Hindbrain Double-Negative Feedback Mediates Palatability-Guided Food and Water Consumption Full text

Hunger and thirst have distinct goals but control similar ingestive behaviors, and little is known about neural processes that are shared between these behavioral states. We identify glutamatergic neurons in the peri-locus coeruleus (periLCⱽᴳᴸᵁᵀ² neurons) as a polysynaptic convergence node from separate energy-sensitive and hydration-sensitive cell populations. We develop methods for stable hindbrain calcium imaging in free-moving mice, which show that periLCⱽᴳᴸᵁᵀ² neurons are tuned to ingestive behaviors and respond similarly to food or water consumption. PeriLCⱽᴳᴸᵁᵀ² neurons are scalably inhibited by palatability and homeostatic need during consumption. Inhibition of periLCⱽᴳᴸᵁᵀ² neurons is rewarding and increases consumption by enhancing palatability and prolonging ingestion duration. These properties comprise a double-negative feedback relationship that sustains food or water consumption without affecting food- or water-seeking. PeriLCⱽᴳᴸᵁᵀ² neurons are a hub between hunger and thirst that specifically controls motivation for food and water ingestion, which is a factor that contributes to hedonic overeating and obesity.

Yamato Tachibana (Tachibana; Citrus tachibana) is an endemic fruit and represents one of the oldest citrus species in Japan; it is grown in the Mie Prefecture. It has been attracting attention for its cultural heritage and unique scent. To evaluate biological activities of Tachibana, we fed several parts of the Tachibana fruit (whole fruit, pulp [albedo and segment wall], and flavedo) to adult zebrafish and found that Tachibana increased body weight and plasma triglycerides besides increasing overall food intake. We then created a simple fluorescence-based feeding assay using dried rotifer sheets and larval zebrafish (6 days postfertilization) to screen the various extracts of Tachibana parts. We found that water extracts of Tachibana pulp increased feeding volume in zebrafish. Although citrus species are believed to prevent obesity and obesity-associated diseases in general, our findings showed that water extracts of Tachibana increase food intake in zebrafish and lead to an increase in body weight. We suggest that Tachibana might reverse appetite loss in lean populations and may prove beneficial in aiding fish cultivation.

The 2012 GHI report focuses particularly on the issue of how to ensure sustainable food security under conditions of water, land, and energy stress. Demographic changes, rising incomes and associated consumption patterns, and climate change, alongside persistent poverty and inadequate policies and institutions, are all placing serious pressure on natural resources.In this report, IFPRI describes the evidence on land, water, and energy scarcity in developing countries and offers two visions of a future global food system - an unsustainable scenario in which current trends in resource use continue, and a sustainable scenario in which access to food, modern energy, and clean water improves significantly and ecosystem degradation is halted or reversed. Concern Worldwide and Welthungerhilfe provide on-the-ground perspectives on the issues of land tenure and title as well as the impacts of scarce land, water, and energy on poor people in Sierra Leone and Tanzania.

The usefulness of replacement of caloric sugars by low-calorie sweeteners (LCS) for weight management has been questioned on the grounds that the uncoupling of LCS sweet taste and dietary energy may confuse physiological mechanisms, leading potentially to higher energy and sugar intake. The aim of the present study was to determine whether LCS beverages compared to water, when consumed with meals, differ in their effects on energy and food intake in acute trials and after long-term habituation. Ad libitum food intake of 166 (80 women; 86 men) healthy non-obese adults (BMI between 19 and 28 kg/m2), infrequent consumers of LCS was measured in four 2-consecutive-day testing sessions (Day 1 in the laboratory, Day 2 free-living). During the first 3 sessions, held one-week apart, participants were required to drink either water or commercial non-carbonated LCS lemonade (330 ml) with their main meals (randomised cross-over design). On Day 1, motivational ratings were obtained using visual analogue scales and ad libitum food intakes (amounts and types of foods selected) were measured using the plate waste method. On Day 2, participants reported their ad libitum intakes using a food diary. After Session 3, participants were randomly assigned to the LCS habituation group or to the water control group. The habituation (660 ml LCS lemonade daily vs 660 ml water) lasted 5 weeks. The fourth and final test session measured food intakes and motivational ratings after habituation. Water and LCS beverage did not differ in their effects on total energy intake, macronutrient intakes or the selection of sweet foods and on motivational ratings. Similar results were obtained in both LCS-naïve and LCS-habituated individuals.
