Control of aflatoxin in groundnut products with emphasis on sampling, analysis, and detoxification

The use of selected mathematical models to describe the distribution of aflatoxin in groundnut kernels, roasted peanuts, peanut butter, and groundnut cake is investigated to facilitate the design of statistically sound sampling plans for these commodities. A subsampling mill was developed, to enable representative, comminuted subsamples to be rapidly produced from large samples of groundnut kernels. Methods for the accurate analysis of the aflatoxin content of groundnut products utilizing bonded-phase cleanup procedures in combination with high performance liquid chromatography (HPLC) and high performance thin-layer chromatography (HPTLC) are described. The application of enzyme-linked immunosorbent assay (ELISA) methods to the analysis of peanut butter is studied. A procedure for the detoxification of groundnut cake using ammonia gas at high temperatures and moderate pressures was developed, and preliminary toxicity trials were completed using ammoniated material generated by a 50-kg capacity reaction vessel
