Callus induction and plantlet/organ regeneration from immature embryos of five inbred lines of corn (Zea mays L.)

Five inbred lines (P10, PS2, Tzi 9, CBR 5031S6, and I12) of corn (Zea mays L.) were cultured in vitro. These inbred lines are currently used by the Cereals Division of the Institute in its breeding program and have the following characteristics: 1) P10 and PS2 have high combining abilities; 2) Tzi9 and CBR5031S6 are resistant to corn borer at the leaf-feeding stage; and 3) PS2 and I12 are susceptible to corn borer at the leaf-feeding stage. Embryogenic, compact and friable calli were initiated from immature embryos of these lines using N6 medium containing 0.5-2.0 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) and 3 and 12 percent sucrose. The calli retained their embryogenic or organogenic state for one month. Regeneration into plantlets, shoots or roots occurred thereafter. These embryogenic/organogenic calli and regenerated plantlets, shoots or roots will be used as starting materials for establishing the in vitro culture systems needed in the subsequent Bacillus thuringiensis endotoxin gene transfer work
