Characterization of cadmium-resistant Klebsiella pneumoniae MCC 3091 promoted rice seedling growth by alleviating phytotoxicity of cadmium Full text

Cadmium (Cd) phytotoxicity in agricultural land is a major global concern now-a-days resulting in very poor yield. Plant growth-promoting rhizobacteria (PGPR)-mediated bioremediation is one of the convenient strategies for detoxification of Cd from the soil and for plant growth promotion under Cd stress. The selected strain K5 was identified as Klebsiella pneumoniae based on MALDI-TOF MS ribosomal protein and 16S rDNA sequence-based homology. The strain possessed several PGP traits viz. IAA production (3413 μg/mL), phosphate solubilization (80.25 ppm), ACC deaminase activity (40 ng α-ketobutyrate/mg protein/h), N₂ fixation ability (1.84 μg N₂ fixed/h), etc. and has the highest Cd resistance (4000 μg/mL) among Cd-resistant PGPR so far reported. This strain efficiently accumulated Cd and remained viable under Cd stress as confirmed by AAS-TEM-EDX analysis and viability test, respectively. The significant (p < 0.05) positive effect of the strain was reflected in various plant growth parameters like increased seed germination (50 to 90%), root length (5-fold), shoot length (about 2-fold), root fresh weight (> 2-fold), and shoot fresh weight (1.23-fold) under Cd stress compared with uninoculated set. Moreover, the positive impact of this strain on antioxidant enzyme activity (CAT, MDA, SOD) and several other biochemical parameters (proline, α-amylase, protease, total sugar, total protein, chlorophyll content) were also measured that favors plant growth promotion under Cd stress. Besides, the K5 strain also decreased stress-ethylene level under Cd stress and reduction of Cd accumulation in seedling (> 1.5-fold) was conducive to alleviate Cd phytotoxicity. Hence, K. pneumoniae strain K5 can be used as a phytostimulating and Cd-bioremediating biofertilizer for sustainable agriculture in heavy metal-contaminated sites.

An abandoned herbicide factory site was used as an example of how planning should be considered for development of the site for transportation use in Dalian, China. Exposure pathways and parameters for three types of transportation use (land for a traffic hub, land for an urban road, and land for a subway) were developed. Twenty-five sampling sites were selected and 38 soil samples were collected in March 2015. Hexachlorobenzene and benzo(a)pyrene which were extracted by Soxhlet extraction and detected by gas chromatography mass spectrometry were the most significant pollutants detected. The maximum concentration of the two pollutants in the surface layer (0–0.5 m) were 0.57 and 3.10 mg/kg, and in the bottom layer (1.0 m) were 2.57 and 3.72 mg/kg, respectively. In this study, risk assessment results based on the established exposure scenario and parameters showed that there was a significant difference in traffic hub land use under specific exposure pathway and common insensitive land use exposure pathways (direct ingestion of soil, dermal contact with soil, and inhalation of soil-derived dust). Commonly considered hexachlorobenzene and benzo(a)pyrene carcinogenic risk values exceeded the maximum acceptable level (10⁻⁶) and were found to be 23.9-fold and 189-fold higher than the carcinogenic risk values, respectively. Parameter sensitivity analysis data showed that for transportation use, the two parameters “EFOa” and “OSIRa” were the most significant factors associated with variation of the carcinogenic risk value. For traffic hub land use, urban road land use, and subway land use, the main exposure pathways were through “inhalation of soil vapors outdoors (from surface soil),” “direct ingestion of soil,” and “inhalation of soil vapors indoors (from bottom soil),” which contributed 84.75, 73.00, and 100.00% to the total risk value, respectively.

The present study aims to assess the removal of 3-amino-5-methylisoxazole (AMI), a recalcitrant by-product resulting from the biological breakdown of some pharmaceuticals, applying a solar photo-Fenton process assisted by ferrioxalate complexes (SPFF) (Fe³⁺/H₂O₂/oxalic acid/UVA-Vis) and classical solar photo-Fenton process (SPF) (Fe²⁺/H₂O₂/UVA-Vis). The oxidation ability of SPFF was evaluated at different iron/oxalate molar ratios (1:3, 1:6, and 1:9, with [total iron] = 3.58 × 10⁻² mM and [oxalic acid] = 1.07 × 10⁻¹, 2.14 × 10⁻¹ and 3.22 × 10⁻¹ mM, respectively) and pH values (3.5–6.5), using low iron contents (2.0 mg Fe³⁺ L⁻¹). Additionally, the use of other organic ligands such as citrate and ethylenediamine-N,N′-disuccinic acid (EDDS) was tested. The oxidation power of the classical SPF was assessed at different pH values (2.8–4.0) using 2.0 mg Fe²⁺ per liter. Furthermore, the effect of AMI concentration (2–20 mg L⁻¹), presence of inorganic ions (Cl⁻, SO₄ ²⁻, NO₃ ⁻, HCO₃ ⁻, NH₄ ⁺), and radical scavengers (sodium azide and D-mannitol) on the SPF method at pH 3.5 was also assessed. Experiments were done using a lab-scale photoreactor with a compound parabolic collector (CPC) under simulated solar radiation. A pilot-scale assay was conducted using the best operation conditions. While at near neutral pH, an iron/oxalate molar ratio of 1:9 led to the removal of 72 % of AMI after 90 min of SPFF, at pH 3.5, an iron/oxalate molar ratio of 1:3 was enough to achieve complete AMI degradation (below the detection limit) after 30 min of reaction. The SPF process at pH 3.5 underwent a slower AMI degradation, reaching total AMI degradation after 40 min of reaction. The scale up of SPF process showed a good reproducibility. Oxalic and oxamic acids were identified as the main low-molecular-weight carboxylic acids detected during the pilot-scale SPF reaction. Graphical abstract ᅟ

