Micropropagation of Dianthus baratus and D. chinensis through cotyledonary node culture [India]

Cotyledonary node explants (2-3 mm) of Dianthus barbatus and D. chinensis were cultured on B5, LS, MS and MSB basal media supplemented with BAP and NAA. The best response in terms of multiple shoot formation was observed on LS mediun with 2.0 mg/1 BAP and 0.50 mg/1 NAA. D. barbatus showed a higher frequency of shoot formation as compared to D. chinensis. In six weeks over 100 shoots were observed in D. barbatus and 60-65 in D. chinensis. Every two weeks inoculi of 6-8 shoots were transferred onto fresh medium for further shoot differentiation. Shoots at every subculture were rooted on hormone-free medium. Rooted plants were transferred to soil where they flowered normally and resembled seed-grown plants.
