Isolation and culture of lipoxygenase lacking cell lines through cell culture of soybean

The aim of this experiment is to select lipoxygenase lacking mutant cell line by in vitro mutagenesis in soybean. MS medium supplemented with 0.01mg/l picloram plus 0.2mg/l BA was effective for callus induction and cell suspension culture using cotyledon tissue. Under different EMS concentrations applied for mutation induction, 0.05% EMS resulted in decreasing cell growth rate down to 30%. Using a large number of colonies originated from the mutagenized cells, screening for lipoxygenase mutant cell line was carried out based on the carotene bleaching tests and lipoxygenase-3 deficient mutant line was selected. In the analysis of fatty acid composition isolated mutant cell line showed the low content of linolenic acid
