Changes of mineral elements in explant of Magnolia x Soulangiana Soul. - Bod. during the culturing in vitro

Chemical elements in parts of the explant culture of Magnolia x soulangiana Soul. - Bod. during in vitro culturing on Standardi and Catalano medium were analyzed. Content was determined on an atomic spectrophotometer IL VIDEO 12. Ca, Mg and Fe contents were determined by an absorption and K and Na by an emission method, Zn by absorption with correction according to Smith-Heftje. Calcium had the highest quantitative representation among the selected elements and accumulated mostly in leaves (97.98 mg per g), less in the callus and least in the stem. Its maximum was found at the end of culturing. From the microelements group, Zn and Mn had a similar dynamism of accumulation, reaching two maximum values on the 50th and 140th day of culturing. At the biginning of the experiment the successive order of elements was K, Ca, Na, Zn, Mn, Fe and Cu. At the end of the period the concentration of Ca in comparison with K and Fe increased markedly with a parallel decrease of Cu. The order of elements also changed. The results point to an important role of Ca and K in the growth stimulation of the explant culture
