The suspension culture of rice single cell

Single cell was isolated by use of callus drawn from the maturation embryo culture. For use of the callus of rice embryo as a material for isolating of cell, it was effective to add 2,4-D 2 ppm to Ms medium. The callus of third succession with the age of 10 days was better and 15 days of the isolation period was suitable. For the isolation of single cell, it was reasonable to add 2,4-D 2 ppm and sugar 2-3%. Using urea maselojium R-10 0.5%, the cell 4x10** (5)/ml could be isolated in 7-10 days. In the single cell culture, it is very effective when the original cell density was 3x10** (5)/ml, and the callus formation was faster when NAA 1 ppm was added
