[Detection of potato viruses S and X in potato tubers in Mashhad]

In this study enzyme-linked immunosorbent assay (ELISA) was used to test for potato virus S(PVS) and potato virus X(PVX) in potato tubers. Tubers were collected from different fields in Mashhad region in randomized manner, they were placed in a darkened room at 4C until dormancy was broken, and then allowed to sprout at room temperature (approximately 22 C) for 3-4 weeks. Sprouts were tested for viruses by ELISA. Survey showed that PVS and PVX were found in all crops tested at an overall average of 58% and 46%, respectively. It is concluded that PVS is nearly more important than PVX in Mashhad. Microelisa plates were coated with Gammaglobulin at 2 microg/mg and enzyme-conjugated globulin was used at 1:400. The tissues wre triturated in PBS-Tween 20, containing 2% PVP and 0.2% ovalbumin. The Elisa results with PVS and PVX indicated that there was no significant difference between taking tuber sprouts and taking leaves from greenhouse grown plants
