Inducing, isolation and characterization of mutant strains with enhanced productivity of beta galactosidase

Aspergillus oryzae H 26 - producer of extracellular beta galactosidase was subjected to a three-stage mutagenic impact with N-methyl-N'-nitro-N-nitrosoguanidin, ultraviolet rays, and Ro rays. Screening of the mutants was carried out against a selective background of suitable nutrient media. The clones with possibly enhanced productivity of beta galactosidase were cultivated in submerged conditions which resulted in isolating a mutant whose enzyme activity was 5 times higher. The selected active mutants were also characterized with regard to the rate of hydrolysis of lactose contained in deproteinised rennet whey.