The Yellow River Delta is the most intact estuary wetland in China and suffers from great pressure of metals. Seventy-seven surface sediment samples were collected in the delta, and contents of Cu, Pb, Cd, Cr, Zn, Ni, and Mn were analyzed by inductively coupled plasma spectrometry and those of Hg and As by atomic fluorescence spectrometry. The results showed that means of metal contents (ppm, dry weight) were as follows: Hg, 0.04; Cr, 61.72; Cu, 20.97; Zn, 60.73; As, 9.47; Pb, 21.91; Cd, 0.12; Ni, 27.24; and Mn, 540.48. 43.8% of Hg and 14.3% of Cd were from the allogenic source while others from the authigenic source. The results of the geoaccumulation indexes appeared that 6.5% of sites from the estuarine and the Gudao areas were moderately polluted by Hg. All ecological risk index values of Hg and 37.7% of Cd were more than 40, which were the main factors of strongly and moderately potential ecological risks of 37.7% of sites in the delta. High Cd contents may be due to the alkaline conditions of the delta and the unreasonable management of the farmland, while the abnormal distribution of Hg to the wet or dry deposition and the erosion of the seawater. It was suggested to monitor Hg content in the atmosphere of the Yellow River Delta. The results were expected to update the pollution status of metals in the delta and created awareness of preserving the sound condition of the Yellow River Delta.

The important criteria in anaerobic digestion is the rate-limiting step which decides the fate of value-added products especially from waste-activated sludge (WAS). Hence, the present study investigated the effect of magnesium (Mg²⁺) addition on anaerobic digestion of WAS. The lab-scale experiments were conducted at 25 °C with Mg²⁺ doses ranging from 0.01 to 0.2 mol/L. Maximum total volatile fatty acids (VFAs) production (372.78 mg COD/L) occurred at a Mg²⁺ dose of 0.2 mol/L, which was about eight times higher than the control tests. Further, Mg²⁺ addition facilitated sludge dewaterability and phosphorus removal. The mechanism of improved VFAs generation was analyzed from the view of both chemical and biological effects. Chemical effect significantly enhanced the release of calcium and iron in WAS, resulting in the disintegration of WAS, which benefited hydrolysis and acidification processes. Illumina MiSeq sequencing analysis revealed that enrichment of functional bacteria and the increase of bacterial diversity were obtained in the 0.2 mol Mg²⁺/L experiment, while the influence was negative on the reactor with 0.025 mol/L Mg²⁺. Meanwhile, methanogens were accordantly inhibited in the experiments with Mg²⁺ addition.

This study assessed the biodegradation of mixtures of polycyclic aromatic hydrocarbons (PAHs) by indigenous bacteria in river sediment. Microcosms were constructed from sediment from the Chao Phraya River (the main river in Thailand) by supplementation with high concentrations of fluorene, phenanthrene, pyrene (300 mg kg⁻¹ of each PAH), and acenaphthene (600 mg kg⁻¹). Fluorene and phenanthrene were completely degraded, whereas 50% of the pyrene and acenaphthene were removed at the end of the incubation period (70 days). Community analyses revealed the dynamics of the bacterial profiles in the PAH-degrading microcosms after PAH exposure. Actinobacteria predominated and became significantly more abundant in the microcosms after 14 days of incubation at room temperature under aerobic conditions. Furthermore, the remaining PAHs and alpha diversity were positively correlated. The sequencing of clone libraries of the PAH-RHDα genes also revealed that the dioxygenase genes of Mycobacterium sp. comprised 100% of the PAH-RHDα library at the end of the microcosm setup. Moreover, two PAH-degrading Actinobacteria (Arthrobacter sp. and Rhodococcus ruber) were isolated from the original sediment sample and showed high activity in the degradation of phenanthrene and fluorene in liquid cultivation. This study reveals that indigenous bacteria had the ability to degrade high concentrations of mixed PAHs and provide clear evidence that Actinobacteria may be potential candidates to play a major role in PAH degradation in the river sediment.

