2020
Fikriye Alev Akçay | Ayşe Avcı
In this study, effects of medium components and inoculum size on the protease production by Bacillus sp. EBTA6 that was isolated from a home-made Tarhana sample were investigated. The cell-free supernatant of bacterium cultured on a shaking incubator for 24 h was used to test protease activity as the response. With a total number of 11 factors, 12 different experiments were run and the highest experimental protease activity was measured as 2280.4 U/mL. Results were analyzed statistically by ANOVA and the most efficient factors were detected as yeast extract, dipotassium phosphate, casein, and peptone with a contribution of 93.78, 2.19, 1.96, 1.31%, respectively. For validation of the selected factors, a further experiment was performed by using of yeast extract (9.98 g/L), dipotassium phosphate (1.27 g/L), casein (8.69 g/L), and peptone (9.88 g/L) obtained from the design equation. The experimental response was found as 2411.4 U/mL which was only 5.5% higher than the predicted response showing that the model was applicable.

2019
Olga Büyükleblebici
Aksaray Malakli Shepherd Dog is a native Anatolian race which is raised in Aksaray and it is marked recently. This study is prepared to reveal significative biochemical properties of Aksaray Malakli Dog. Blood samples were collected from 20 Malakli dogs which are raised in various animal shelters in Aksaray region. Biochemical parameters; serum total protein, albumin, globulin, blood urea nitrogen, creatinine, glucose, total cholesterol, triglyceride, calcium, phosphor, iron, ferritin, and ALP, AST, ALT, creatine enzyme activities were measured calorimetrically. Obtained results were 6.25 g/dL, 3.62 g/dL, 2.63 g/dL, 16.53 mg/dL, 0.88 mg/dL, 91.10 g/dL, 186.85 mg/dL, 47.65 mg/dL, 9.53 mg/dL, 4.36 mg/dL, 112.45 µg/dL, 0.51 ng/dL respectively. and enzyme levels were 42.50 IU/L, 31.90 IU/L, 43.30 IU/L, 166.4 IU/L, respectively. Serum Paraoxonase 1 and hepcidine levels were measured with ELISA kits manually. Obtained results were 33.0 ng/ml and 23.55 ng/ml respectively. With this study, we determined some serum biochemical parameters of Malakli dog and we tried to find out the differences between the other Anadolu Çoban dogs. Obtained results may be useful for the next studies about morbidity-health status of these dogs.

The present study investigated the effects of the prebiotic inulin on the growth parameters, digestive enzyme activities and antioxidant status of rainbow trout (Oncorhynchus mykiss). After acclimation, fish (34±0.22 g; n = 135) were assigned into 9 tanks (15 fish per tank) and triplicate groups were fed a control diet (G-0) or diets containing 1% (G-1) or 2% (G-2) inulin. At the end of the trial (8 weeks), growth factors (final weight, weight gain, specific growth rate (SGR), food conversion ratio (FCR), protein efficiency ratio (PER), Economic conversion ratio (ECR), digestive enzyme activities (pepsin, trypsin, amylase and lipase) and antioxidant status superoxide dismutase (SOD), catalase (CAT) and malondialdehyde (MDA) level of liver tissue were assessed. At the end of the study highest survival rate was observed in the fish fed with 1% G-1 inulin group. Similarly, digestive enzyme activities were significantly higher dietary G-1 group. Subsequently, elevated growth performance (final weight, SGR and FCR) was observed in trout fed with 1% compared to the control group. The effect of the dietary inulin on digestion was partly observed by assaying the activity of pepsin, intestinal amylase, trypsin and lipase. Apart from pepsin activity, results indicated that dietary supplementation of inulin is beneficial and may also have differential effects upon digestive enzyme activities. Antioxidant enzyme activity of SOD and CAT in liver tissue was generally higher in the supplemented inulin diets than in the control and significantly higher in the 1% inulin group. A significant decrease in MDA level of tissue was observed in all inulin supplemented groups when compared to the control group. These results indicate that inulin be considered as a beneficial dietary supplement for growth performance, digestive enzyme activities and antioxidant status of rainbow trout.
