Improvement of a plate culture method for Thiobacillus thiooxidans and isolation of a new strain

The improvement of Thiobacillus thiooxidans by conventional genetic manipulation has not been successful, mainly due to the difficulties in cultivating the bacterium on a solid medium. In this paper we descibe the development of an improved solid medium on which an isolate obtained from the culture of T. thiooxidans ON 106 can produce significantly large colonies. The improved medium is 9K medium described by Silverman and Lundgren supplemented with 0.3 % K2S4O6 instead of FeSO4 as an energy source and solidified with a 0.6 % gellan gum. The growth of isolate on the medium containing 0.1 % glucose, glucuronic acid or rhamnose (these are components of gellan gum) instead of K2S4O6 could not be observed even after 7 days. On the basis of these results and chemotaxonomic characteristics; the major quinone is ubiquinone 8, it has 3-OH C14: 0, 0, and its G + C content of DNA is 52%, the isolate is identified as belonging to T. thiooxidans
