoculated with Rhizobium leguminosarum. XAS results showed that in both ZnS and ZnSO₄ treatments, the most significant changes in speciation occurred between the rhizosphere and the root, and involved an increase in the proportion of organic acids and thiol complexes. In ZnS treatments, Zn phytate and Zn citrate were the dominant organic acid complexes, whilst Zn histidine also appeared in roots exposed to ZnSO₄. Inoculation with bacteria was associated with the appearance of Zn cysteine and Zn formate in roots, suggesting that these two forms are driven by bacterial metabolism. In contrast, Zn complexation with phytate, citrate and histidine is attributed to plant responses, perhaps in the form of exudates, some with long range influence into the bulk soil, leading to shallower speciation gradients.

Studies in the literature concern the toxicity of nanoparticles either in a Petri dish or in agar media-based tests. Therefore, for environmental relevance, individual and binary mixtures of metal oxide nanoparticles (M-NPs) cadmium oxide (CdO-NP) and copper oxide (CuO-NP) were tested in this study for their effect on Vigna radiata in soil with and without the addition of sawdust. Seed germination was 67% in 100 mg CuO-NP in soil without sawdust. Seeds failed to germinate in 100 mg CdO +100 mg CuO-NPs in soil without the addition of sawdust and germination was 83% at the same concentration in soil with sawdust. In sawdust added to soil, when compared with control (soil without M-NPs), the maximum reduction in shoot (82%) and root (80%) length and wet (61%) and dry (54%) weight of plant was recorded in CdO-NP treated soil. Similarly, compared with control (soil without sawdust and M-NPs), the percent reduction in shoot (61%) and root (70%) length and wet (44%) and dry (48%) weight was highest in CdO-NP treated soil not supplemented with sawdust. In a binary mixture test (CdO-NP + CuO-NP), the addition of sawdust promoted the above plant growth parameters compared with individual CdO-NP and CuO-NP tests. Cadmium (511 mg kg⁻¹ for individual and 303 mg kg⁻¹ for binary mixture tests) and Cu (953 mg kg⁻¹ for individual and 2954 mg kg⁻¹ for binary mixture tests) accumulation was higher in plants grown in soil without sawdust. The beneficial effect of sawdust addition was observed in seed germination, plant growth, and metal accumulation. With or without sawdust, the binary mixture of CdO and CuO was antagonistic. These results indicate that sawdust can prevent M-NP-induced toxicity and reduce metal accumulation in plant tissues.

Since the Industrial Revolution, the global ambient O3 concentration has more than doubled. Negative impact of O3 on some common crops such as wheat and soybeans has been widely recognized, but there is relatively little information about maize, the typical C4 plant and third most important crop worldwide. To partly compensate this knowledge gap, the maize cultivar (Zhengdan 958, ZD958) with maximum planting area in China was exposed to a range of chronic ozone (O3) exposures in open top chambers (OTCs). The O3 effects on this highly important crop were estimated in relation to two O3 metrics, AOT40 (accumulated hourly O3 concentration over a threshold of 40 ppb during daylight hours) and POD6 (Phytotoxic O3 Dose above a threshold flux of 6 nmol O3 m−2 s−1 during a specified period). We found that (1) the reduced light-saturated net photosynthetic rate (Asat) mainly caused by non-stomatal limitations across heading and grain filling stages, but the stomatal limitations at the former stage were stronger than those at the latter stage; (2) impact of O3 on water use efficiency (WUE) of maize was significantly dependent on developmental stage; (3) yield loss induced by O3 was mainly due to a reduction in kernels weight rather than in the number of kernels; (4) the performance of AOT40 and POD6 was similar, according to their determination coefficients (R2); (5) the order of O3 sensitivity among different parameters was photosynthetic parameters > biomass parameters > yield-related parameters; (6) Responses of Asat to O3 between heading and gran filling stages were significantly different based on AOT40 metric, but not POD6. The proposed O3 metrics-response relationships will be valuable for O3 risk assessment in Asia and also for crop productivity models including the influence of O3.

Bush bean (Phaseolus vulgaris) was exposed to atmospheric deposition of As, Cd and Pb in a polluted and a reference area. The atmospheric deposition of these elements was significantly related to the concentrations in leaves, stems and pods at green harvest. Surprisingly there was also a clear relation for As and Pb in the seeds at dry harvest, even though these seeds were covered by the husks. Root uptake of accumulated atmospheric deposits was not likely in such a short term experiment, as confirmed by the fact that soil pore water analysis did not reveal significant differences in trace element concentrations in the different exposure areas. For biomonitoring purposes, the leaves of bush bean are the most suitable, but also washed or unwashed pods can be used. This means that the obtained relationships are suitable to estimate the transfer of airborne trace elements in the food chain via bush bean.

