Embryogenesis and organogenesis induction in oil palm callus

Primary callus induced from young leaves of D x P oil palm seedling had the highest relative growth rate when cultured in MS medium component with reduced 2,4-D concentration from 2.5 mg/l to 1 mg/l and 2,4-D at the same concentration with BA at 0.10 mg/l after 5 months of culture. After 5 months of subculture, there was a 100 percent embryogenic callus formation in 1 mg/l 2,4-D containing medium. Somatic embryos were formed at a higher rate in the medium containing 1 mg/l 2,4-D and 0.1 mg/l BA. Somatic embryos germinated well, expressed as green spot formation in normal MS and 1/2 MS rather than in phytohormone added medium, within 5 months of observation.
