Serological diagnosis and risk factors for Coxiella burnetii in goats and sheep in a semi-arid region of Northeastern Brazil Full text

Abstract Coxiella burnetii is a zoonotic agent transmitted mainly by small ruminants. In Brazil the disease has been classified as a notifiable disease since 2013, when human cases were reported. This study aimed to identify risk factors associated with the presence of anti- Coxiella burnetii antibodies in goats and sheep in a semiarid region of Northeastern Brazil. Sera of 412 goats and 403 sheep from municipality of Petrolina, Pernambuco, were examined by the Indirect Fluorescent Antibody Test (IFAT) against antigens of C. burnetii. Information about management variables (independent variables) that could be associated with the presence of the microorganism (dependent variables) were obtained from the supervisor of each farm. It was determined that 2.2% (9/412) of the goats and 2.1% (9/403) of the sheep had antibodies reactive to C. burnetii. The presence of anti-C. burnetii antibodies was associated with the dry area of the Sequeiro (a region in the northern part of the municipality of Petrolina) (P = 0.025), male sheep (P = 0.020), and intensive goat breeding (P = 0.005). This study therefore showed the presence of anti-C. burnetii antibodies in goat and sheep, confirming for the first time that this agent is likely circulating among goat herds in the Caatinga Biome, semi-arid of Brazil.

Abstract Recently, the importance of wild-living rodents for maintenance of pathogens of the family Anaplasmataceae in the environment was investigated. These mammals play a role as reservoirs for these pathogens and act as hosts for the immature stages of tick vectors. The aim of the present study was to investigate the prevalence of Ehrlichia sp. and Anaplasma sp. in 24 specimens of Azara’s agouti (Dasyprocta azarae) that had been trapped in the Itapiracó Environmental Reserve, in São Luís, Maranhão, northeastern Brazil, using molecular methods. Four animals (16.7%) were positive for Ehrlichia spp. in nested PCR assays based on the 16S rRNA gene. In a phylogenetic analysis based on the 16S rRNA gene, using the maximum likelihood method and the GTRGAMMA+I evolutionary model, Ehrlichia sp. genotypes detected in Azara’s agoutis were found to be closely related to E. canis and to genotypes relating to E. canis that had previously been detected in free-living animals in Brazil. The present work showed the first molecular detection of Ehrlichia sp. in Azara’s agoutis in Brazil.

Abstract This study involved two field trials with the aim of evaluating the efficacy of diflubenzuron, via mineral supplementation, against Haematobia irritans parasitizing cattle. Concomitantly with the main trial, a stall test was conducted to ascertain the effects of a different formulation with the same active ingredient against Rhipicephalus microplus, along with the action of diflubenzuron on the reproductive parameters of R. microplusfemales that had naturally detached from cattle. Against H. irritans, it was observed that the efficacy indexes fordiflubenzuron were low (≤ 31.3% or 44.6%) or null (0.0%) throughout the study. The anti- R. microplus efficacy of diflubenzuron, at weekly intervals, ranged from 0.0 to 13.7% over the entire experimental period. Null efficacy (0.0%) was registered for diflubenzuron in relation to the reproductive parameters of R. microplusfemales that had naturally detached from cattle. The different diflubenzuron formulations, administered via mineral salt supplementation, did not show satisfactory efficacy indexes against H. irritans and R. microplus parasitizing cattle, within the experimental design of the present study. In addition, this agent did not present any deleterious effects on the reproductive parameters of R. microplus females.