Many paddy fields have been contaminated by mercury (Hg) in mining areas of China. In this study, twenty-six rice cultivars and three Hg contaminated paddy fields in different geographic regions were selected for field trials and aimed to investigate the variations and similarities in total Hg (THg) and methylmercury (MeHg) accumulations in brown rice (seeds) across sites. Our results revealed widescale cultivar variation in THg (13–52 ng g−1 at Wanshan) and MeHg (3.5–23 ng g−1) accumulation and %MeHg (17.7–89%) in seeds. The ability to translocate is an important factor in the levels of THg and MeHg in seed. Cultivar tended to stability in THg accumulation across sites. Some cultivars accumulated lower concentrations of both THg and MeHg in seeds at fields seriously contaminated by Hg. Present results suggest that appropriate cultivar selection is a possible way to reduce THg and MeHg accumulation in seeds of rice grown in Hg-contaminated regions.

Activated charcoal (AC) amendment has been suggested as a promising method to immobilize organic contaminants in soil. We performed pot experiments with rice and soybean grown in agricultural soil polluted by aromatic arsenicals (AAs). The most abundant AA in rice grains and soybean seeds was methylphenylarsinic acid (MPAA). MPAA concentration in rice grains was significantly reduced to 2% and 3% in 0.2% AC treated soil compared to untreated soil in the first year of rice cultivation. In the second year, MPAA concentration in rice grains was significantly reduced to 15% in 0.2% AC treated soil compared to untreated soil. MPAA concentration in soybean seeds was significantly reduced to 44% in 0.2% AC treated soil compared to untreated soil. AC amendment was effective in reducing AAs in rice and soybean.

Guidelines provided by the OECD and EPPO allow the use of single-species tests performed in greenhouses to assess the risk of herbicides to non-target terrestrial plant communities in the field. The present study was undertaken to investigate the use of greenhouse data to determine effects of herbicides with a different mode of action on the biomass, seed production and emergence of field-grown plants. In addition, a single species approach was compared with a mixed species approach. Effects on the biomass of greenhouse and field-grown plants were found to be related at different effect levels, indicating that it might be possible to translate results from greenhouse studies to field situations. However, the use of single-species tests may not be valid. The response of a single plant species to sublethal herbicide dosages differed to the response of the same species grown in a mixture with other species. The use of single-species greenhouse tests in the ecological risk assessment of crop protection products may only be valid for single species in the field, not for vegetations.

Remediation programmes are considered to be complete when human risk-based criteria are met. However, these targets are often unsatisfied with the ecological parameters that may be important with regard to future soil use. Five soil subsamples, collecting along a pilot-scale soil column after electrokinetic treatment, were studied, from which about 42.0%-93.3% soil Cu had been successfully removed. A series of biological assays including soil microbial biomass carbon, basal soil respiration, soil urease activity, earthworm assays, and seed assays were used to evaluate their ecological risks. The results showed that the bioassay data from the treatment variants did not supposedly reflecting the decreased soil Cu concentrations after the electrokinetic treatment, but were highly correlated with some soil physicochemical characteristics. It suggests that bioassays are necessary to assess the ecotoxicity of soil after electrokinetic treatment.

We studied the effects of long-term exposure (nine years) of birch (Betula papyrifera) trees to elevated CO(2) and/or O(3) on reproduction and seedling development at the Aspen FACE (Free-Air Carbon Dioxide Enrichment) site in Rhinelander, WI. We found that elevated CO(2) increased both the number of trees that flowered and the quantity of flowers (260% increase in male flower production), increased seed weight, germination rate, and seedling vigor. Elevated O(3) also increased flowering but decreased seed weight and germination rate. In the combination treatment (elevated CO(2)+O(3)) seed weight is decreased (20% reduction) while germination rate was unaffected. The evidence from this study indicates that elevated CO(2) may have a largely positive impact on forest tree reproduction and regeneration while elevated O(3) will likely have a negative impact.

Most available exposure–response relationships for assessing crop loss due to elevated ozone (O3) have been established using data from chamber and open-top chamber experiments, using a simulated constant O3 concentration exposure (square wave), which is not consistent with the diurnal variation of O3 concentration that occurs in nature. We investigated the response of oilseed rape (Brassica napus L.) to O3 as affected by two exposure regimes: one with a diurnal variation (CF100D) and another with a constant concentration (CF100). Although the two exposure regimes have the same mean O3 concentration and accumulated O3 concentration above 40 ppb (AOT40), our results show that O3 at CF100D reduced biomass and number of pods/plant more than O3 at CF100. Both O3 exposures resulted in larger seed weights/100 pods compared to CF. Numbers of seeds/100 pods were reduced by CF100, while numbers of seeds/100 pods in the CF100D chambers were comparable to those in CF. Our results suggest that chamber experiments that use a constant O3 exposure may underestimate O3 effects on biomass and yields. Diurnal variation of O3 concentration should be considered when designing O3 exposure experiment.
