Hplc Analysis of Certain Phenolic Compounds and Carotenoid in Two Calendula Species

Calendula officinalis (pot marigold), a medicinal ornamental plant belonging to the Asteraceae family, has various medicinal activities such as anticancer, antimicrobial, antioxidant and antitumor. The retention time of quercetin (6.445 min), kaempferol (11.246 min), caffeic acid (2.333 min), beta carotene (9.614 min) standards, which were planned to be analyzed by HPLC, was determined. The HPLC conditions that are most suitable for these metabolites have been determined. The amounts of quercetin, kaempferol, caffeic acid and beta carotene in the seedling leaves of these two Calendula species on the 0., 15., 30., 45., 60. days were compared. Caffeic acid and beta carotene have been detected in the leaves. However, quercetin and kaempferol compounds were not detected. It was determined that the caffeic acid and beta carotene amounts of C. officinalis species were higher than that of C. arvensis. The highest caffeic acid in both C. officinalis and C. arvensis seedlings (0.4042±0.0123 µg/g dry weight for C. officinalis, 0.1918±0.0437 µg / g dry weight for C. arvensis) and beta carotene amounts (0.8520±0.0692 µg/g dry weight for C. officinalis, 0.6389±0.0189 µg/g dry weight for C. arvensis) were determined on 60. day. In addition, it was determined that the amount of these metabolites differed according to the seedling development period.

In the present study, we aimed to isolate and biochemically identify tyramine-producing lactic acid bacteria (LAB) from black and green fermented olive samples produced by traditional methods and obtained from different provinces of Turkey. A total of 36 LABs, including Enterococcus spp. (2 isolates), Lactobacillus spp. (16 isolates), and Lactococcus spp. (18 isolates) were isolated from the fermented olive samples. Among them, 27 isolates could decarboxylate tyrosine amino acid; however, the decarboxylase enzyme activity of the remaining 9 isolates was negative. The ability of LAB isolates with positive enzyme activity to produce tyramine was evaluated using high-performance liquid chromatography (HPLC), and 6 isolates were found to be significant tyramine producers in vitro, producing tyramine at concentrations ranging from 107.251 to 207.618 mg L–1.

Apples, rich in phenolic compounds, are widely grown in abundance and consumed as fruit which reduces the risk of many illnesses. In this study, physico-chemical properties old apple cultivars from east of Turkey were investigated. Identification and quantification of phenolic compounds of freeze dried fresh apple varieties (fruit, pulp and peel) were analysed by using the HPLC method. Total phenolic content and antioxidant capacity of samples was also found in 25.00-11.63 mg GAE/g DW and 49.86-86.12 µmol TE/g DW. The highest antioxidant activity was identified in the peel of the Kara Sakı apple whereas the lowest was identified in the pulp of Ak Sakı. Varieties of apple differ significantly in terms of ferulic acid and hidroxycinnamic acid. (-)-Epicatechin was found in the peel of Kara Sakı apple at the very most and p- coumaric acid was found in the pulp of Kara Sakı at the very least. Hidroxycinnamic acid was identified only in the peel. It is approved to these cultivars show unique long shelf life (unusual for apples) without any storage condition due to the significantly presence of ferulic acid.