Abstract Visceral leishmaniasis (VL) is a disease caused by the protozoa Leishmania infantum and can cause an inflammatory reaction in the gastrointestinal tract, however the role of granulocytic cells (neutrophils, eosinophils, and mast cells) in the intestine of dogs infected is not fully understood. We performed a quantitative analysis these cells in the intestinal wall of dogs with canine visceral leishmaniasis (CVL). Twenty dogs were assigned to one of three groups: group 1 (G1, n=8), dogs with CVL and L. infantum amastigotes in the intestine; group 2 (G2, n=9), dogs with CVL but without intestinal amastigotes; and group 3 (G3, n=3), uninfected dogs (control group). Granulocytic cells were counted in the crypt-villus unit (mucosa), submucosa, and muscle layer of the intestinal mucosa. Cell counts were higher in the intestinal wall of dogs from G2 followed by G1 and G3 (p≤0.05). In G1, there was a low inverse correlation between parasite burden of the small intestine and granulocyte counts (r= -0.1, p≤0.01). However, in G2 dogs, mast cell and eosinophil numbers showed positive correlation (r=0.85, p≤0.01). The granulocytic cell hyperplasia observed in the intestine of L. infantum-infected dogs suggests that these cells may be involved in the cell-mediated immune response for parasite elimination.

Abstract This study aimed to perform a morphological, molecular and phylogenetic characterization of Borrelia theileri obtained from infected Rhipicephalus microplus in Brazil. Fifty engorged R. microplus females from cattle in the municipality of Seropédica, Rio de Janeiro, were analyzed for spirochetes by hemolymph smear. Macerated eggs and positive ticks, as well as blood from the bovine infested by these ticks, were analyzed the glpQ, flaB and hpt genes by PCR. The PCR products were purified and sequenced for analysis and construction of a phylogenetic tree. Only 2% (1/50) of the ticks generated a positive result by both smear and PCR. The spiral forms (n = 50) had (media ± SD) a mean length of 19.17 ± 4.12 µm, diameter of 0.2935 ± 0.0469 and number of turns 8.44 ± 2.59. Sequence alignments of the three evaluated genes exhibited 98% similarity to B. theileri isolates, occurring in a clade highly related to B. theileri strain KAT. Egg maceration samples were positive for the three evaluated genes, whereas bovine blood was negative by PCR. This is the most detailed characterization of B. theileri in the Americas to-date, presenting morphological, molecular and phylogenetic data, including the transovarial transmission of the spirochete in the host tick.

Abstract The occurrence of Eimeria Schneider, 1875 in mammals of the order Didelphimorphia indicates the infection-predisposition of these animals, which in turn is mainly determined for their eating habits. The objective of this work was to evaluate the parasitism of Eimeria spp. in marsupials of the Atlantic Forest of the southern region of Bahia. Fecal samples were collected from marsupials captured in the regions of Ilhéus, Una, Belmonte and Mascote, with traps of the Sherman model (23 × 8 × 9 cm), Tomahawk (50 × 17 × 17 cm) and pitfall and analyzed by Sheather's modified centrifugal-flotation method. Oocysts were identified by microscopical evaluation of their morphology and morphometry. Didelphis aurita Wied-Neuwied, 1826, Gracilinanus agilis Burmeister, 1854, Monodelphis americana Müller, 1776, Marmosa demerarae O. Thomas, 1905 and Marmosa murina Linnaeus, 1758 were parasitized by Eimeria philanderi Lainson & Shaw, 1989 and Eimeria gambai Carini, 1938. Mixed parasitism for these two coccidia was observed in two of the 56 marsupials sampled. In conclusion, this work registers new hosts for E. philanderi and E. gambai, as well as the state of Bahia as a new distribution site for these coccidia.

Abstract Bovine coccidiosis is a disease of major importance in cattle herds across the world. The disorder mainly affects young calves, and E. bovis and E. zuernii are considered the most pathogenic species of the genus, however, E. alabamensis have been described in grazing calves. In this study, the prevalence of Eimeria spp. was evaluated in calves on dairy farms in the northern region of the state of Paraná, Brazil. Four hundred calves on 44 dairy farms were tested for the presence of coccidian oocysts. The positives were re-examined and the oocysts were morphometrically analyzed for species identification. All the farms were contaminated and 205 animals (51.25%) presented Eimeria spp. oocysts. Among these, 146 animals (71.22%) were co-infected by two or more species of coccidia. Ten species of Eimeria were identified: E. bovis (in 30.25% of the positive samples), E. alabamensis (26.75%), E. zuernii (22.00%), E. ellipsoidalis (18.50%), E. auburnensis (13.75%), E. canadensis (8.00%), E. cylindrica (7.25%), E. subspherica (5.00%), E. bukidnonensis (3.00%) and E. brasiliensis (0.75%). This study demonstrates the high prevalence of Eimeria spp. in the northern region of Paraná, Brazil, and detection for the first time in our region the pathogenic species E. alabamensis.

