In vitro reaction of three sunflower inbred lines to different concentrations of oxalic acid - the toxin of Sclerotinia sclerotiorum

Three inbred lines, coded RF 7849, RF 7906 and RF 7898, were chosen for their distinct reactions to Sclerotinia sclerotiorum. The first two were known to be tolerant and middle-tolerant, and the last one susceptible. Immature embryos were excised 5 days post-anthesis and cultured on modified Murashige-Skoog's inorganic salts, supplemented with 62,5 mg/l and 250,0 mg/l oxalic acid and the fungal filtrate. The introduction of the oxalic acid in the basal MS culture medium, determined a decrease of the induced callus percentage and of the regenerants number, proportionally with the increase of the toxin concentration. When the MS medium was supplemented with the fungal filtrate, the decrease was higher. The preliminary results demonstrate that screening in vitro for disease resistance is a reliable method, which shows a good correlation with the reaction of the three inbred lines in field environments.