During ovary maturation of crabs, vitellogenin (Vg), a precursor molecule of vitellin (Vn) needed for embryogenesis, can be produced in large quantities in the hepatopancreas and then transported to the ovary by the hemolymph. In the present study, effects of Cd on Vg accumulation in the hepatopancreas and Vg transportation of the freshwater crab Sinopotamon henanense were investigated. We also studied the impacts of Cd on the mRNA expression of genes involved in energy metabolism, protein metabolism, and metallothionein (MT) and glutathione (GSH) synthesis. After Cd treatment, the Vg concentration and the Vg mRNA expression in the hepatopancreas were downregulated. Pearson’s correlation coefficient showed that the Vg level in the hepatopancreas correlated positively with those of the ovary and hemolymph (correlation coefficients 0.844 and 0.749, respectively), suggesting that the Vg transport from the hepatopancreas to the ovary can be impaired by Cd. The levels of carbohydrate and protein in the hepatopancreas of Cd-exposed crabs were decreased, and an inhibited protein metabolism was also observed. Energy production related isocitrate dehydrogenase and cytochrome C oxidase mRNA expressions, and MT and GSH synthesis increased after 10 days of Cd treatment and decreased after 20 days. Cd also caused a time-dependent upregulation of malondialdehyde. Our findings showed that Cd decreased Vg accumulation in the hepatopancreas due to partially excessive energy consumption and an activated defense system in the hepatopancreas, suggesting a possible regulatory mechanism in S. henanense which is the competitive advantage of energy reserves in metabolic Cd stress responses over the high-energy flux during vitellogenesis to ensure a continuous supply of metabolic energy. Moreover, the damage of Vg accumulation in the hepatopancreas caused by Cd could lead to an insufficient accumulation of Vn in the ovary and cause a retardation of oocyte development.

Copper ions were first adsorbed by zeolite 4A synthesized from bauxite tailings, the desorption of Cu(II) using Na₂EDTA solutions was performed, and the recycling of zeolite 4A in adsorption and desorption was systematically investigated. It was observed that the Cu(II) removal efficiency was directly dependent on the initial pH value. The maximum removal efficiency of Cu(II) was 96.2% with zeolite 4A when the initial pH value was 5.0. Cu(II) was completely absorbed in the first 30 min. It was also observed that the desorption efficiency and zeolite recovery were highly dependent on the initial pH and concentration of Na₂EDTA in the solution. The desorption efficiency and percent of zeolite recovered were 73.6 and 85.9%, respectively, when the Na₂EDTA solution concentration was 0.05 mol L⁻¹ and the pH value was 8. The recovered zeolites were pure single phase and highly crystalline. After 3 cycles, the removal efficiency of Cu(II) was as high as 78.9%, and the zeolite recovery was 46.9%, indicating that the recovered zeolites have good adsorption capacity and can repeatedly absorb Cu(II).

Ferrihydrite (Fh) has been recently used in water treatment for removing dissolved organic matter (DOM), but its governing interactions with low-molecular weight DOM are largely unknown. This study aimed to elucidate the influence of chemical structure of DOM on the interactions between functional groups of DOM and Fh using various surrogates representing DOM in natural waters. We tested four surrogate compounds: L-glutamic acid, resorcinol, L-serine, and tannic acid, which represent the main chemical groups of carboxylic and hydroxyl groups; and the Suwannee River NOM (SRNOM) that represents the composition of DOM in natural aquatic systems. Batch adsorption experiments revealed that the DOM adsorption onto Fh was significantly influenced by the steric arrangements of –COOH and –OH functional groups. Both L-serine with α-carboxyl group and resorcinol with hydroxyl groups in meta-position were marginally removed by Fh, indicating that the adsorption of DOM on Fh was determined by their chemical structures and the relative positions of carboxylate and hydroxyl groups. The adsorption of L-glutamic acid was controlled by the pH-dependent ligand exchange of γ-carboxyl groups, which was similar to the SRNOM adsorption. In contrast, adsorption of tannic acid was not affected by pH, which can be explained by a two-step adsorption, namely, ligand exchange followed by multi-layer adsorption to the partitioning phase. The results of kinetic experiments demonstrated that adsorption of DOM by Fh was significant and rapid. The kinetic adsorption data can be expressed by the pseudo-second-order equation, indicating that the adsorption step might be the rate-limiting step.

Anaerobic sediments of urban watercourses are subjected to industrial pollution and frequently tend to accumulate heavy metals. The biocatalyzed oxidation and reduction of sulphur compounds that occur within the sediment are key reactions that determine mobility of metals such as that occurred in mine acidic drainage reactions. The aim of this work was to study the application of these processes using heap leaching technology for the remediation of anaerobic contaminated sediments from Reconquista River basin. The bioleaching potentiality for remediation was demonstrated through batch tests in shake flasks with different pulp densities of anaerobic sediment containing 338 mg kg⁻¹ of Zn and 117 mg kg⁻¹ of Cu. Subsequently, bioleaching heap systems were compiled into columns of 12-cm height and 6-cm diameter, fitted with perlite to improve drainage. In order to assess the effect of elementary sulphur over the mobility of metals from the bioheap to the aqueous solution, increasing concentrations of elementary sulphur (1, 2, 5 % w/w) were added. After 3 months of acidification generated by periodic watering, the extraction of 70 % of the initial Zn and 43 % of the initial Cu was achieved. Polluted sediments from waterways as Reconquista River should not be indiscriminately manipulated if acid drainage is possible. Remediation by a simple and economically viable strategy like heap leaching is feasible.
