Electrophoretic analysis of buckwheat (Fagopyrum esculentum Moench) seed proteins during germination

Qualitative changes in total seed proteins in germinated buckwheat (Fagopyrum esculentum Moench) seeds in a range of 30 to 82 kDa were studied by SDS-PAGE. During germination, seed proteins were sequentially degraded by endogenous proteolytic enzymes. On the third day of germination, primary protein bands disappeared and new bands emerged as a result of the cleavage of limited number of peptide bonds. The electrophoretic pattern of degradation products was almost the same for all protein phenotypes, and consequently for all buckwheat cultivars. After about six days of germination the SDS-PAGE method indicated only some minor protein components below 30 kDa. The degradation of proteins was also studied separately in endosperm and cotyledons.
