Changes of endogenous phytohormones of apple rootstocks during vitrification in vitro culture

Change of endogenous phytohormones in plantles from apple rootstock shoots during vitrification in vitro culture were determined by high pressure liquid chromatography. The results showed that the content of GA3, IAA and ABA were very low in vitrified leaves and shoot tips, but cytokinins (CTK) content increased significantly. When plant vitrified heavily CTK content became very low. During subculture period the content of GA3 and IAA in leaves decreased and CTK content increased, meanwhile the rate of GA3/CTK, IAA/CTK decreased significantaly, about 5-10 days before the symptom of vitrification occurred. It is concluded that the content of GA3, IAA and CTK and the rate of GA3/CTK, IAA/CTK was correlated with the vitrification of apple plantlets in vitro culture