Abstract This study investigated the parasites fauna of four freshwater ornamental fish species in aquarium shops of Macapá, Amapá State, in addition to survey the commercialized fish species and sanitary conditions of aquarium shops. Different native and non-native ornamental fish species were found in aquarium shops, mainly Poecilidae. We examined 30 specimens of Xiphophorus maculatus, 30 Danio rerio, 30 Paracheirodon axelrodi, and 30 Corydoras ephippifer for parasites. Of the 120 fish examined, 22.5% were parasitized by one or more species and a total of 438 parasites were collected and identified. Parasites such as: Ichthyophthirius multifiliis, Monogenea, undermined Digenea metacercariae, Acanthostomum sp. metacercariae, Camallanus spp., Bothriocephalus acheilognathi and Echinorhynchus sp. infected the hosts examined. Endoparasites in the larval stage showed the greatest diversity and Camallanus spp. was found in all hosts species examined. Paracheirodon axelrodi (43.3%) was the most parasitized host, while C. ephippifer (6.7%) was the least parasitized. Despite the low ectoparasites level, six species of endoparasites was observed, demonstrating that prophylactic and quarantine procedures were not fully adequate. Therefore, failures in prophylactic procedures on any link in the production industry of ornamental fish may cause parasite transmission to ornamental fish captured in different environments and localities.

Abstract To describe the parasitic community of the Magellanic Horned Owl, Bubo magellanicus (Aves, Strigiformes), 19 carcasses from central Chile were analyzed. Ectoparasites were collected through plumage inspection, while endoparasites were collected through traditional techniques of parasitological necropsy. Sixteen owls were infected with at least one species of ectoparasite (84.21%) or endoparasite (31.58%). Eleven of 19 birds (57.89%) harbored feather mites of the three species Pandalura cirrata (42.11%), Glaucalges attenuatus (47.37%), and Kramerella sp. (10.53%), whereas 16 individuals (84.21%) harbored the chewing louse Strigiphilus chilensis. Only six birds (31.58%) were infected with helminths; the nematodes Capillaria tenuissima (26.32%) and Dispharynx nasuta (5.26%); the acanthocephalan Centrorhynchus spinosus (5.26%); and the trematode Neodiplostomum sp. (5.26%). Apart from S . chilensis, all parasites comprised new records for B . magellanicus.

Abstract The aim of this study was to evaluate the presence of anti-Toxocara antibodies in naturally infected broiler chickens (n = 189) from the state of Paraná, southern Brazil. The chickens were reared in a semi-intensive system by small family farmers (n = 7). An enzyme-linked immunosorbent assay (ELISA) was performed to detect the presence of anti- Toxocara spp. IgY after serum adsorption with Ascaridia galli antigens. An overall seroprevalence of 67.7% (128/189; 95% CI = 61.1-74.4) was observed. The frequency of positive animals by farm ranged from 29.6% to 100%. The optical density and reactivity index values observed in ELISA test indicated the possible chronicity of infection of the evaluated chickens. Associations between the presence of antibodies and the area where the chickens were reared (p = 0.382) or the population density of dogs on the farm (p = 0.785) were not observed. This study shows a high prevalence of Toxocara spp. antibodies in broiler chickens reared in semi-intensive systems and provides evidence that chickens are a good indicator of environmental contamination by larva migrans agents. Further studies are necessary to assess the risk factors associated with poultry infection and the likelihood of toxocariasis transmission to humans via the ingestion of free-range chicken meat.
