Embryogenesis from the protoplast culture of Glycine soja

Protoplasts were isolated from the cotyledons of Glycine soja enzymatically and cultured. Freshly isolated protoplasts were initially plated in K8P liquid medium and then diluted with K8 medium to reduce the osmoticum level. G. soja produced somatic embryos from the suspension cultures of protoplast derived calluses. They were initially cultured in Uchimiya and Murashige medium [1] and then inoculated in the embryoid induction media. Embryoid induction was achieved when cultured in M1 medium containing Murashige and Skoog basic salts [2] with reduced nitrogen content and 2 mg/l 2,4-D and then subcultured sequentially in the M2 medium containing 2 mg/l 2.4-D and 0.1uM ABA and Mo medium containing only MS basic salts with reduced nitrogen content.
