Investigations into the certain detection of Xanthomonas campestris pv. pelargonii in geranium cuttings

In cooperation with eight German propagation companies investigations into the certain detection of Xanthomonas campus pv. pelargonii in geranium cuttings were carried out. The results showed that artificially infected cuttings with different, defined levels of pathogen content can be made available with the help of a specific inoculation technique. Cuttings containing bacteria can be produced in large numbers for investigations into optimizing diagnostic methods and for technique comparison within the group of geranium growers and commercial testing laboratories. Different cultivars of Zonale and Peltatum hybrids showed different background levels in the ELISA testing of healthy plants. With the help of the extinction values thus determined it was possible to define specific clusters for different cultivars and/or origins with average values between 0.02 and 0.15. Accordingly, crucial differences in the delimitation of healthy from potentially ill plant material were obtained depending on the assignment of the control group to a particular cultivar. Low bacterial concentrations could be determined inexpensively and at the same time with certainty with the help of an optimized isolation technique using a specific buffer and YDC medium. Special attention was paid to the suitability of the techniques for routine use in propagation companies as well as in commercial testing laboratories.
