Studies on the anther culture of Oryza sativa L. 2. Variation of S pollen grains by cultural environments in anther culture

The effects of seasonal changes and low temperature treatment at the meiotic stage on S pollen ratio and callus formation from cultured anther was investigated in 10 rice cultivars. The number of S pollen grains was significantly increased in winter, and was higher in japonica type cultivars, late ripening habit, with high sporophytic potentiality. The effect of low temperature at the meiotic stage on the increase of S pollen grains was more noticeable at 22deg C than 17deg C because of the increase of degenerated pollen grains. The low temperature treatment at the tetrad stage (auricle distance; -1 - 0 cm) was better than at the first meiotic stage (auricle distance; -4 - -5 cm). Callus formation was positively related to the rate of S pollen grains and it was more increased in winter than in summer. The rate of S pollen grains and callus formation was increased in order of japonica type later-ripening cultivars (Daecheongbyeo, Nagdongbyeo and Chucheongbyeo), japonica type early-ripening cultivars (Sobaegbyeo and odaebyeo) and tongil type cultivars (Chilseongbyeo, Gayabyeo, Milyang 23, Samgangbyeo and Pungsanbyeo)
