Kırıkkale Delice İlçesi Yerel Üzüm Popülasyonun Bazı Morfolojik Özellikleri

Bu çalışma, Kırıkkale’nin Delice ilçesine özgü yerel bir üzüm popülasyon olan “Delice Üzümü”nün morfolojik ve kimyasal özelliklerini detaylı bir şekilde incelemeyi amaçlamaktadır. Araştırma kapsamında, Delice Üzümü’nün salkım büyüklüğü, tane şekli, kabuk rengi ve kalınlığı gibi morfolojik özellikleri laboratuvar ve saha çalışmaları ile belirlenmiştir. Ayrıca, üzümün kimyasal bileşimi, suda çözünür kuru madde (SÇKM) oranı, pH değeri, tartarik asit miktarı ve şıra randımanı gibi parametreler üzerinden değerlendirilmiştir. Elde edilen sonuçlar, Delice Üzümü’nün yüksek kaliteli bir üzüm popülasyonu olduğunu göstermektedir. Morfolojik incelemelerde, salkımların orta büyüklükte ve sıklıkta, tanelerin ise yumurta şeklinde ve tatlı olduğu belirlenmiştir. Kimyasal analiz sonuçları, SÇKM oranının %24-25, pH değerinin 3.7, tartarik asit miktarının 0.416 g/L ve şıra randımanının 500-550 ml olduğunu ortaya koymuştur. Bu bulgular, Delice Üzümü’nün hem sofralık tüketim hem de pekmez ve pestil gibi yöresel ürünlerin yapımında kullanılabilirliğini desteklemektedir. Çalışma, aynı zamanda Delice ilçesinin ekolojik ve toprak özelliklerinin üzüm yetiştiriciliği için uygun olduğunu, bu nedenle yerel çeşitlerin korunması ve geliştirilmesinin genetik çeşitlilik ve biyolojik çeşitliliğin sürdürülmesi açısından kritik önem taşıdığını vurgulamaktadır. Bu çalışma, yerel üzüm çeşitlerinin tarımsal ve ekonomik potansiyelini ortaya koyarak, gelecekteki araştırmalara ve sürdürülebilir tarım uygulamalarına katkıda bulunmayı hedeflemektedir.
Show more [+] Less [-]Molecular Detection and Characterization of New Emerging Viruses by PCR Analysis in Hatay and Tekirdag Vineyards

The improvements on the next generation sequencing or high-trough output technologies allowed the discovery of several unknown viruses in grapevines and also in other plants . The most important new emerging grapevine viruses were identified as Grapevine pinot gris virus (GPGV), Grapevine syrah virus 1 (GSyV-1), Grapevine red blotch-associated virus (GRBaV) and Grapevine roditis leaf discoloration virus (GRLDaV). The aim of the present study was to investigate the occurrence and characterization of these viruses in Tekirdag and Hatay viticulture production areas by PCR and DNA sequencing analyses. Totally 191 and 111 grapevine samples showing both virus-like symptoms and symptomless were collected from Tekirdağ and Hatay provinces, respectively. Among the tested samples GPGV and GSyV-1 were detected in both local and imported cultivars by the infection rate of 43.62 % and 1.04% in Tekirdağ , respectively. In Hatay provice, only GSyV-1 was detected by the infection rate of 0.9 % and all tested samples were negative for GPGV, GRBaV, GRLDaV. RT-PCR results showed that DNA fragments of 411 bp, 302 bp and 618 bp corresponding to the part of the coat protein (CP), movement protein (MP) and the replicase genes of GPGV were successfully amplified in Tekirdağ samples. All PCR products of GPGV were directly sequenced on both strands. All the nucleotide sequences of CP, MP and 5’ UTR and N-terminus of replicase genes shared the highest sequence identity with different GPGV isolates deposited in Genbank
Show more [+] Less [-]Histopathological Research of the Therapeutic Effects of Grape (Vitis vinifera L.) Seeds Extracts on Cadmium-Exposed Carp (Cyprinus carpio L. 1758)

This study was designed to investigate the therapeutic effects of Vitis vinifera L. seed extract (5 ppm and 10 ppm) on gill and liver tissues histopathology of carp (Cyprinus carpio L. 1758) exposed to cadmium chloride (CdCl2=Cd) (20 ppb) acute and subchronic period. A total of 140 fish in 14 groups were used in the experiment. The experiment was designed as in two different periods (acute and chronic) and 7 different dose groups (control, vehicle, V. vinifera seed extract 5 ppm, V. vinifera seed extract 10 ppm, Cd 20 ppb, V. vinifera seed extract 5ppm + Cd 20 ppb and V. vinifera seed extract 10 ppm + Cd 20 ppb). At the end of the experiment, the gill and liver tissues dissected from the fish were fixed, dehydrated, cleared, and paraffin impregnated. Appropriately sized blocks were prepared from the tissues, sections of 5-7 µm thickness were taken and covered by staining with hematoxylin-eosin. Stained preparations were examined under a light microscope and photographed. It was determined that all dose groups in the acute period and control groups were not different histopathological. It was determined that the histopathological damage caused by the cadmium group in the subchronic period and tissue damage were reduced in the dose groups with cadmium added with V. vinifera seed extract. As a result of the study, it was concluded that V. vinifera seed extract could have a therapeutic effect on the gill and liver tissues of carp exposed to cadmium.

In this study, total phenolic compounds which possess health-promoting effects and provide basic nutrition were aimed to determine the alterations in different grapevine leaves. Total phenolic content was determined using Folin-Ciocalteu colorimetric method by using a spectrophotometer. At the result of the study, it was determined that contents of total phenolic in leaves collected from different grape cultivars were varied to cultivars and grape leaves had high phenolic compounds as berries. Therefore the grape leaves especially wine grapes such as Narince, Boğazkere ve Kalecik Karası may be used as an easily accessible source of natural antioxidants and food supplement at pharmaceutical and food industry.
