Haploid plants production and screening for potato leafroll resistant in potato

Flower buds of size 4-6 mm., with the anther length ca. 3-4 mm. are collected and kept at 6-10 deg. celsius for 48 h. After thorough surface sterilization, the anthers were excised and cultured in MS-PE, NN, LS and modified NN media. Almost every tested media support the callus formation, though some anthers turn brown after a short period of culturing. No direct embryogenesis occurred, however the adventitious shoot could be obtained from the root derived from the anther culture in the combination of MS and NN medium. Ploidy level of the anther derived plants can be determined by counting the number of chloroplasts in the guard cell. Tetraploidy has 15-21 and diploidy has 10-12 chloroplasts per cell. Testing for PLRV resistant in the diploidy, S. acaule and tetraploidy clones are done by ELISA technique. All the tested S. acaule are highly resistant to PLRV and could be further used for the PLRV resistant genes pooling in the fusion experiment.