Bu çalışmada, Türkiye’de başlıca zeytin yetiştiriciliği yapan Ege bölgesinin bazı illerinden (Muğla, Aydın, İzmir ve Manisa) 2 hasat dönemi (2007-2008 ve 2008-2009) süresince yerli zeytin çeşitlerinin (Gemlik, Memecik, Ayvalık, Uslu ve Domat) yağı incelenmiştir. Bu yağların toplam fenolik madde ve fenolik bileşimi belirlenmiştir. Yağların toplam fenolik madde içeriği, 2007-2008 hasat dönemi örneklerinde 23,69-153,64 mg kafeik asit/kg, 2008-2009 hasat dönemi örneklerinde 16,18-136,22 mg kafeik asit/kg aralığında belirlenmiştir. Tüm zeytinyağı örneklerinde tespit edilen fenolik maddeler; tirozol, oleuropein, 4-hidroksifenil asetik asit, luteolin, vanilik asit, hidroksitirozol, rutin, sinnamik asit, verbaskozit, hidroksi fenilkarboksilik asit, sirinjik asit, 3,4-dihidroksibenzoik asit, kafeik asit, ferulik asit, p-kumarik asit, taksifolin ve apigenindir. Tirozol ve oleuropeinin, 2007-2008 hasat döneminde 1,80-13,39 mg/kg, 1,26-19,50 mg/kg ve 2008-2009 hasat döneminde ise 1,76-11,66 mg/kg, 0,20-13,12 mg/kg aralığında en fazla miktarda değişen bileşenler olduğu saptanmıştır.

Dracocephalum moldavica L. has been used as both treatment of coronary heart disorders and food supplements. In the present study, determination of total phenolic and flavonoid content in methanol, ethanol and methanol/ethanol extracts of leaf was studied in Iranian Moldavian balm for the first time. Antioxidant activity of the studied plant was also measured. The methanol extract had the highest phenolic and flavonoid content, anthocyanin, DPPH and H2O2 radical scavenging activity. The ethanol extract showed the least amount of all. The methanol/ethanol extract showed the highest amount in two oxides including nitric and superoxide radical scavenging activities; it also showed the highest Ferric Reducing Ability Power. The obtained chromatograms of the plant using High Performance Liquid Chromatography showed that the highest and the lowest found phenolic compounds were caffeic acid and vanilic acid, respectively. The results show that this plant is a suitable natural antioxidant to reduce the oxidative stress in human being.

Gümüşhacıköy Basma is famous for its aromatic structure in oriental type tobacco. However, it is known that new types of oriental tobacco have become widespread by adding Taşova, Erbaa and Niksar lines to the production area of this type of tobacco, especially due to the decrease in production since the beginning of 2000s. In this study, yield and quality performances of some oriental tobacco parents and hybrids in Gümüşhacıköy ecological conditions and heterosis and heterobeltiosis values of these hybrids were emphasized. Hybrid lines were obtained in 2016 by half-diallel hybridization from Xanthi-2A, Erbaa, Nail, Katerini and Canik 10821 (Sıtmasuyu) genotypes. The study was carried out with 5 parents and 4 F1 populations with their hybrids. Yield and yield related characteristics (plant height, number of leaves, leaf width and height), as well as glucose, fructose, nicotine, chlorogenic acid and routine contents were examined. It was determined that genotypes showed significant differences in all traits except leaf length. Hybrids outperformed their parents in all parameters except plant height and number of leaves. This advantage was 8.42% in yield, 7.73% in quality, 8.64% in reducing sugar, 3.49% in nicotine and 15.20% in chlorogenic acid + routine. The effect of leaf width on yield and chlorogenic acid + routine content on quality were determined. As a result of the study, when heterosis and heterobeltiosis data were evaluated, it was found that Xanthi 2A x Katerini hybrid showed promising results.

More than ninety percent of the ruminant livestock in Nigeria lies in the hands of herders who keep them under extensive and semi-intensive management systems, whereby the animals rely only on natural pasture and crop residues for survival. In this work, the mycoflora and aflatoxin levels of ten farms were determined by sampling crop residues on farms grazed by cattle. Samples of the remains of farm harvest were surface-disinfected and cultured using standard microbiological techniques while aflatoxins in the left over harvest were determined using High Performance Liquid Chromatography (HPLC) with fluorescence detection. Fungal counts in leftover harvest ranged from 1.2 x 106 to 3.8 x106cfu/g. Aspergillus flavus, A. terreus, A.parasiticus, Rhizopus sp and a yeast, Candida sp were most prevalent on all the investigated crop residues. Aflatoxin B1 (AFB1) on the crop residues ranged between 3.0 and 13.30 μg/Kg, while the levels of AFG1 were between 2.30 and 4.50 μg/Kg. Results of the present study is indicative that the accumulation of these doses of AFB1 can lead to transfer of AFB1 into cattle and subsequently into milk. So there is an urgent need to control the feeding pattern of cattle in order to protect the health of the consuming public.

Mycotoxin occurrence in foods, especially in uncontrolled produced traditional foods causes serious health problems. In this study, traditional Küp cheese samples were collected from different part of Anatolian region in Turkey (Ankara, Nevşehir and Yozgat) and analyzed to determine Aflatoxin M1 (AFM1) level. AFM1 analysis was carried out by, immunoaffinity column (IAC) clean-up and high performance liquid chromatography (HPLC) attached with fluorescence detector (FL) The level of AFM1 in all samples was in the range of 16 and 136 ng/kg which is lower than the maximum tolerance limit of the Turkish Codex Regulations (250 ng/kg). The levels of contamination indicated that more detailed and continuous monitoring is required to increase the public health conscious and reduce consumers’ exposure to AFM1.

Yapılan bu çalışmada ısıl işleme alternatif termosonikasyon (TS) yöntemi ile taze elma suyu farklı genlik (%60, 80, 100), sıcaklık (40, 50, 60 ve 70°C) ve sürelerde (5, 10, 15, 20, 25 ve 30 dakika) pastörize edilmiştir. Bu yöntemin elma suyunda bulunan polifenoloksidaz (PPO) ile peroksidaz (POD) ile fenolik bileşiklere etkisi araştırılmıştır. Fenolik bileşikler HPLC ve FTIR spektroskopisi kullanılarak belirlenmiştir. İnaktivasyon sonuçlarına bakıldığında elma suyunda %100 genlikte, 70°C sıcaklıkta 15 dakika işlem sonunda PPO enziminin %99’u inaktif olurken, aynı koşullarda POD enziminin %94,5’i inaktif olmuştur. %80 genlik, 60°C sıcaklık ve 15 dakika toplam fenolik madde ve antioksidan aktivitenin en iyi korunduğu işlem parametreleri olarak belirlenmiştir. HPLC ile belirlenen fenolik bileşikler; kateşin, epikateşin, klorojenik asit, kafeik asit ve kamferoldur. FTIR spektrumlarına bakıldığında fenolik bileşiklerde önemli bir değişiklik olmadığı belirlenmiştir. Aynı zamanda HPLC ve FTIR ile belirlenen fenolik bileşiklerin benzer olduğu tespit edilmiştir. Termosonikasyon elma sularının işlenmesinde enzimlerin inaktivasyonu ve biyoaktif bileşiklerin korunmasında umut verici bir yöntem olarak önerilebilmektedir.

The aim of the study is to evaluate the ethanol extract of Rheum ribes root (RTE), as well as the root and young shoots (RYSE), for its chemical composition, antioxidant properties and cytotoxic effects. Total polyphenols (TPC), total flavonoid (TFC), radical scavenging activity (DPPH), the ferric reducing antioxidant power (FRAP) of Rheum ribes extracts were determined using colorimetric methods. Mineral contents and vitamin values of samples were determined by ICP-MS and HPLC, respectively. The cytotoxic effect of Rheum ribes extracts was determined on different cells using XTT assay. The cytotoxic effects of RTE and RYSE on cancer cells were evaluated with regard to apoptosis. According to results, mineral contents, vitamine A and C values were very high in both samples. Ethanolic extracts of Rheum ribes young shoot exhibited a selective cytotoxic effect on all cancer cells compared to WI-38 cells, and the IC50 values of the extract in the cancer cells between 26.10 to 54.81 μg/mL. Ethanolic extracts of RTE and RYSE induced apoptosis on MCF-7 cells. The ethanolic extracts of Rheum ribes has effective cytotoxic and antioxidant activity. More research is needed to determine the cytotoxic effect mechanisms on cancer cells.
